Abstract

In one aspect, the invention provides a purified and modified phytase enzyme from Escherichia coli K12 appA phytase. The enzyme has phytase activity and improved thermal tolerance as compared with the wild-type enzyme. In addition, the enzyme has improved protease stability at low pH. Glycosylation of the modified phytase provided a further improved enzyme having improved thermal tolerance and protease stability. The enzyme can be produced from native or recombinant host cells and can be used to aid in the digestion of phytate where desired. In one aspect, the phytase of the present invention can be used in foodstuffs to improve the feeding value of phytate rich ingredients.

Claims (53)

1. A formulation comprising:

(A) a polypeptide having phytase activity wherein the polypeptide is encoded by a nucleic acid comprising a variant of the nucleic acid of SEQ ID NO:7, wherein said variant has a nucleotide sequence which differs from SEQ ID NO:7 solely by:

(i) substituting nucleotide 389 with G, substituting nucleotide 390 with A, substituting nucleotide 437 with T, substituting nucleotide 438 with G, substituting nucleotide 439 with G, substituting nucleotide 470 with C, substituting nucleotide 472 with T, substituting nucleotide 476 with T, substituting nucleotide 477 with G, substituting nucleotide 478 with T, substituting nucleotide 689 with G, substituting nucleotide 690 with A, substituting nucleotide 691 with G, substituting nucleotide 728 with T, substituting nucleotide 729 with A, substituting nucleotide 730 with T, substituting nucleotide 863 with T, substituting 864 with G, substituting nucleotide 1016 with G, or any combination thereof; or

(ii) substituting nucleotide 389 with G, substituting nucleotide 390 with A, substituting nucleotide 437 with T, substituting nucleotide 438 with G, substituting nucleotide 439 with G, substituting nucleotide 470 with C, substituting nucleotide 472 with T, substituting nucleotide 476 with T, substituting nucleotide 477 with G, substituting nucleotide 478 with T, substituting nucleotide 689 with G, substituting nucleotide 690 with A, substituting nucleotide 691 with G, substituting nucleotide 728 with T, substituting nucleotide 729 with A, substituting nucleotide 730 with T, substituting nucleotide 863 with T, substituting 864 with G, and substituting nucleotide 1016 with G;

(B) a polypeptide having phrase activity wherein said polypeptide comprises an amino acid sequence which is a variant of SEQ ID NO:8, wherein said variant differs from SEQ ID NO:8 solely by:

(i) substituting amino acid 68 with E, substituting amino acid 84 with W, substituting amino acid 95 with P, substituting amino acid 97 with C, substituting amino acid 168 with E, substituting amino acid 181 with Y, substituting amino acid 226 with C, substituting amino acid 277 with D, or any combination thereof; or

(ii) substituting amino acid 68 with E, substituting amino acid 84 with W, substituting amino acid 95 with P, substituting amino acid 97 with C, substituting amino acid 168 with E, substituting amino acid 181 with Y, substituting amino acid 226 with C, and substituting amino acid 277 with D;

(C) the polypeptide of (A) or (B) wherein the polypeptide lacks a leader sequence;

(D) the polypeptide of(A), (B) or (C) wherein the polypeptide further comprises a heterologous sequence;

(E) the polypeptide of (D) wherein the heterologous sequence encodes a leader or signal peptide, or an N-terminal identification peptide which provides stabilization or allows for simplified purification;

(F) the polypeptide of (A), (B), (C), (D) or (E) wherein the polypeptide is glycosylated;

(G) the polypeptide of (A), (B), (C), (D), (E) or (F) and a stabilizing agent; or

(H) the polypeptide of (A), (B), (C), (D), (E), (F) or (G) and a stabilizing agent which comprises urea, polyol, sorbitol, and/or glycerol.

2. The formulation of claim 1 , wherein the formulation is or comprises a dietary supplement.

3. The formulation of claim 1 , further comprising at least one probiotic, at least one vitamin, at least one additional enzyme, at least one mineral or metal, at least one herb or plant extract, at least one amino acid or amino acid derivative, or any combination thereof.

The formulation of claim 3 , wherein the mineral or metal is selected from the group consisting of aluminum, antimony, barium, beryllium, bismuth, boron, bromide, bromine, cadmium, calcium, cerium, cesium, chloride, chromium, cobalt, copper, dysprosium, erbium, europium, fluoride, fluorine, gadolinium, gallium, germanium, gold, hafnium, holmium, indium, iodine, iridium, iron, lanthanum, lithium, lutetium, magnesium, manganese, molybdenum, neodymium, nickel, niobium, osmium, palladium, phosphorous, platinum, potassium, praseodymium, promethium, rhenium, rhodium, rubidium, ruthenium, samarium, scandium, selenium, silicon, silver, sodium, strontium, sulfur, tantalum, tellurium, terbium, thorium, thulium, tin, titanium, tungsten, vanadium, ytterbium, yttrium, zinc, zirconium and any combination thereof.

