Abstract

Environmental containment methods ( 16 ) for a flow cytometer ( 21 ) which provides an adjustably controllable environment in which biological particles can be differentiated and droplets separated into collection containers ( 15 ).

Claims (62)

1. A method of maintaining a controlled environment, comprising the steps of:

a. providing an conduit having an orifice element;

b. flowing a liquid in said conduit;

c. generating a fluid stream at said orifice element;

d. collecting said fluid stream in a container;

e. providing an enclosure having an aperture element, wherein said container has a location in said enclosure, and wherein said fluid stream has a trajectory from said orifice element to said container in said enclosure;

f. introducing a mixture of gases into said enclosure which flow through said aperture element; and

g. maintaining said fluid stream having said trajectory from said orifice element to said container in said enclosure within said mixture of gases,

wherein said step of introducing said mixture of gases into said enclosure comprises adjusting percent ratio of gases in said enclosure to a percent ratio of gases different than atmosphere.

2. A method of maintaining a controlled environment as described in claim 1 , wherein said percent ratio of gases different than atmosphere comprises an anaerobic environment.

3. A method of maintaining a controlled environment as described in claim 2 , wherein said anaerobic environment comprises about seven percent hydrogen, about five percent carbon dioxide, and about 88 percent nitrogen.

4. A method of maintaining a controlled environment as described in claim 3 , further comprising the step of detecting oxygen in said enclosure.

5. A method of maintaining a controlled environment as described in claim 4 , further comprising the step of generating a signal in response to detection of said oxygen in said enclosure.

6. A method of maintaining a controlled environment as described in claim 1 , further comprising the step of filtering said mixture of gases prior to introduction into said enclosure.

7. A method of maintaining a controlled environment as described in claim 6 , wherein said step of filter said mixture of gases prior to introduction into said enclosure comprises filtering said mixture of gases through a HEPA filter.

8. A method of maintaining a controlled environment comprising the steps of:

a. providing an conduit having an orifice element;

b. flowing a liquid in said conduit;

c. generating a fluid stream at said orifice element;

d. collecting said fluid stream in a container;

e. providing an enclosure having an aperture element, wherein said container has a location in said enclosure, and wherein said fluid stream has a trajectory from said orifice element to said container in said enclosure;

f. introducing a mixture of gases into said enclosure which flow through said aperture element;

g. maintaining said fluid stream having said trajectory from said orifice element to said container in said enclosure within said mixture of gases; and

h. controlling the temperature of said mixture of gases in said enclosure.

9. A method of maintaining a controlled environment as described in claim 1 or 8 , further comprising the step of entraining material in said fluid stream.

10. A method of maintaining a controlled environment as described in claim 1 or 8 , further comprising the step of entraining particles in said fluid stream.

11. A method of maintaining a controlled environment as described in claim 10 , wherein said particles comprise biological particles.

12. A method of maintaining a controlled environment as described in claim 11 , wherein said biological particles comprise cells.

13. A method of maintaining a controlled environment, comprising the steps of:

a. providing an conduit having an orifice element;

b. flowing a liquid in said conduit;

c. generating a fluid stream at said orifice element;

d. collecting said fluid stream in a container;

e. providing an enclosure having an aperture element, wherein said container has a location in said enclosure, and wherein said fluid stream has a trajectory from said orifice element to said container in said enclosure;

f. introducing a mixture of gases into said enclosure which flow through said aperture element;

g. maintaining said fluid stream having said trajectory from said orifice element to said container in said enclosure within said mixture of gases; and

entraining non-biological particles in said fluid stream.

14. A method of maintaining a controlled environment, comprising the steps of:

a. providing an conduit having an orifice element;

b. flowing a liquid in said conduit;

c. generating a fluid stream at said orifice element;

d. collecting said fluid stream in a container;

e. providing an enclosure having an aperture element, wherein said container has a location in said enclosure, and wherein said fluid stream has a trajectory from said orifice element to said container in said enclosure;

f. introducing a mixture of gases into said enclosure which flow through said aperture element; and

g. maintaining said fluid stream having said trajectory from said orifice element to said container in said enclosure within said mixture of gases; and

h. entraining biological particles in said fluid stream, wherein said biological particles comprise anaerobic organisms.

15. A method of maintaining a controlled environment, comprising the steps of:

a. providing an conduit having an orifice element;

b. flowing a liquid in said conduit;

c. generating a fluid stream at said orifice element;

d. collecting said fluid stream in a container;

e. providing an enclosure having an aperture element, wherein said container has a location in said enclosure, and wherein said fluid stream has a trajectory from said orifice element to said container in said enclosure;

f. introducing a mixture of gases into said enclosure which flow through said aperture element; and

g. maintaining said fluid stream having said trajectory from said orifice element to said container in said enclosure within said mixture of gases; and

h. entraining biological particles in said fluid stream, wherein said biological particles comprise aerobic organisms.

16. A method of maintaining a controlled environment as described in claim 1 , 10 , 13 , 14 or 15 wherein said conduit having an orifice element comprises a flow cytometer nozzle.

17. A method of maintaining a controlled environment as described in claim 16 , further comprising the step of oscillating said fluid stream in which particles are entrained.

18. A method of maintaining a controlled environment as described in claim 17 , further comprising breaking droplets off from said fluid stream below said orifice element of said flow cytometer nozzle, wherein a portion of said droplets contain one of said particles entrained in said fluid stream.

19. A method of maintaining a controlled environment as described in claim 18 , further comprising the step of differentiating said particles contained in said droplets based upon at least one particle characteristic.

20. A method of maintaining a controlled environment as described in claim 19 , further comprising the step of sorting differentiated particles contained in said droplets.

21. A method of maintaining a controlled environment as described in claim 1 , 8 , 13 , 14 or 15 , further comprising the step of filtering said mixture of gases after egress from said enclosure.

22. A method of maintaining a controlled environment as described in claim 21 , wherein said step of filtering said mixture of gases after egress from said enclosure comprises filtering said mixture of gases through a HEPA filter.
