Abstract

A method and apparatus for measuring the presence or absence of reaction between a first and second material of interest by measuring osmotic pressure changes of reaction detector. The reaction detector is capable of measuring the small changes in pressure that occur due to osmotic pressure shifts during a catalytic or binding reaction at species concentrations down to approximately 10 −7 M.

Claims (25)

1. A method for monitoring reactivity in reactions between a first and second material of interest, said method comprising the steps of:

a) providing an osmotic reaction detector comprising a semi-permeable membrane and a pressure sensor;

b) introducing one of a reaction mixture and a reference solution into said osmotic reaction detector, said reaction mixture comprising a carrier fluid, a first material of interest and a second material of interest and said reference solution comprising any one or combination of the first material, the second material, a surrogate reference material and a carrier fluid;

c) positioning said osmotic reaction detector such that said semi-permeable membrane is in fluid contact with and positioned between said reaction mixture and said reference solution; and

d) monitoring an output from said pressure sensor after the positioning of said osmotic reaction detector, wherein any changes in pressure are measured in the reference solution and occur as a result of a change in concentration in said reaction mixture of one or more of said first material, second material, surrogate reference material or reaction products formed from said first and second materials.

2. A method according to claim 1 , wherein said method is performed using a plurality of osmotic reaction detectors, wherein at least two osmotic reaction detectors of the plurality utilize at least one different first or second materials.

3. A method according to claim 2 , wherein said method for monitoring at least two osmotic reaction detectors are performed substantially simultaneously.

4. A method according to claim 2 , wherein one of said reference solution or reaction mixture in the positioning step is held within a titer plate.

5. A method according to claim 4 , wherein the plurality of osmotic reaction detectors are positioned by an automated robotic arm.

6. A method according to claim 1 ,

wherein said method monitors reactions in said osmotic reaction detectors at initial concentration of said first and second materials of interest in said reaction mixture in the range of approximately 5×10 −5 M to 5×10 −7 M.

7. A method according to claim 1 , wherein said method monitors reactions at initial concentration of said first and second materials of interest in said reaction mixture as low as approximately 5×10 −7 M.

8. A method according to claim 1 , wherein said method measures osmotic pressure changes in a range of approximately 1.2 N/m 2 to 130 N/m 2 .

9. A method according to claim 1 , wherein said pressure sensor is a diaphragm having an effective spring constant of from about 0.1 to 2.5 N/m.

10. A method according to claim 1 , wherein said osmotic pressure changes are measured from approximately 1.2 N/m 2 or greater.

11. A method according to claim 1 , wherein said pressure sensor is a diaphragm operatively associated with said osmotic reaction detector and wherein step d) is performed by measuring the amount of deformation in said diaphragm.

12. A method according to claim 11 , wherein said amount of deformation is measured by a change in capacitance of said diaphragm.

13. A method according to claim 11 , wherein said amount of deformation is measured by a change in the deflection of light incident on said diaphragm.

14. A method according to claim 1 , wherein at least one of said first and second materials of interest is an antibody, an antigen, a nucleic acid, a protein, a ligand, an oligonucleotide, or a polypeptide.

15. A method according to claim 1 , wherein said pressure sensor is a reference solution mixture height detector in an osmotic cell and step d) is performed by measuring the change in reference solution height in said osmotic reaction detector.

16. A method according to claim 15 , wherein said reference solution height is measured by determining the resistance between two spaced apart conductors positioned in said osmotic reaction detector.

17. A method according to claim 1 , wherein said semi-permeable membrane is impermeable to said first and second materials of interest and said surrogate reference material and is permeable to said carrier fluid.

A method according to claim 1 , wherein said semi-permeable membrane is impermeable to at least one of said first material, said second material and said surrogate reference material, and is permeable to at least one of said first material and said second material and said surrogate reference material or products of a reaction between said first material and said second material.

19. A method according to claim 1 , wherein said reference solution comprises at least one of said first and second materials of interest.

20. A method according to claim 1 , wherein a total combined initial unit concentration of said first and second materials of interest in said reaction mixture is equal to an initial unit concentration of solute molecules in said reference solution.
