Abstract

The present invention provides compounds which inhibit poly(ADP-ribose) polymerase (“PARP”), compositions containing these compounds and methods for using these PARP inhibitors to treat, prevent and/or ameliorate the effects of the conditions described herein.

Claims (29)

1. A compound of the Formula:

where

R 1 is H, halogen or alkyl;

R 2 is H, halogen or alkyl;

R 3 is independently H, amino, hydroxy, —NH—NH 2 , halogen-substituted amino, —O-alkyl, —O-aryl, or an alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl or —OR 6 or —NR 6 R 7 or —CH 2 NR 6 R 7 where R 6 and R 7 are each independently H or an alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl and

R 4 is independently H, amino, —NH—NH 2 , —CO—NH—NH 2 , halogen-substituted amino, —O—C 2 -C 6 alkyl, —O-aryl or an alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl, —COR 8 where R 8 is H, hydroxy, an alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl or —CH 2 OH or —OR 6 or —CH 2 NR 6 R 7 or —CH 2 R 6 or —NR 6 R 7 where R 6 and R 7 are each independently H or alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl or aryl substituted alkyl.

2. The compound of claim 1

where

R 1 is H, F, Cl, methoxy or methyl;

R 2 is H, F, Cl, methoxy or methyl;

R 3 is independently H, amino, hydroxy, —NH—NH 2 , halogen-substituted amino, —O-alkyl, —O-aryl or an alkyl, alkenyl, —COR 8 where R 8 is H, an alkyl, alkenyl or —OR 6 or —NR 6 R 7 where R 6 and R 7 are each independently H or an alkyl or alkenyl and

R 4 is independently H, amino, —NH—NH 2 , —CO—NH—NH 2 , halogen-substituted amino, —O—C 2 -C 6 alkyl, —O-aryl, an alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl, —COR 8 where R 8 is H, hydroxy, an alkyl or alkenyl or —CH 2 OH or —CH 2 R 6 or —OR 6 or —CH 2 NR 6 R 7 or —NR 6 R 7 where R 6 and R 7 are each independently H, an alkyl, alkenyl, aryl substituted alkyl or heterocycloalkyl.

3. A compound is selected from the group consisting of:

A method of treating diseases or conditions selected from the group consisting of neural tissue damage resulting from ischemia and reperfusion injury, depression, osteoarthritis, gout chronic pain, acute pain, bowel disorder, neurophathic pain, renal failure, retinal ischemia and septic shock in an animal comprising administering to said animal an effective amount of a compound of Formula II:

where

R 1 is H, halogen or alkyl;

R 2 is H, halogen or alkyl;

R 3 is independently H, amino, hydroxy, —NH—NH 2 , halogen-substituted amino, —O-alkyl, —O-aryl, or an alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl or —OR 6 or —NR 6 R 7 or —CH 2 NR 6 R 7 where R 6 and R 7 are each independently H or an alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl and

R 4 is independently H, amino, —NH—NH 2 , —CO—NH—NH 2 , halogen-substituted amino, —O-alkyl, —O-aryl or an alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl, —COR 8 where R 8 is H, hydroxy, a alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl or —CH 2 OH or —OR 6 or —CH 2 NR 6 R 7 or —CH 2 R 6 or —NR 6 R7 where R 6 and R 7 are each independently H or alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl or aryl substituted alkyl.

5. The method of claim 4 , wherein said ischemia or reperfusion injury is cerebral injury after cardiac arrest and cardio-pulmonary resuscitation.

6. The method of claim 4 , wherein the septic shock is endotoxic shock.

7. The method of claim 4 , wherein the bowel disorder is colitis.

8. The method of claim 4 , wherein the bowel disorder is Crohn's Disease.

9. The method of claim 4 , where

R 1 is H, F, Cl, methoxy or methyl;

R 2 is H, F, Cl, methoxy or methyl;

R 3 is independently H, amino, hydroxy, —NH—NH 2 , halogen-substituted amino, —O-alkyl, —O-aryl or an alkyl, alkenyl, —COR 8 where R 8 is H, an alkyl, alkenyl or —OR 6 or —NR 6 R 7 where R 6 and R 7 are each independently H or an alkyl or alkenyl and

R 4 is independently H, amino, —NH—NH 2 , —CO—NH—NH 2 , halogen-substituted amino, —O-alkyl, —O-aryl, an alkyl, alkenyl, alkynyl, cycloalkyl, heterocycloalkyl, optionally substituted aryl, optionally substituted heteroaryl, —COR 8 where R 8 is H, hydroxy, an alkyl or alkenyl or —CH 2 OH or —CH 2 R 6 or —OR 6 or —CH 2 NR 6 R 7 or —NR 6 R 7 where R 6 and R 7 are each independently H, an alkyl, alkenyl, aryl substituted alkyl or heterocycloalkyl.

A method of treating diseases or conditions selected from the group consisting of neural tissue damage resulting from ischemia and reperfusion injury, depression, osteoarthritis, gout chronic pain, acute pain, bowel disorder, neurophathic pain, renal failure, retinal ischemia and septic shock in an animal comprising administering to said animal an effective amount of a compound selected from the group consisting of:
