Abstract

The purification and isolation of various genes which encode mammalian cell surface polypeptides. Nucleic acids, proteins, antibodies, and other reagents useful in modulating development of cells, e.g., lymphoid and myeloid, are provided, along with methods for their use.

Claims (22)

1. An isolated or purified polypeptide having at least 90% sequence identity to an extracellular domain of SEQ ID NO: 12 or 14, wherein the polypeptide associates with a DAP12 polypeptide having the amino acid sequence of SEQ ID NO: 2 or 6.

2. The isolated or purified polypeptide of claim 1 , wherein the polypeptide has at least 95% sequence identity to the extracellular domain of SEQ ID NO: 12 or 14, wherein the polypeptide associates with a DAP12 polypeptide having the amino acid sequence of SEQ ID NO: 2 or 6.

3. The isolated and purified polypeptide of claim 1 fused to a heterologous protein.

4. The isolated and purified polypeptide of claim 3 , wherein the heterologous protein is an immunoglobulin peptide.

5. The isolated and purified polypeptide of claim 3 , wherein the heterologous protein is a detection or purification tag selected from the group consisting of a FLAG, His6, β-galactosidase, trpE, Protein A, β-lactamase, alpha amylase, alcohol dehydrogenase, and yeast alpha mating factor.

6. A composition comprising the isolated and purified polypeptide of claim 1 and a pharmaceutically acceptable carrier.

7. An isolated or purified polypeptide having at least 90% sequence identity to SEQ ID NO: 12 or 14, wherein the polypeptide associates with a DAP12 polypeptide having the amino acid sequence of SEQ ID NO: 2 or 6.

8. The isolated or purified polypeptide of claim 7 , wherein the polypeptide has at least 95% sequence identity to SEQ ID NO: 12 or 14, wherein the polypeptide associates with a DAP12 polypeptide having the amino acid sequence of SEQ ID NO: 2 or 6.

9. The isolated and purified polypeptide of claim 7 fused to a heterologous protein.

10. The isolated and purified polypeptide of claim 9 , wherein the heterologous protein is an immunoglobulin peptide.

11. The isolated and purified polypeptide of claim 9 , wherein the heterologous protein is a detection or purification tag selected from the group consisting of a FLAG, His6, β-galactosidase, trpE, Protein A, β-lactamase, alpha amylase, alcohol dehydrogenase, and yeast alpha mating factor.

12. A composition comprising the isolated and purified polypeptide of claim 7 and a pharmaceutically acceptable carrier.

13. An isolated or purifed polypeptide comprising an extracellular domain of the amino acid sequence of SEQ ID NO: 12 or 14.

14. The isolated and purified polypeptide of claim 13 fused to a heterologous protein.

15. The isolated and purified polypeptide of claim 14 , wherein the heterologous protein is an immunoglobulin peptide.

16. The isolated and purified polypeptide of claim 14 , wherein the heterologous protein is a detection or purification tag selected from the group consisting of a FLAG, His6, β-galactosidase, trpE, Protein A, β-lactamase, alpha amylase, alcohol dehydrogenase, and yeast alpha mating factor.

17. A composition comprising the isolated and purified polypeptide of claim 13 and a pharmaceutically acceptable carrier.

18. An isolated or purified polypeptide comprising the amino acid sequence of SEQ ID NO: 12 or 14.

19. The isolated and purified polypeptide of claim 18 fused to a heterologous protein.

20. The isolated and purified polypeptide of claim 19 , wherein the heterologous protein is an immunoglobulin peptide.

21. The isolated and purified polypeptide of claim 19 , wherein the heterologous protein is a detection or purification tag selected from the group consisting of a FLAG, His6, β-galactosidase, trpE, Protein A, β-lactamase, alpha amylase, alcohol dehydrogenase, and yeast alpha mating factor.

22. A composition comprising the isolated and purified polypeptide of claim 18 and a pharmaceutically acceptable carrier.
