Total analyte quantitation
View Patent ↗Methods for determining total analyte concentrations and amounts, especially in combination with analyte separations are provided. Microfluidic devices are used to separate analyte mixtures and detect the individual analytes. Signal areas are summed for each individual analyte to quantitate the total analyte amount. Separate measurements of the total analyte sample are also used to determine total analyte concentration.
1. A method for separating analytes and determining a total analyte concentration or amount, the method comprising:
(i) electrokinetically flowing at least two analytes through a first channel;
(ii) waiting a specified time, during which specified time the at least two analytes flow into an intersection of the first channel and a separation channel;
(iii) injecting the at least two analytes at the intersection into the separation channel;
(iv) separating the at least two analytes in the separation channel, which separating results in two or more separated analytes;
(v) detecting the two or more separated analytes, which detecting results in two or more individual analyte signals; and,
(vi) determining a total analyte concentration or amount from the two or more individual analyte signals, wherein each of the two or more signals comprises an area or peak height, and wherein the determining step comprises summing the areas of the two or more signals or summing the peak heights of the two or more signals to determine the total analyte concentration or amount.
2. The method of claim 1 , wherein the at least two analytes are known analytes for which flow rates at specified voltages are known, and wherein the specified time is determined based on the known flow rates.
3. The method of claim 1 , wherein the at least two analytes have different electrokinetic mobilities, wherein at least one of the at least two analytes comprises a slowest analyte, and waiting the specified time comprises waiting until the slowest analyte reaches the intersection.
4. The method of claim 1 , wherein the separating step comprises electrophoretically separating the at least two analytes.
5. The method of claim 1 , wherein the method further comprises determining a concentration or amount of at least one of the two or more separated analytes based on the individual analyte signal of the at least one separated analyte.
6. The method of claim 5 , the method further comprising calculating a ratio or difference of the amount of at least one of the at least two analytes to the total analyte amount or to a portion of the total analyte amount.
