Abstract

The present invention provides trans-complementation systems for expressing gene products in plants. In general, the invention provides systems including a carrier vector and a producer vector, both based on plant viruses. The producer vector is defective for at least one function needed for successful systemic infection of a plant, e.g., replication, cell-to-cell movement, or long distance movement. The carrier vector supplies the missing function in trans. Certain producer vectors lack a functional coat protein coding sequence, in which case the corresponding producer vector supplies coat protein in trans. The invention also provides novel plant viral vectors and methods of use, e.g., to produce polypeptides or active RNAs in plants.

Claims (19)

1. A producer vector comprising (a) one or more components of a tobacco mosaic virus; and (b) a polynucleotide of interest, wherein the vector is defective for cell-to-cell movement or systemic movement, and wherein the vector comprises at least one polynucleotide encoding a replicase protein of a plant virus and comprises sufficient non-coding portions to allow self-replication.

A producer vector comprising (a) one or more components of an alfalfa mosaic virus; and (b) a polynucleotide of interest, wherein the vector is defective for cell-to-cell movement or systemic movement, and wherein the vector comprises at least one polynucleotide encoding a replicase protein of a plant virus and comprises sufficient non-coding portions to allow self-replication.

3. The producer vector of claim 1 or 2 , wherein the vector is defective for both cell-to-cell movement and systemic movement.

4. A method of expressing a polynucleotide of interest comprising:

(a) introducing the vector of claim 3 into a plant or plant cell;

(b) maintaining the plant cell or plant cell under conditions and for a time sufficient that the polynucleotide is expressed.

5. The method of claim 4 , wherein the polynucleotide encodes a pharmaceutical protein of interest.

6. The method of claim 4 , wherein the method comprises steps of:

(a) introducing a second vector into the plant or plant cell, wherein the second vector, or both, comprises a gene that encodes a replicase protein;

(b) maintaining the plant or plant cell under conditions and for a time sufficient that the polynucleotide is expressed in at least some plant cells.

7. The method of claim 6 , wherein the second vector comprises a polynucleotide of interest, the method comprising steps of:

maintaining the plant or plant cell under conditions and for a time sufficient that the polynucleotide of the second vector is expressed in at least some plant cells.

8. The method of claim 7 , wherein the second vector comprises a gene that encodes a replicase protein.

9. The method of claim 6 , wherein the first and second polypeptides are polypeptide chains of a multimeric protein.

10. The method of claim 9 , wherein the first and second polypeptides encode therapeutically active proteins.

11. The producer vector of claim 1 or 2 , wherein the polynucleotide of interest encodes a pharmaceutical protein of interest.

12. A method of expressing a polynucleotide of interest comprising:

(a) introducing the vector of claim 1 or 2 into a plant cell;

(b) maintaining the plant cell under conditions and for a time sufficient that the polynucleotide is expressed.
