Abstract

Disclosed are methods for sterilizing cross-linked gelatin as well as to sterilized cross-linked gelatin. In particular, the methods of this invention employ E-beam sterilization techniques.

Claims (22)

1. A method for preparing a sterile, cross-linked gelatin composition for use in vivo, comprising:

providing a cross-linked gelatin composition;

exposing the cross-linked gelatin composition to a dose of E-beam irradiation, the dose of E-beam irradiation being sufficient to sterilize the cross-linked gelatin composition, and the dose of E-beam irradiation being sufficient to increase the tensile strength of the cross-linked gelatin composition to a tensile strength of at least about 2.0 lbs/in 2 .

2. The method of claim 1 wherein the composition after the irradiation has a water absorbability of at least about 30 grams of water per gram of the composition.

3. The method of claim 1 wherein the composition further comprises an antimicrobial agent, a growth factor, a thrombus enhancing agent, or mixtures thereof.

4. The method of claim 3 wherein the dose of the irradiation is from about 17 to about 25 kGy.

5. The method of claim 1 , in which the dose of E-beam irradiation is sufficient to increase the tensile strength of the cross-linked gelatin composition by more than approximately 80%.

6. A method for preparing a sterile, cross-linked gelatin composition for use in vivo, comprising:

providing a cross-linked gelatin composition;

exposing the cross-linked gelatin composition to a dose of E-beam irradiation, the dose of E-beam irradiation being sufficient to sterilize the cross-linked gelatin composition, the dose of E-beam irradiation being sufficient to retain no less than approximately 85% of the water absorbability of the cross-linked gelatin composition, and the dose of E-beam irradiation being sufficient to increase the tensile strength of the composition to a tensile strength of at least about 2.0 lbs/in 2 .

7. The method of claim 6 wherein the composition after the irradiation has a water absorbability of at least about 30 grams of water per gram of the composition.

8. The method of claim 6 wherein the composition further comprises an antimicrobial agent, a growth factor, a thrombus enhancing agent, or mixtures thereof.

9. The method of claim 8 wherein the dose of the irradiation is from about 17 to about 25 kGy.

10. A method for preparing a package of sterile, cross-linked gelatin composition for use in vivo, comprising:

providing a packaging element;

providing a cross-linked gelatin composition;

enclosing the cross-linked gelatin composition in the packaging element; and

exposing the packaging element to a dose of E-beam irradiation, the dose of E-beam irradiation being sufficient to sterilize the entire package, and the dose of E-beam irradiation being sufficient to increase the tensile strength of the cross-linked gelatin composition to a tensile strength of at least about 2.0 lbs/in 2 .

11. The method of claim 10 wherein the composition after the irradiation has a water absorbability of at least about 30 grams of water per gram of the composition.

12. The method of claim 10 wherein the composition further comprises an antimicrobial agent, a growth factor, a thrombus enhancing agent, or mixtures thereof.

13. The method of claim 12 wherein the dose of the irradiation is from about 17 to about 25 kGy.

14. The method of claim 10 , in which the dose of E-beam irradiation is sufficient to increase the tensile strength of the cross-linked gelatin composition by more than approximately 80%.
