Immunotherapeutic methods targeted towards stratum corneum chymotryptic enzyme
View Patent ↗The present invention discloses the protease stratum corneum chymotrytic enzyme (SCCE) is specifically over-expressed in ovarian and other malignancies. A number of SCCE peptides can induce immune responses to SCCE, thereby demonstrating the potential of these peptides in monitoring and the development of immunotherapies for ovarian and other malignancies.
1. A method of providing immunotherapy towards stratum corneum chymotrytic enzyme (SCCE) in an individual, comprising the step of:
inoculating an individual with the SCCE protein encoded by the DNA sequence shown in SEQ ID NO:30 or an SCCE peptide shown in SEQ ID NO: 32, 33, 35, 36, 86 or 137, wherein said SCCE protein or SCCE peptide is processed and presented on the surface of dendritic cells such that said SCCE protein or SCCE peptide presented on the surface of the dendritic cells elicits an immune response in said individual, thereby providing immunotherapy towards SCCE in the individual.
2. The method of claim 1 , wherein said individual has ovarian cancer, lung cancer, prostate cancer or colon cancer.
3. The method of claim 1 , further comprising:
isolating T cells from said individual; and measuring immune responses induced by said SCCE protein or SCCE peptide, wherein an increased level of immune responses compared to those exhibited by cells from normal individual indicates that said individual has been treated with said SCCE protein or SCCE peptide.

The method of claim 3 , wherein said immune response is T cell proliferation induced by said SCCE protein or SCCE peptide, frequency of cytokine-secreting T cells specific to said SCCE protein or SCCE peptide and frequency of T cells expressing T cell receptor specific to said SCCE protein or SCCE peptide.
