Abstract

The genome sequences and the nucleotide sequences coding for the PWD circovirus polypeptides, such as the circovirus structural and non-strucutral polypeptides, vectors including the sequences, and cells and animals transformed by the vectors are provided. Methods for detecting the nucleic acids or polypeptides, and kits for diagnosing infection by a PWD circovirus, also are provided. Method for selecting compounds capable of modulating the viral infection are further provided. Pharmaceutical, including vaccines, compositions for preventing and/or treating viral infections caused by PWD circovirus and the use of vectors for preventing and/or treating diseases also are provided.

Claims (58)

1. A host cell transformed with vector capable of expressing at least one polypeptide encoded by a nucleic acid sequence comprising a sequence having at least 90% sequence identity to SEQ. ID. NO: 25.

2. The host cell of claim 1 , wherein the vector is a baculovirus vector.

3. A host cell transformed with vector capable of expressing at least one polypeptide encoded by a nucleic acid sequence comprising a sequence having at least 95% sequence identity to SEQ. ID. NO: 25.

4. The host cell of claim 3 , wherein the vector is a baculovirus vector.

5. A vaccine composition, comprising the transformed host cell of claim 1 and a pharmaceutically or veterinarily acceptable carrier, wherein said nucleic acid sequence encodes an immunogenic protein that induces a protective response effective against infection by a piglet weight loss disease virus.

6. A vaccine composition, comprising the transformed host cell of claim 2 and a pharmaceutically or veterinarily acceptable carrier, wherein said nucleic acid sequence encodes an immunogenic protein that induces a protective response effective against infection by a piglet weight loss disease virus.

7. A vaccine composition, comprising the transformed host cell of claim 3 and a pharmaceutically or veterinarily acceptable carrier, wherein said nucleic acid sequence encodes an immunogenic protein that induces a protective response effective against infection by a piglet weight loss disease virus.

8. A vaccine composition, comprising the transformed host cell of claim 4 and a pharmaceutically or veterinarily acceptable carrier, wherein said nucleic acid sequence encodes an immunogenic protein that induces a protective response effective against infection by a piglet weight loss disease virus.

9. An immunogenic composition, comprising an immunizing amount of the transformed host cell of claim 1 and a pharmaceutically or veterinarily acceptable carrier.

10. An immunogenic composition, comprising an immunizing amount of the transformed host cell of claim 2 and a pharmaceutically or veterinarily acceptable carrier.

11. An immunogenic composition, comprising an immunizing amount of the transformed host cell of claim 3 and a pharmaceutically or veterinarily acceptable carrier.

12. An immunogenic composition, comprising an immunizing amount of the transformed host cell of claim 4 and a pharmaceutically or veterinarily acceptable carrier.

13. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 5 .

14. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 6 .

15. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 7 .

16. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 8 .

17. A method of recovering protein expressed by ORF′2 from PCVB, said method including the steps a) infecting cells in growth media with a vector comprising a nucleic acid sequence having at least 90% sequence identity to SEQ. ID. NO: 25; b) causing said vector to express said protein; and c) recovering said expressed protein.

18. The method of claim 17 , wherein the vector is a viral vector.

19. The method of claim 17 , wherein the vector is a baculovirus vector.

20. A method of recovering protein expressed by ORF′2 from PCVB, said method including the steps a) infecting cells in growth media with a vector comprising a nucleic acid sequence having at least 95% sequence identity to SEQ. ID. NO: 25; b) causing said vector to express said protein; and c) recovering said expressed protein.

21. The method of claim 20 , wherein the vector is a viral vector.

22. The method of claim 20 , wherein the vector is a baculovirus vector.

23. A method of preparing a composition for inducing an immune response against PCVB, said method comprising the steps of: a) transferring a recombinant DNA comprising a nucleic acid sequence having at least 90% sequence identity to SEQ. ID. NO: 25 into a virus; b) infecting cells with said virus, said cells being in a growth media; c) causing said virus to express the protein encoded by said nucleic acid sequence; d) recovering said recombinant protein; and e) combining said recovered protein with a suitable adjuvant or other pharmaceutically acceptable carrier or excipient.

24. The method of claim 23 , wherein said method further including the step of obtaining said DNA from a transfer vector.

25. The method of claim 23 , said method further including the step of amplifying said nucleic acid from a strain of PCVB prior to cloning said nucleic acid into said transfer vector.