5. The formulation of claim 1 , further comprising an herb, a plant extract, diatomaceous earth, charcoal, choline, inositol, biotin, PABA, alpha-lipoic acid, a carotenoid, beta carotene, coenzyine Q10, chondroitin, melatonin, lecithin, brewer's yeast, or a combination thereof.

The formulation of claim 1 , further comprising alfalfa, ginseng, American ginseng, Asian red ginseng, Asian white ginseng, Siberian ginseng, Brazilian ginseng, astragalus, bilberry, back cohosh, cascara sagrada, cat's claw, cayenne, dong quai, echinacea, eucalyptus, feverfew, garlic, ginkgo biloba, goldenseal, gotu kola, horsetail, maca, a mushroom, Maitake mushroom, Reishi mushroom, Shiitake mushroom, leuzea, rhodiola, milk thistle, noni, pau d'arco, papaya, pygeum, saw palmetto, schizandra, senna, suma, wild yam, willow, yucca, wheat grass, barley grass, parsley, broccoli, acerola cherries, aloe vera, quercitin, pine bark, grape seed, green tea, red wine, grapefruit extract, ginger, oat straw, sarsaparilla, oil, walnut oil, safflower oil, soybean oil, peanut oil, fish oil, salmon oil, evening primrose oil, borage oil, bee pollen, bee propolis, royal jelly, bran, oat bran, wheat bran, fiber, soy, psyllium, apple pectin, protein, egg protein, milk protein, soy protein, rice protein, whey, algae, spirulina, Chlorella, dulse, kelp, D. salina, or a combination thereof.

The formulation of claim 3 , wherein the probiotic is selected from the group consisting of a Lactobacillus species, L. acidophilus, L. bifidus, L. sporogenes, L. casei, L. rhamnosus, L. plantarum, S. thermophilus , a Bifidobacterium species, an Escherichia , an Enterococcus , a Bacillus and a Saccharomyces species.

The formulation of claim 3 , wherein the additional enzyme is selected from the group consisting of a phytase, an amylase, a bromelain, a cellulase, a chymopapain, a diastase, a glucoamylase, a hemicellulase, a hyaluronidase, an invertase, a lactase, a lipase, a maltase, a pancreatin, a papain, a pectinase, a pepsin, a plasmin, a protease, a rennin and any combination thereof.

The formulation of claim 3 , wherein the vitamin is selected from the group consisting of vitamin B, Thiamine (Vitamin B1), Riboflavin (Vitamin B2), Nicotinic acid (Niacin, Vitamin B3), Pantothenic acid (Vitamin B5), Pyridoxine (Vitamin B6), B7, Folic acid (Vitamin B9), Cyanocobalamin (Vitamin B 12), vitamin C, a vitamin D, vitamin D1, vitamin D2, vitamin D3, vitamin E, a vitamin K, vitamin K1, vitamin K2, vitamin G, vitamin H, vitamin P and any combination thereof.

The formulation of claim 3 , wherein the amino acid or amino acid derivative is selected from the group consisting of Isoleucine, Leucine, Lysine, Phenylalanine, Threonine, Tryptophan, Valine, Methionine, Cysteine, Alanine, Arginine, Aspartic Acid, Glutamic Acid, Glycine, Histidine, Proline, Serine, Asparagine, Glutamine, Tyrosine, taurine, glucosamine and any combination thereof.

11. The formulation of claim 1 , comprising vitamin D3 or calcium or both.

12. The formulation of claim 1 , further comprising potassium, glucose, CaCl 2 or a combination thereof.

The formulation of claim 1 , further comprising at least one enzyme selected from the group consisting of an α-galactosidase, a β-galactosidase, a lactase, a phytase, a β-glucanase, an endo-β-1,4-glucanase, an endo-β-1,3(4)-glucanase, a cellulase, a xylosidase, a galactanase, an arabinogalactan endo-1,4-β-galactosidase, an arabinogalactan endo-1,3-β-galactosidase, an endoglucanase, an endo-1 ,2-β-glucanase, an endo-1,3-α-glucanase, an endo-1,3-β-glucanase, a pectin degrading enzyme, a pectinase, a pectinesterase, a pectin lyase, a polygalacturonase, an arabinanase, a rhamnogalacturonase, a rhamnogalacturonan acetyl esterase, a rhamnogalacturonan-α-rhamnosidase, a pectate lyase, an α-galacturonisidase, a mannanase, a β-mannosidase, a mannan acetyl esterase, a xylan acetyl esterase, a protease, a xylanase, an arabinoxylanase, a lipase, a phospholipase, and a cutinase.

