Abstract

Methods are described for the production of neurons or neuronal progenitor cells. Multipotent neural stem cells are proliferated in the presence of growth factors and erythropoietin which induces the generation of neuronal progenitor cells. The erythropoietin may be exogenously applied to the multipotent neural stem cells, or alternatively, the cells can be subjected to hypoxic insult which induces the cells to express erythropoietin.

Claims (11)

1. A method for screening the effects of drugs or other agents on neuronal cells, comprising:

a) preparing a cell culture comprising a cell population enriched in neuronal cells by

(i) inducing multipotent neural stem cells to proliferate;

(ii) exposing said multipotent neural stem cells to hypoxic conditions; and

(iii) allowing said multipotent neural stem cells to differentiate into a differentiated cell population containing neuronal cells, wherein said differentiated cell population is enriched in neuronal cells compared to when step (ii) above is not performed;

b) contacting said enriched differentiated neuronal cell population with at least one drug or other agent; and

c) determining if said drug or other agent has an effect on said enriched differentiated neuronal cell population.

2. The method of claim 1 , wherein said multipotent neural stem cells are exposed to hypoxic conditions for between 1 and 8 hours.

3. The method of claim 1 , wherein said enriched differentiated neuronal cell population contains at least 6% neurons.

4. The method of claim 1 , wherein the source of said multipotent neural stem cells is a human.

5. The method of claim 1 , wherein the source of said multipotent neural stem cells is a fetal mammal.
