Abstract

Aryl substituted imidazoquinoline compounds, pharmaceutical compositions containing the compounds, intermediates, and methods of use of these compounds as immunomodulators, for inducing or inhibiting cytokine biosynthesis in animals and in the treatment of diseases including viral, and neoplastic, are disclosed.

Claims (132)

1. A compound of formula (II):

wherein:

R is selected from the group consisting of alkyl, alkoxy, hydroxy, and trifluoromethyl;

n is 0 or 1;

R 1 is —R 4 ;

R 2 is selected from the group consisting of:

—R 4 ;

—X—R 4 ,

—X—Y—R 4 , and

—X—R 5 ;

R 3 is selected from the group consisting of:

-Z-Ar,

-Z-Ar′—Y—R 4 ,

-Z-Ar′—X—Y—R 4 ,

-Z-Ar′—R 5 , and

-Z-Ar′—X—R 5 ;

Ar is selected from the group consisting of aryl and heteroaryl both of which can be unsubstituted or can be substituted by one or more substituents independently selected from the group consisting of alkyl, alkenyl, alkoxy, methylenedioxy, haloalkyl, haloalkoxy, halogen, nitro, hydroxy, hydroxyalkyl, mercapto, cyano, carboxy, formyl, aryl, aryloxy, arylalkoxy, heteroaryl, heteroaryloxy, heteroarylalkoxy, heterocyclyl, heterocyclylalkyl, amino, alkylamino, and dialkylamino;

Ar′ is selected from the group consisting of arylene and heteroarylene both of which can be unsubstituted or can be substituted by one or more substituents independently selected from the group consisting of alkyl, alkenyl, alkoxy, haloalkyl, haloalkoxy, halogen, nitro, hydroxy, hydroxyalkyl, mercapto, cyano, carboxy, formyl, aryl, aryloxy, arylalkoxy, heteroaryl, heteroaryloxy, heteroarylalkoxy, heterocyclyl, heterocyclylalkyl, amino, alkylamino, and dialkylamino;

each X is independently selected from the group consisting of alkylene, alkenylene, alkynylene, arylene, heteroarylene, and heterocyclylene wherein the alkylene, alkenylene, and alkynylene groups can be optionally interrupted or terminated with arylene, heteroarylene, or heterocyclylene, and optionally interrupted by one or more —O— groups;

each Y is independently selected from the group consisting of:

Z is selected from the group consisting of a bond, alkylene, alkenylene, and alkynylene;

each R 4 is independently selected from the group consisting of hydrogen, alkyl, alkenyl, alkynyl, aryl, arylalkylenyl, aryloxyalkylenyl, alkylarylenyl, heteroaryl, heteroarylalkylenyl, heteroaryloxyalkylenyl, alkylheteroarylenyl, and heterocyclyl wherein the alkyl, alkenyl, alkynyl, aryl, arylalkylenyl, aryloxyalkylenyl, alkylarylenyl, heteroaryl, heteroarylalkylenyl, heteroaryloxyalkylenyl, alkylheteroarylenyl, and heterocyclyl groups can be unsubstituted or substituted by one or more substituents independently selected from the group consisting of alkyl, alkoxy, hydroxyalkyl, haloalkyl, haloalkoxy, halogen, nitro, hydroxy, mercapto, cyano, aryl, aryloxy, arylalkyleneoxy, heteroaryl, heteroaryloxy, heteroarylalkyleneoxy, heterocyclyl, amino, alkylamino, dialkylamino, (dialkylamino)alkyleneoxy, and in the case of alkyl, alkenyl, alkynyl, and heterocyclyl, oxo;

each R 5 is independently selected from the group consisting of:

each R 6 is independently selected from the group consisting of ═O and ═S;

each R 7 is independently C 2-7 alkylene;

R 8 is selected from the group consisting of hydrogen, alkyl, alkoxyalkylenyl, and arylalkylenyl;

R 9 is selected from the group consisting of hydrogen and alkyl;

each R 10 is independently C 3-8 alkylene;

A is selected from the group consisting of —O—, —C(O)—, —S(O) 0-2 —, —CH 2 —, and —N(R 4 )—;

