Abstract

A method and apparatus for the manipulation of colloidal particulates and biomolecules at the interface between an insulating electrode such as silicon oxide and an electrolyte solution. Light-controlled electrokinetic assembly of particles near surfaces relics on the combination of three functional elements: the AC electric field-induced assembly of planar aggregates; the patterning of the electrolyte/silicon oxide/silicon interface to exert spatial control over the assembly process; and the real-time control of the assembly process via external illumination. The present invention provides a set of fundamental operations enabling interactive control over the creation and placement of planar arrays of several types of particles and biomolecules and the manipulation of array shape and size. The preset invention enables sample preparation and handling for diagnostic assays and biochemical analysis in an array format, and the functional integration of these operations. In addition, the present invention provides a procedure for the creation of material surfaces with desired properties and for the fabrication of surface-mounted optical components.

Claims (12)

1. A method of decoding an assay in which a substantially planar array of beads, said planar array including differently-encoded beads and wherein different ligands are attached to differently-encoded beads, is used to bind to and determine the existence of and identity of optically labeled analytes which are present in a sample, said optically labeled analytes capable of binding to one or more of the ligands and wherein following said binding, the optical labels of said optically labeled analytes can be optically detected, comprising the following steps:

(i) imaging the substantially planar array of beads to identify the relative locations of different beads within the array;

(ii) placing the sample in contact with the substantially planar array of beads under binding conditions whereby the optically labeled analytes can bind to the ligands;

(iii) imaging the substantially planar array of beads to identify the relative locations of beads bearing ligands within the array which bound to optically labeled analytes by optically detecting said optical labels; and

(iv) comparing the images in steps (i) and (iii) so as to identify the ligands which are bound to optically labeled analytes or the optically labeled analytes which bound to ligands.

2. The method of claim 1 wherein the optical label for the analytes is fluorescence.

3. The method of claim 1 wherein the spatial coordinates of the optically labeled analytes directly indicates the identity of the agent.

4. The method of claim 1 wherein the beads are fluorescently encoded.

5. The method of claim 1 wherein the beads are made of polystyrene.

6. The method of claim 1 wherein the beads are encoded with a binary encoding system.

7. The method of claim 6 wherein the binary encoding system is performed by attaching oligonucleotides to the beads.

8. The method of claim 1 wherein the beads are encoded by attaching oligonucleotides thereto.
