Abstract

The present invention provides a purified and isolated nucleic acid encoding a sodium/iodide symporter. The present invention also provides purified sodium/iodide symporter, a vector comprising nucleic acid encoding sodium/iodide symporter, a host cell transformed with the vector, and a method for producing recombinant sodium/iodide symporter. In addition, the present invention provides nucleic acid probes and mixtures thereof specific for sodium/iodide symporter nucleic acid and antibodies immunoreactive with sodium/iodide symporter. The present invention also provides a method for diagnosing and treating thyroid disorders associated with non-functional sodium/iodide symporter. Furthermore, the present invention provides a method for the selective ablation of tissue. The present invention also provides a method for identifying an iodide transport protein in non-thyroid tissue. Finally, the present invention provides a non-human, transgenic model for a thyroid disorder.

Claims (13)

1. An isolated labeled nucleic acid molecule consisting of (i) at least 8 contiguous nucleotides from the coding region of a nucleic acid encoding the amino acid sequence set forth in SEQ ID NO:2 or from the complement of the coding region of a nucleic acid encoding the amino acid sequence set forth in SEQ ID NO:2, and (ii) a detectable label bound to the nucleic acid molecule by a method selected from the group consisting of polymerase chain reaction (PCR), nick translation, end labeling, fill-in end labeling, polynucleotide kinase exchange reaction and random priming.

2. The labeled nucleic acid molecule of claim 1 , wherein the nucleic acid molecule consists of at least 8 contiguous nucleotides from the coding region of the sequence set forth in SEQ ID NO:1 or its complement.

3. The labeled nucleic acid molecule of claim 1 , where the nucleic acid is DNA or RNA.

4. The isolated labeled nucleic acid molecule of claim 1 , wherein the detectable label is bound to the nucleic acid molecule using polymerase chain reaction (PCR).

5. The isolated labeled nucleic acid molecule of claim 1 , wherein the detectable label is bound to the nucleic acid molecule using nick translation.

6. The isolated labeled nucleic acid molecule of claim 1 , wherein the detectable label is bound to the nucleic acid molecule using end labeling.

7. The isolated labeled nucleic acid molecule of claim 1 , wherein the detectable label is bound to the nucleic acid molecule using fill-in end labeling.

8. The isolated labeled nucleic acid molecule of claim 1 , wherein the detectable label is bound to the nucleic acid molecule using polynucleotide kinase exchange reaction.

9. The isolated labeled nucleic acid molecule of claim 1 , wherein the detectable label is bound to the nucleic acid molecule using random priming.

10. The isolated labeled nucleic acid molecule of claim 1 , wherein the detectable label is a fluorescent label.

11. The isolated labeled nucleic acid molecule of claim 1 , wherein the detectable label is biotin.

12. The isolated labeled nucleic acid molecule of claim 1 , wherein the detectable label is a radioactive label.

13. The isolated labeled nucleic acid molecule of claim 1 , wherein the detectable label is 35 S, 32 P or 3 H.