Q is selected from the group consisting of a bond, —C(R 6 )—, —C(R 6 )—C(R 6 )—, —S(O) 2 —, —C(R 6 )—N(R 8 )—W—, —S(O) 2 —N(R 8 )—, —C(R 6 )—O—, and —C(R 6 )—N(OR 9 )—;

V is selected from the group consisting of —C(R 6 )—, —O—C(R 6 )—, —N(R 8 )—C(R 6 )—, and —S(O) 2 —;

W is selected from the group consisting of a bond, —C(O)—, and —S(O) 2 —;

a and b are independently integers from 1 to 6 with the proviso that a+b is ≦7; and

wherein heteroaryl is selected from the group consisting of furyl, thienyl, pyridyl, quinolinyl, isoquinolinyl, indolyl, isoindolyl, trialkyl, pyrrolyl, tetrazolyl, imidazolyl, pyrazolyl, oxazolyl, thiazolyl, benzofuranyl, benzothiophenyl, carbazolyl, benzoxazolyl, pyrimidinyl, benzimidazolyl, quinoxalinyl, benzothiazolyl, naphthyridinyl, isoxazolyl, isothiazolyl, purinyl, quinazolinyl, pyrazinyl, 1-oxidopyridyl, pyridazinyl, triazinyl, tetrazinyl, oxadiazoyl, and thiadiazolyl; and heterocyclyl is selected from the group consisting of the fully saturated and partially unsaturated derivatives of the above heteroaryl groups and from the group consisting of pyrrolidinyl, tetrahydrofuranyl, morpholinyl, thiomorpholinyl, piperidinyl, piperazinyl, thiazolidinyl, imidazolidinyl, isothiazolidinyl, tetrahydropyranyl, quinuclidinyl, homopiperidinyl, and homopiperazinyl;

or a pharmaceutically acceptable salt thereof.

2. The compound or salt of claim 1 wherein -Z is a bond.

3. The compound or salt of claim 1 wherein R 3 is -Z-Ar.

4. The compound or salt of claim 1 wherein R 3 is selected from the group consisting of phenyl, pyridyl, pyrrolyl, thienyl, and furyl; each of which can be unsubstituted or can be substituted by one or more substituents selected from the group consisting of halogen, alkyl, hydroxy, hydroxyalkyl, alkoxy, carboxy, and cyano.

5. The compound or salt of claim 1 wherein R 3 is -Z-Ar′—Y—R 4 , -Z-Ar′—X—Y—R 4 , or -Z-Ar′—R 5 .

6. The compound or salt of claim 5 wherein Ar′ is phenyl or pyridyl;

Y is selected from the group consisting of:

—S(O) 0-2 —

—C(O)—,

—N(R 8 )-Q-,

—C(R 6 )—N(R 8 )—, and

—C(R 6 )—N(OR 9 )—;

wherein Q is selected from the group consisting of a bond, —C(O)—, and —S(O) 2 —; and R 8 is selected from the group consisting of hydrogen, C 1-4 alkyl, and alkoxyalkylenyl;

X is C 1-4 alkylene;

R 4 is selected from the group consisting of alkyl, aryl, heteroaryl, and heterocyclyl; and

R 5 is

7. The compound or salt of claim 1 wherein R 1 is selected from the group consisting of alkyl, arylalkylenyl, aryloxyalkylenyl, hydroxyalkyl.

8. The compound or salt of claim 1 wherein R 2 is selected from the group consisting of hydrogen, alkyl, and alkoxyalkylenyl.

9. A compound of formula (IIa):

wherein:

R is selected from the group consisting of alkyl, alkoxy, hydroxy, and trifluoromethyl;

n is 0 or 1;

R 1 is —R 4 ;

R 2 is selected from the group consisting of:

—R 4 ,

—X—R 4 ,

—X—Y—R 4 , and

—X—R 5 ;

R 3 is selected from the group consisting of:

-Z-Ar and

-Z-Ar′—Y—R 4 ;

each X is independently selected from the group consisting of alkylene, alkenylene, alkynylene, arylene, heteroarylene, and heterocyclylene wherein the alkylene, alkenylene, and alkynylene groups can be optionally interrupted by arylene, heteroarylene or heterocyclylene or by one or more —O— groups;

each Y is independently selected from the group consisting of:

