Abstract

This invention relates to plant breeding and the protection of plants from insects. More specifically, this invention includes novel transformation events of cotton plants comprising one or more polynucleotide sequences, as described herein, inserted into specific site(s) within the genome of a cotton cell. In highly preferred embodiments, said polynucleotide sequences encode “stacked” Cry1F and Cry1Ac lepidopteran insect inhibitory proteins. However, the subject invention includes plants having single cry1F or cry1Ac events, as described herein. Additionally, the invention is related to cotton plants derived from that transformation event and to assays for detecting the presence of the event in a sample. More specifically, the present invention provides DNA and related assays for detecting the presence of certain insect-resistance events in cotton. The assays are based on the DNA sequences of recombinant constructs inserted into the cotton genome and of the genomic sequences flanking the insertion sites. These sequences are unique. Based on these insert and border sequences, event-specific primers were generated. PCR analysis demonstrated that these cotton lines can be identified in different cotton genotypes by analysis of the PCR amplicons generated with these event-specific primer sets. Thus, these and other related procedures can be used to uniquely identify these cotton lines. Kits and conditions useful in conducting the assays are also provided. These materials and methods can also be used to assist breeding programs to further develop traits in cotton.

Claims (18)

1. A cotton seed comprising in its genome cry1F cotton event 281-24-236 and cry1Ac cotton event 3006-210-23 and having representative seed deposited with American Type Culture Collection (ATCC) with Accession No. PTA-6233.

2. A cotton plant produced by growing the seed of claim 1 .

3. A cotton seed produced by a plant of claim 2 wherein said seed comprises in its genome cry1F cotton event 281-24-236 as present in a cotton seed deposited with American Type Culture Collection (ATCC) with Accession No. PTA-6233.

4. A cotton seed produced by a plant of claim 2 wherein said seed comprises in its genome cry1Ac cotton event 3006-210-23 as present in a cotton seed deposited with American Type Culture Collection (ATCC) with Accession No. PTA-6233.

5. A part of the cotton plant of claim 2 wherein said part is selected from the group consisting of pollen, ovule, flowers, bolls, shoots, roots, and leaves and said part comprises at least one of said events.

6. An insect-resistant progeny cotton plant of the plant of claim 2 wherein said insect resistance is due to expression of at least one of said events in said genome.

7. A cotton plant comprising a genome comprising a DNA sequence selected from residues 2055–12,768 of SEQ ID NO:1 and 508–8920 of SEQ ID NO:2.

8. The plant of claim 7 wherein said cotton plant comprises residues 2055–12,768 of SEQ ID NO:1.

9. The plant of claim 7 wherein said cotton plant comprises residues 508–8920 of SEQ ID NO:2.

10. A cotton seed comprising a genome comprising SEQ ID NO:1.

11. A cotton seed comprising a genome comprising SEQ ID NO:2 in its genome.

12. The cotton seed of claim 10 further comprising SEQ ID NO:2 in its genome.

13. A plant produced by growing a seed of claim 10 .

14. A plant produced by growing a seed of claim 11 .

15. A plant produced by growing a seed of claim 12 .

16. A cotton seed produced by a plant of claim 7 and comprising a DNA sequence selected from residues 2,055–12,768 of SEQ ID NO:1 and 508–8,920 of SEQ ID NO:2.

17. A cotton plant produced by growing the seed of claim 3 .

18. A cotton plant produced by growing the seed of claim 4 .
