Rapidly disintegrating methylcellulose tablets
View Patent ↗The present invention relates to a rapidly disintegrating tablet for oral administration. The tablet has a first phase and a second phase blended with the first phase. The first phase has a compacted mixture of methylcellulose having a viscosity of >1000 centipoise and a diluent. The methylcellulose is the sole active ingredient in the first phase. There is also a process for preparing a rapidly disintegrating tablet.
1. A rapidly disintegrating tablet for oral administration comprising
a first phase having a compacted mixture of methylcellulose with a viscosity of >1000 centipoise, and a diluent, wherein the methylcellulose is the sole active, wherein the diluent is an edible calcium salt; and
a second phase that is blended with the first phase wherein the second phase includes at least two ingredients selected from the group consisting of a disintegrating agent, a wetting agent, a lubricant, and a second diluent.
2. The tablet of claim 1 , wherein the ratio of methylcellulose to diluent in the first phase is from about 2:1 to about 4:1.
3. The tablet of claim 1 , wherein the edible calcium salt is selected from the group consisting of dibasic calcium phosphate dihydrate, calcium phosphate anhydrous, tribasic calcium phosphate, and any mixtures thereof.
4. The tablet of claim 1 , wherein the first phase includes a disintegrating agent.
5. The tablet of claim 4 , wherein the disintegrating agent is present in an amount about 3% w/w to about 8% w/w.

The tablet of claim 4 , wherein the disintegrating agent is selected from the group consisting of sodium starch glycolate, sodium carboxymethylcellulose, sodium croscarmellose, carboxymethylcellulose, veegum, alginate, agar, tragacanth, locust bean, karaya, pectin, crospovidone, and any mixtures thereof.
7. The tablet of claim 1 , wherein the first phase includes a binding agent.
8. The tablet of claim 7 , wherein the binding agent is present in an amount about 4% w/w to about 6.5% w/w.
9. The tablet of claim 7 , wherein the binding agent is selected from the group consisting of PVP, hydroxypropylcellulose, hydroxypropyl methylcellulose, acacia, gelatin, tragacanth, pregelatinized starch, starch, and any mixtures thereof.
10. The tablet of claim 1 , wherein the second phase includes at least three ingredients selected from the group consisting of a disintegrating agent, a wetting agent, a lubricant, and a second diluent.
11. The tablet of claim 1 , wherein the second phase includes a wetting agent.
12. The tablet of claim 11 , wherein the wetting agent is selected from the group consisting of sodium lauryl sulfate, magnesium lauryl sulfate, and any mixture thereof.
13. The tablet of claim 1 , wherein the second phase includes a lubricant.
14. The tablet of claim 13 , wherein the lubricating agent is selected from the group consisting of magnesium stearate, calcium stearate, sodium stearate, colloidal silicon dioxide, Syloid, stearic acid, talc, and any mixtures thereof.
15. The tablet of claim 1 , wherein the second phase includes a second diluent.
16. The tablet of claim 15 , wherein the second diluent is selected from the group consisting of microcrystalline cellulose, corn starch, pregelatinized starch, and any mixtures thereof.
17. The tablet of claim 1 , wherein the tablet is a laxative.
18. A process of preparing a rapidly disintegrating tablet comprising
granulating methylcellulose and granulating a diluent, wherein the diluent is an edible calcium salt
blending the granulated methylcellulose and the granulated diluent together to form a first phase;
blending a second phase including at least two ingredients selected from the group consisting of a disintegrating agent, a wetting agent, a lubricant, and a second diluent;
mixing together the first phase and the second phase to form the tablet.
19. The process of claim 18 , wherein the edible calcium salt is selected from the group consisting of dibasic calcium phosphate dihydrate, calcium phosphate anhydrous, tribasic calcium phosphate, and any mixtures thereof.
20. The process of claim 18 , wherein the ratio of methylcellulose to diluent in the first phase is from about 2:1 to about 4:1.
21. The process of claim 18 , wherein the first phase includes a disintegrating agent.
22. The process of claim 21 , wherein the disintegrating agent is present in an amount about 3% w/w to about 8% w/w.

The process of claim 21 , wherein the disintegrating agent of the first phase is selected from the group consisting of sodium starch glycolate, sodium carboxymethylcellulose, sodium croscarmellose, carboxymethylcellulose, veegum, alginate, agar, tragacanth, locust bean, karaya, pectin, crospovidone, and any mixtures thereof.
24. The process of claim 18 , wherein the first phase includes a binding agent.
25. The process of claim 24 , wherein the binding agent is present in an amount about 4% w/w to about 6.5% w/w.
26. The process of claim 24 , wherein the binding agent is selected from the group consisting of PVP, hydroxypropylcellulose, hydroxypropyl methylcellulose, acacia, gelatin, tragacanth, pregelatinized starch, starch, and any mixtures thereof.
27. The process of claim 18 , wherein the second phase includes a wetting agent.
28. The process of claim 27 , wherein the wetting agent is selected from the group consisting of sodium lauryl sulfate, magnesium lauryl sulfate, and any mixture thereof.
29. The process of claim 18 , wherein the second phase includes a disintegrating agent.
30. The process of claim 29 , wherein the disintegrating agent is present in an amount about 3% w/w to about 8% w/w.
31. The process of claim 29 , wherein the disintegrating agent of the second phase is selected from the group consisting of sodium starch glycolate, sodium carboxymethylcellulose, sodium croscarmellose, carboxymethylcellulose, veegum, alginate, agar, tragacanth, locust bean, karaya, pectin, crospovidone, and any mixtures thereof.
32. The process of claim 18 , wherein the second phase includes a lubricant.
33. The process of claim 32 , wherein the lubricating agent is selected from the group consisting of magnesium stearate, calcium stearate, sodium stearate, colloidal silicon dioxide, Syloid, stearic acid, talc, and any mixtures thereof.
34. The process of claim 18 , wherein the second phase includes at least three ingredients selected from the group consisting of a disintegrating agent, a wetting agent, a lubricant, and a second diluent.