Z is selected from the group consisting of a bond, alkylene, alkenylene, and alkynylene;

Ar is selected from the group consisting of aryl and heteroaryl both of which can be unsubstituted or can be substituted by one or more substituents independently selected from the group consisting of alkyl, alkoxy, haloalkyl, haloalkoxy, halogen, nitro, hydroxy, mercapto, cyano, carboxy, formyl, aryl, aryloxy, arylalkoxy, heteroaryl, heteroaryloxy, heteroarylalkoxy, heterocyclyl, heterocyclylalkyl, amino, alkylamino, and dialkylamino;

Ar′ is selected from the group consisting of arylene and heteroarylene both of which can be unsubstituted or can be substituted by one or more substituents independently selected from the group consisting of alkyl, alkoxy, haloalkyl, haloalkoxy, halogen, nitro, hydroxy, mercapto, cyano, carboxy, formyl, aryl, aryloxy, arylalkoxy, heteroaryl, heteroaryloxy, heteroarylalkoxy, heterocyclyl, heterocyclylalkyl, amino, alkylamino, and dialkylamino;

each R 4 is independently selected from the group consisting of hydrogen, alkyl, alkenyl, alkynyl, aryl, heteroaryl, heterocyclyl wherein the alkyl, alkenyl, alkynyl, aryl, heteroaryl, and heterocyclyl groups can be unsubstituted or substituted by one or more substituents independently selected from the group consisting of alkyl, alkoxy, haloalkyl, haloalkoxy, halogen, nitro, hydroxy, mercapto, cyano, carboxy, formyl, aryl, aryloxy, arylalkoxy, heteroaryl, heteroaryloxy, heteroarylalkoxy, heterocyclyl, heterocyclylalkyl, amino, alkylamino, dialkylamino, and in the case of alkyl, alkenyl, alkynyl, and heterocyclyl, oxo;

each R 5 is independently selected from the group consisting of:

R 6 is selected from the group consisting of ═O and ═S;

R 7 is C 2-7 alkylene;

each R 8 present is independently selected from the group consisting of hydrogen, alkyl, and arylalkyl;

R 9 is selected from the group consisting of hydrogen and alkyl;

A is selected from the group consisting of —O—, —S(O) 0-2 —, —NR 4 —, and —CH 2 —;

Q is selected from the group consisting of —CR 6 —, —SO 2 —, —CR 6 —NR 8 —W—, —SO 2 —NR 8 —, —CR 6 —O—, and —CR 6 —N(OR 9 )—;

V is selected from the group consisting of —CR 6 —, —O—CR 6 —, and —NR 8 —CR 6 —;

W is selected from the group consisting of a bond, —C(O)—, and —SO 2 —;

a and b are independently integers from 1 to 6 with the proviso that a+b is ≦7; and

wherein heteroaryl is selected from the group consisting of furyl, thienyl, pyridyl, quinolinyl, isoquinolinyl, indolyl, isoindolyl, triazolyl, pyrrolyl, tetrazolyl, imidazolyl, pyrazolyl, oxazolyl, thiazolyl, benzofuranyl, benzothiophenyl, carbazolyl, benzoxazolyl, pyrimidinyl, benzimidazolyl, quinoxalinyl, benzothiazolyl, naphthyridinyl, isoxazolyl, isothiazolyl, purinyl, and quinazolinyl; and heterocyclyl is selected from the group consisting of all of the fully saturated and partially unsaturated derivatives of the above heteroaryl groups and from the group consisting of pyrrolidinyl, tetrahydrofuranyl, tetrahydropyranyl, morpholinyl, thiomorpholinyl, piperidinyl, piperazinyl, thiazolidinyl, isothiazolidinyl, and imidazolidinyl;

or a pharmaceutically acceptable salt thereof.

