Abstract

The invention provides methodologies and apparatus for producing acellular soft-tissue implants, both in small quantities and in commercializable quantities. Such soft-tissue implants include vascular graft substitutes. An acellular graft is produced by subjecting the tissue sample to an induced pressure mediated flow of an extracting solution, followed by inducing a pressure mediated flow of a treating solution, then washing the treated tissue to produce the acellular graft. The acellular grafts produced are uniform and non-immunogenic. The inventive method allows for the production of multiple decellularized soft tissue implants, where processing time is significantly less than prior art processes and the number of implants produced per day is increased over prior art processes. In clinical use, the decellularized grafts produced exhibit significantly improved in long-term durability and function.

Claims (75)

1. A process for preparing an acellular soft tissue graft comprising:

extracting at least one soft tissue sample with a buffered alkaline extraction solution which contains at least one nonionic detergent and at least one endonuclease, and which is hypotonic to the cells in said soft tissue sample,

washing said extracted tissue in a first washing solution comprising water to produce a first washed tissue,

treating said first washed tissue with a first processing solution comprising an anionic detergent to produce a first processed tissue,

washing said first processed tissue in a second washing solution comprising water to produce a second washed tissue and,

storing said second washed tissue in a storage solution comprising at least one decontaminating agent and water.

2. The process of claim 1 , wherein said extraction solution has an alkaline pH.

3. The process of claim 2 , wherein said extracting solution comprises at least one of polyoxyethylene alcohol, polyoxyethylene isoalcohol, polyoxyethylene p-t-octyl phenol, polyoxyethylene nonylphenol, polyoxyethylene esters of fatty acids, and polyoxyethylene sorbitol esters.

4. The process of claim 3 , wherein said nonionic detergents are at a concentration of from about 0.001% (w:v) to about 10% (w:v).

5. The process of claim 4 , wherein said nonionic detergents are at a concentration of from about 0.1% (w:v) to about 2% (w:v).

6. The process of claim 1 , wherein said first processing solution comprises at least one buffer, has an alkaline pH and an osmolality which is hypertonic to the cells in said soft tissue sample.

7. The process of claim 6 , wherein said first processing solution comprises at least one of sodium dodecylsulphate, sodium dodecylsulphonate, sodium dodecyl-N-sarcosinate, sodium laurylsulfate, and sodium suramin.

8. The process of claim 7 , wherein said anionic detergents are at a concentration of from about 0.001% (w:v) to about 4% (w:v).

9. The process of claim 8 , wherein said anionic detergents are at a concentration of from about 0.1% (w:v) to about 2% (w:v).

10. The process of claim 1 , wherein said first processing solution or said extracting solution comprises at least one of a monovalent salt, a divalent salt, and an antimicrobial agent.

The process of claim 10 , wherein said first processing solution or said extracting solution comprises at least one of sodium chloride, potassium chloride, lithium chloride, calcium chloride, sodium phosphate, calcium hydroxide, potassium sulfate, lithium sulfate, calcium phosphate, potassium phosphate, lithium phosphate, ammonium chloride, magnesium chloride, and calcium sulfate.

12. The process of claim 11 , wherein said first processing solution or said extracting solution comprises at least one of calcium chloride, calcium hydroxide, magnesium chloride, lithium chloride, potassium chloride, and sodium chloride.

13. The process of claim 1 , wherein said storage solution comprises ultrapure, endotoxin-free, water.

14. The process of claim 1 , wherein said decontaminating agent comprises at least one antimicrobial agent.

15. The process of claim 14 , wherein said decontaminating agent comprises at least one of chlorine dioxide, ethanol, isopropanol, methanol, glycerol, and methylparaben.

16. The process of claim 1 , wherein said endonuclease is a broad spectrum endonuclease which degrades deoxyribonucleic acids (DNA) and ribonucleic acids (RNA).

17. The process of claim 16 , wherein said broad-spectrum endonuclease is a recombinant endonuclease.

18. The process of claim 16 , wherein said broad-spectrum endonuclease is present in said hypotonic buffered extracting solution in a concentration sufficient to degrade deoxyribonucleic acids and said ribonucleic acids present in said soft tissue sample.

19. The process of claim 18 , wherein said concentration of said broad-spectrum endonuclease is from about 20 IU to about 400 IU per 1 ml volume of tissue.

20. The process of claim 16 , wherein an activity of said broad-spectrum endonuclease is enhanced by said one or more nonionic detergents.

21. The process of claim 1 , wherein said extracting comprises subjecting said soft tissue sample to a pressure mediated flow of said extracting solution.

22. The process of claim 1 , wherein said treating comprises subjecting said extracted tissue to a pressure mediated flow of said first processing solution.

23. The process of claim 1 , wherein said washing comprises subjecting said first processed tissue to a pressure mediated flow of said washing solution.

24. The process of claim 1 , wherein said extracting is carried out for a time period of from about 6 hours to about 24 hours.

25. The process of claim 24 , wherein said extracting is carried out for a time period of from about 12 hours to about 16 hours.

26. The process of claim 1 , wherein said extracting is carried out at a temperature of from about 4° C. to about 42° C.

27. The process of claim 26 , wherein said extracting is carried out at a temperature of from about 20° C. to about 27° C.

