Abstract

The present invention provides a composition comprising fluorescent semiconductor nanocrystals associated to a compound, wherein the nanocrystals have a characteristic spectral emission, wherein said spectral emission is tunable to a desired wavelength by controlling the size of the nanocrystal, and wherein said emission provides information about a biological state or event.

Claims (33)

1. A method of detecting an interaction between a composition and a biological moiety comprising:

providing a first composition capable of a characteristic spectral emission, the composition comprising a first compound capable of interacting non-covalently with a first biological moiety and a first semiconductor nanocrystal associated with the first compound;

providing a second composition capable of a characteristic spectral emission and capable of interacting non-covalently with a second biological moiety;

allowing a sample comprising at least said first or second biological moieties to non-covalently interact with the first composition and the second composition;

exciting the first composition and the second composition with an excitation source, thereby producing a spectral emission; and

detecting an interaction of at least one biological moiety by monitoring the spectral emission.

The method of claim 1 , wherein monitoring the spectral emission occurs in assays selected from the group consisting of: immunochemistry, immunocytochemistry, immunobiology, immunofluorescence, DNA sequence analysis, fluorescence resonance energy transfer, flow cytometry, fluorescence activated cell sorting, diagnostics in biological systems, and high-throughput screening.

3. The method of claim 1 , wherein the first composition comprises a molecular complex with a molecule associated with the nanocrystal complexed to a second molecule that interacts with the biological moiety.

4. The method of claim 1 , wherein the spectral emission is tunable to a desired wavelength by controlling the size of the nanocrystal.

5. The method of claim 1 , wherein the noncovalent interaction comprises hydrophobic interaction, hydrophilic interaction, electrostatic interaction, van der Waals interaction, or magnetic interaction.

6. The method of claim 1 , wherein the first biological moiety comprises a small molecule.

7. The method of claim 1 , wherein the first biological moiety comprises a protein, peptide or antibody.

8. The method of claim 1 , wherein the first biological moiety comprises a nucleic acid.

9. The method of claim 8 , wherein the nucleic acid comprises DNA or RNA.

10. The method of claim 1 , wherein the first biological moiety comprises an amino acid.

11. The method of claim 1 , wherein the first biological moiety comprises a ligand.

12. The method of claim 1 , wherein the first biological moiety comprises an antigen.

13. The method of claim 1 , wherein the first biological moiety comprises a cell.

14. The method of claim 1 , wherein the first biological moiety comprises a subcellular organelle.

15. The method of claim 1 , wherein the semiconductor nanocrystal is water-soluble.

16. The method of claim 1 , wherein the semiconductor nanocrystal comprises a core comprising a first semiconductor material, and a layer overcoating the core comprising a second semiconductor material.

17. The method of claim 1 , wherein the spectral emission provides information about a biological state or event.

18. The method of claim 17 , wherein the spectral emission provides information about the amount of biological moiety in the sample.

19. The method of claim 17 , wherein the spectral emission provides information about the presence of the biological moiety in the sample.

20. The method of claim 19 , wherein the biological state or event includes: biological interactions, biological processes, alterations of biological processes, alterations of biological moieties, structure of biological moieties, composition of biological moieties, conformation of biological moieties, or localization of biological moieties.

21. The method of claim 1 , wherein the characteristic spectral emission of the first composition is distinct from the characteristic spectral emission of the second composition.

22. The method of claim 21 , wherein the second composition includes an organic fluorescent dye.

23. The method of claim 21 , wherein the second composition includes a second compound capable of interacting non-covalently with the second biological moiety and a second semiconductor nanocrystal associated with the second compound.

24. A method of detecting an interaction between a composition and a biological moiety comprising:

combining a sample comprising a biological moiety with a first composition, wherein the first composition is capable of a spectral emission and comprises a compound and a semiconductor nanocrystal conjugated to the compound;

providing a second composition capable of a characteristic spectral emission and capable of interacting non-covalently with a second biological moiety;

exciting the semiconductor nanocrystal; and

monitoring the spectral emission of the sample to detect a non-covalent interaction between the first composition and the biological moiety or between the second composition and the second biological moiety.