10. The compound or salt of claim 9 wherein R 1 is alkyl or hydroxyalkyl.

11. The compound or salt of claim 9 wherein R 2 is R 4 .

12. The compound or salt of claim 9 wherein R 2 is alkyl or alkoxyalkyl.

13. The compound or salt of claim 9 wherein R 3 is -Z-Ar.

14. The compound or salt of claim 13 wherein -Z- is a bond.

15. The compound or salt of claim 13 wherein —Ar is unsubstituted aryl or heteroaryl.

16. The compound or salt of claim 15 wherein —Ar is phenyl, thienyl or pyridyl.

17. The compound or salt of claim 9 wherein R 3 is attached at the 7-position.

18. The compound or salt of claim 9 wherein R 3 is attached at the 8-position.

19. The compound or salt of claim 9 wherein n is 0.

20. A compound of Formula (VIII):

wherein:

R 2 is selected from the group consisting of:

—R 4 ,

—X—R 4 ,

—X—Y—R 4 , and

—X—R 5 ;

R 3 is selected from the group consisting of:

-Z-Ar,

-Z-Ar′—Y—R 4 ,

-Z-Ar′—X—Y—R 4 ,

-Z-Ar′—R 5 , and

-Z-Ar′—X—R 5 ;

each X is independently selected from the group consisting of alkylene, alkenylene, alkynylene, arylene, heteroarylene, and heterocyclylene wherein the alkylene, alkenylene, and alkynylene groups can be optionally interrupted or terminated with arylene, heteroarylene, or heterocyclylene, and optionally interrupted by one or more —O— groups;

each Y is independently selected from the group consisting of:

wherein heteroaryl is selected from the group consisting of furyl, thienyl, pyridyl, quinolinyl, isoquinolinyl, indolyl, isoindolyl, triazolyl, pyrrolyl, tetrazolyl, imidazolyl, pyrazolyl, oxazolyl, thiazolyl, benzofuranyl, benzothiophenyl, carbazolyl, benzoxazolyl, pyrimidinyl, benzimidazolyl, quinoxalinyl, benzothiazolyl, naphthyridinyl, isoxazolyl, isothiazolyl, purinyl, quinazolinyl, pyrazinyl, 1-oxidopyridyl, pyridazinyl, triazinyl, tetrazinyl, oxadiazopyl, and thiadiazolyl; and heterocyclyl is selected from the group consisting of the fully saturated and partially unsaturated derivatives of the above heteroaryl groups and from the group consisting of pyrrolidinyl, tetrahydrofuranyl, morpholinyl, thiomorpholinyl, piperidinyl, piperazinyl, thiazolidinyl, imidazolidinyl, isothiazolidinyl, tetrahydropyranyl, quinuclidinyl, homopiperidinyl, and homopiperazinyl;

or a pharmaceutically acceptable salt thereof.

21. The compound or salt of claim 20 wherein R 2 is selected from the group consisting of hydrogen, C 1-4 alkyl, and C 1-4 alkyl-O—C 1-4 alkylenyl.

The compound or salt of claim 20 wherein R 3 is phenyl or pyridyl, either of which can be unsubstituted or can be substituted by one or more substituents selected from the group consisting of halogen, alkyl, hydroxy, hydroxyalkyl, alkoxy, alkylsulfonylamino, arylsulfonylamino, alkylcarbonylamino, arylcarbonylamino, alkylsulfonylaminoalkylenyl, arylsulfonylaminoalkylenyl, alkylcarbonylaminoalkylenyl, and arylcarbonylaminoalkylenyl.

23. The compound or salt of claim 20 wherein R 4 is selected from the group consisting of C 1-6 alkyl, C 1-6 hydroxyalkyl, C 1-4 alkyl-O—C 1-4 alkylenyl, and aryl-O—C 1-4 alkylenyl.

24. The compound or salt of claim 23 wherein R 4 is selected from the group consisting of 2-methylpropyl, 2-hydroxy-2-methylpropyl, 3-methoxypropyl, and phenoxyethyl.

25. A pharmaceutical composition comprising a therapeutically effective amount of a compound or salt of claim 1 and a pharmaceutically acceptable carrier.

26. A pharmaceutical composition comprising a therapeutically effective amount of a compound or salt of claim 9 and a pharmaceutically acceptable carrier.

27. A pharmaceutical composition comprising a therapeutically effective amount of a compound or salt of claim 20 and a pharmaceutically acceptable carrier.
