Hepatitis C receptor protein CD81
View Patent ↗The present invention relates to the use of CD81 protein and polynucleic acid in the therapy and diagnosis of hepatitis C and pharmaceutical compositions, animal models and diagnostic kits for such purposes.
1. A method for inhibiting binding of the E2 protein of HCV to human cells comprising administering to a human infected with HCV an amount of a CD81 protein effective to bind HCV, wherein the human CD81 protein is a soluble form of the CD81 protein that comprises amino acids 113–201 of the human CD81 amino acid sequence depicted in SEQ ID NO: 21, and a deletion of one or more of the transmembrane domains selected from the group consisting of:
a) transmembrane domain 1 (TM1) consisting of amino acids 12–35 of the amino acid sequence of SEQ ID NO :21,
b) transmembrane domain 2 (TM2) consisting of amino acids 63–84 of the amino acid sequence of SEQ ID NO:2 1,
c) transmembrane domain 3 (TM3) consisting of amino acids 89–112 of the amino acid sequence of SEQ ID NO:21, and
d) transmembrane domain 4 (TM4) consisting of amino acids 202–224 of the amino acid sequence of SEQ ID NO:21.
2. The method of claim 1 , wherein the CD81 protein consists of the sequence of amino acids shown at positions 113–201 of the human CD8l amino acid sequence depicted in SEQ ID NO:21.