26. The method of claim 23 , said PCVB nucleic acid comprising SEQ ID NO: 25.

27. The method of claim 23 , said cells comprising SF+ cells.

28. The method of claim 23 , said virus comprising baculovirus.

29. A method of preparing a composition for inducing an immune response against PCVB, said method comprising the steps of: a) transferring a recombinant DNA comprising a nucleic acid sequence having at least 95% sequence identity to SEQ. ID. NO: 25 into a virus; b) infecting cells with said virus, said cells being in a growth media; c) causing said virus to express the protein encoded by said nucleic acid sequence; d) recovering said recombinant protein; and e) combining said recovered protein with a suitable adjuvant or other pharmaceutically acceptable carrier or excipient.

30. The method of claim 29 , wherein said method further including the step of obtaining said DNA from a transfer vector.

31. The method of claim 29 , said method further including the step of amplifying said nucleic acid from a strain of PCVB prior to cloning said nucleic acid into said transfer vector.

32. The method of claim 29 , said PCVB nucleic acid comprising SEQ ID NO: 25.

33. The method of claim 29 , said cells comprising SF+ cells.

34. The method of claim 29 , said virus comprising baculovirus.

35. A polypeptide encoded by a nucleic acid having at least 90% identity to SEQ ID NO:25.

36. A polypeptide comprising an amino acid sequence having at least 90% identity to SEQ ID NO:26.

37. A polypeptide encoded by a nucleic acid having at least 95% identity to SEQ ID NO:25.

38. A polypeptide comprising an amino acid sequence having at least 95% identity to SEQ ID NO:26.

39. A vaccine comprising the polypeptide of claim 35 and a pharmaceutically or veterinarily acceptable carrier.

40. A vaccine comprising the polypeptide of claim 36 and a pharmaceutically or veterinarily acceptable carrier.

41. A vaccine comprising the polypeptide of claim 37 and a pharmaceutically or veterinarily acceptable carrier.

42. A vaccine comprising the polypeptide of claim 38 and a pharmaceutically or veterinarily acceptable carrier.

43. An immunogenic composition comprising the polypeptide of claim 35 and a pharmaceutically or veterinarily acceptable carrier.

44. An immunogenic composition comprising the polypeptide of claim 36 and a pharmaceutically or veterinarily acceptable carrier.

45. An immunogenic composition comprising the polypeptide of claim 37 and a pharmaceutically or veterinarily acceptable carrier.

46. An immunogenic composition comprising the polypeptide of claim 38 and a pharmaceutically or veterinarily acceptable carrier.

47. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 39 .

48. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 40 .

49. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 41 .

50. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 42 .

51. A diagnostic kit comprising the polypeptide of claim 35 for detecting porcine circovirus type B (PCVB) by an Enzyme-Linked Immunosorbent Assay (ELISA).

52. A diagnostic kit comprising the polypeptide of claim 36 for detecting porcine circovirus type B (PCVB) by an Enzyme-Linked Immunosorbent Assay (ELISA).

53. A diagnostic kit comprising the polypeptide of claim 37 for detecting porcine circovirus type B (PCVB) by an Enzyme-Linked Immunosorbent Assay (ELISA).

54. A diagnostic kit comprising the polypeptide of claim 38 for detecting porcine circovirus type B (PCVB) by an Enzyme-Linked Immunosorbent Assay (ELISA).

55. A method for detecting and quantifying at least one polypeptide of a porcine circovirus type B (PCVB) by an Enzyme-Linked Immunosorbent Assay (ELISA) method, wherein the at least one polypeptide is encoded by a nucleic acid having a nucleotide sequence with at least 90% sequence identity to SEQ ID NO: 25.

56. A method for detecting and quantifying at least one polypeptide of a porcine circovirus type B (PCVB) by an Enzyme-Linked Immunosorbent Assay (ELISA) method, wherein the at least one polypeptide is encoded by a nucleic acid having a nucleotide sequence with at least 95% sequence identity to SEQ ID NO: 25.

57. A method for detecting and quantifying at least one polypeptide of a porcine circovirus type B (PCVB) by an Enzyme-Linked Immunosorbent Assay (ELISA) method, wherein the polypeptide comprises an amino acid sequence with at least 90% sequence identity to SEQ ID NO: 26.

58. A method for detecting and quantifying at least one polypeptide of a porcine circovirus type B (PCVB) by an Enzyme-Linked Immunosorbent Assay (ELISA) method, wherein polypeptide comprises an amino acid sequence with at least 95% sequence identity to SEQ ID NO: 26.