14. The formulation of claim 1 , wherein the formulation is in the form of a powder, a tablet, a concentrate, a gel tab, a capsule, a spray, an aerosol, a lotion, an adhesive patch or a drink.

15. A pharmaceutical composition comprising a pharmaceutically acceptable excipient and the formulation of claim 1 .

16. The pharmaceutical composition of claim 15 formulated for oral delivery.

17. The pharmaceutical composition of claim 15 formulated as a pill, a tablet, a capsule, a spray, an aerosol or a powder.

18. A kit comprising the formulation of claim 1 , or the pharmaceutical composition of claim 15 , and instructions on using the formulation or the pharmaceutical composition.

19. An immobilized phytase comprising:

(A) a polypeptide encoded by a nucleic acid comprising a variant of the nucleic acid of SEQ ID NO:7, wherein said variant has a nucleotide sequence which differs from SEQ ID NO:7 solely by:

(i) substituting nucleotide 389 with G, substituting nucleotide 390 with A, substituting nucleotide 437 with T, substituting nucleotide 438 with G, substituting nucleotide 439 with G, substituting nucleotide 470 with C, substituting nucleotide 472 with T, substituting nucleotide 476 with T, substituting nucleotide 477 with G, substituting nucleotide 478 with T, substituting nucleotide 689 with G, substituting nucleotide 690 with A, substituting nucleotide 691 with G, substituting nucleotide 728 with T, substituting nucleotide 729 with A, substituting nucleotide 730 with T, substituting nucleotide 863 with T, substituting 864 with G, substituting nucleotide 1016 with G, or any combination thereo: or

(ii) substituting nucleotide 389 with G, substituting nucleotide 390 with A, substituting nucleotide 437 with T, substituting nucleotide 438 with G, substituting nucleotide 439 with G, substituting nucleotide 470 with C, substituting nucleotide 472 with T, substituting nucleotide 476 with T, substituting nucleotide 477 with G, substituting nucleotide 478 with T, substituting nucleotide 689 with G, substituting nucleotide 690 with A, substituting nucleotide 691 with G, substituting nucleotide 728 with T, substituting nucleotide 729 with A, substituting nucleotide 730 with T, substituting nucleotide 863 with T, substituting 864 with G, and substituting nucleotide 1016 with G;

(B) a polypeptide which comprises an amino acid sequence which is a variant of SEQ ID NO:8, wherein said variant differs from SEQ ID NO:8 solely by:

(i) substituting amino acid 68 with E, substituting amino acid 84 with W, substituting amino acid 95 with P, substituting amino acid 97 with C, substituting amino acid 168 with E, substituting amino acid 181 with Y, substituting amino acid 226 with C, substituting amino acid 277 with D, or any combination thereof; or

(ii) substituting amino acid 68 with E, substituting amino acid 84 with W, substituting amino acid 95 with P, substituting amino acid 97 with C, substituting amino acid 168 with E, substituting amino acid 181 with Y, substituting amino acid 226 with C, and substituting amino acid 277 with D;

(C) the polypeptide of (A) or (B) wherein the polypeptide lacks a leader sequence;

(D) the polypeptide of (A), (B) or (C) wherein the polypeptide further comprises a heterologous sequence;

(E) the polypeptide of (D) wherein the heterologous sequence encodes a leader or signal peptide, or an N-terminal identification peptide which provides stabilization or allows for simplified purification; or

(F) the polypeptide of (A), (B), (C), (D) or (E) wherein the polypeptide is glycosylated.

20. The immobilized phytase of claim 19 , wherein the polypeptide is immobilized to a bead.

21. The immobilized phytase of claim 20 , wherein the polypeptide is immobilized to a polysorb bead or a polystyrene bead.

22. A dietary supplement comprising the immobilized phytase of claim 19 .

23. A pharmaceutical composition comprising the immobilized phytase of claim 19 .

24. A fertilizer or soil additive comprising the immobilized phytase of claim 19 .

25. A fertilizer or soil additive comprising the formulation of claim 1 .

26. A liquid supplement comprising: the formulation of claim 1 , or the pharmaceutical composition of claim 15 .

27. The liquid supplement of claim 26 , further comprising glucose, potassium, sodium, or calcium.

28. A hydrating agent comprising a formulation of claim 1 .

29. The hydrating agent of claim 28 , further comprising glucose, potassium, sodium or calcium.

30. A tissue culture, cell culture media, or a cell culture media additive, comprising the formulation of claim 1 .

31. A plant food additive comprising the formulation of claim 1 .