28. An acellular tissue graft prepared by the process of claim 1 , wherein said anionic detergent is sodium dodecylsulfate (SDS) and wherein the second washed tissue contains SDS in about 0.001% by weight SDS/wet weight of tissue.

29. A process for preparing an acellular soft tissue graft comprising:

extracting at least one soft tissue sample with a buffered alkaline extraction solution which contains at least one nonionic detergent and at least one endonuclease, and which is hypotonic to the cells in said soft tissue sample,

washing said extracted tissue in a first washing solution comprising water to produce a first washed tissue,

treating said first washed tissue with a first treating solution comprising a solution which is hypertonic to the cells in said soft tissue sample and contains an anionic detergent and salt to produce a first processed tissue,

washing said first processed tissue in a second washing solution comprising water to produce a second washed tissue,

treating said second washed tissue with a second treating solution which comprises a salt and which is hypertonic to the cells in said soft tissue sample, to produce a second treated tissue,

washing said second treated tissue in at least one third washing solution comprising water to produce a third washed tissue and,

storing said third washed tissue in a storage solution comprising at least one decontaminating agent and water.

30. A process for preparing an acellular soft tissue graft comprising:

extracting at least one soft tissue sample with a buffered alkaline extraction solution which contains at least one nonionic detergent and at least one endonuclease, and which is hypotonic to the cells in said soft tissue sample,

washing said extracted tissue in a first washing solution comprising water to produce a first washed tissue,

treating said first washed tissue with a first treating solution comprising a solution which is hypertonic to the cells in said soft tissue sample and contains an anionic detergent and salt to produce a first processed tissue,

washing said first processed tissue in a second washing solution comprising water to produce a second washed tissue,

treating said second washed tissue with a second treating solution which comprises a salt and an anionic detergent which is sodium dodecylsulfate and which is hypertonic to the cells in said soft tissue sample, to produce a second treated tissue,

washing said second treated tissue in at least one third washing solution comprising water and a calcium salt to produce a third washed tissue and,

storing said third washed tissue in a storage solution comprising at least one decontaminating agent and water.

31. A process for preparing an acellular soft tissue graft comprising:

extracting at least one soft tissue sample with a buffered alkaline extraction solution which contains at least one nonionic detergent and at least one endonuclease, and which is hypotonic to the cells in said soft tissue sample,

washing said extracted tissue in a first washing solution comprising water to produce a first washed tissue,

treating said first washed tissue with a first processing solution comprising an anionic detergent which is sodium dodecylsulfate to produce a first processed tissue,

washing said first processed tissue in a second washing solution comprising water and a calcium salt produce a second washed tissue and,

storing said second washed tissue in a storage solution comprising at least one decontaminating agent and water.

32. The process of claim 30 or 31 , wherein the stored tissue comprises at least 1.0 μmole calcium ion precipitated anionic detergent/mg weight of tissue.

33. The process of claim 30 or 31 , wherein the stored tissue comprises from about 0.1 wt % to about 10 wt % calcium ion precipitated anionic detergent.

34. The process of claim 30 or 31 , wherein the stored tissue comprises less than 20 wt % calcium ion precipitated anionic detergent.

35. The process of claim 30 or 31 , wherein the stored tissue comprises from about 0.2wt % to about 20 wt % calcium ion precipitated anionic detergent.

36. An acellular tissue graft, comprising a soft tissue sample substantially free from cellular elements and calcium ion precipitated anionic detergent, produced by the process of claim 1 or claim 29 , wherein recellularization of said acellular tissue graft in vivo or in vitro is retarded.

37. The tissue graft of claim 36 , wherein the tissue sample is cartilage.

38. An acellular tissue graft prepared by the process of claim 1 or claim 29 or claim 30 or claim 31 .

39. The tissue graft of claim 38 , wherein the graft is essentially non-immunogenic.

40. The tissue graft of claim 38 , wherein the graft has essentially the same tensile properties as the soft tissue sample.

41. The tissue graft of claim 38 , wherein the tissue sample is a vein, an artery, or a heart valve.

42. The tissue graft of claim 38 , wherein the tissue sample is a ligament or a tendon.

43. The tissue graft of claim 38 , wherein the tissue sample is fascia, dura mater, pericardium or skin.

44. The tissue graft of claim 38 , wherein the graft remains acellular post implantation.

45. The tissue graft of claim 38 , wherein the graft remains acellular for at least 6 months post implantation.

46. The tissue graft of claim 38 , wherein the graft re-endothelializes on a luminal surface 3 to 6 months post implantation.

47. The tissue graft of claim 38 , wherein the tissue sample is cartilage.

48. The tissue graft of claim 38 , wherein the graft comprises at least 1.0 μmole calcium ion precipitated anionic detergent/mg wet weight of tissue.

49. The tissue graft of claim 38 , wherein the washed tissue comprises from about 0.1 wt % to about 10 wt % calcium ion precipitated anionic detergent.

50. The tissue graft of claim 38 , wherein the stored tissue comprises less than 20 wt % calcium ion precipitated anionic detergent.

51. The tissue graft of claim 38 , wherein the stored tissue comprises from about 0.2 wt % to about 2.0 wt % calcium ion precipitated anionic detergent.
