Abstract

Apparatus and methods are disclosed for treating a sample by selectively controlling sonic energy and/or selectively controlling the location of the sample relative to the sonic energy.

Claims (47)

1. A method for achieving a desired state of fluidization of a sample, comprising:

providing a focused acoustic field to a sample, the focused acoustic field having a frequency of between about 100 kilohertz and about 100 megahertz and a focal zone having a diameter of less than about 2 centimeters;

monitoring emissions of acoustic energy from the sample;

determining the state of fluidization of the sample based, at least in part, on the emissions of acoustic energy from the sample; and

providing a modified focused acoustic field to the sample, the modified focused acoustic field having acoustic wave variables selected, at least in part, based on the determined state of fluidization to achieve the desired state of fluidization of the sample, wherein the selected acoustic wave variables include a frequency of between about 100 kilohertz and about 100 megahertz and a focal zone having a diameter of less than about 2 centimeters.

2. The method of claim 1 , wherein the focused acoustic field has a focal zone smaller than a size of the sample, and the method includes: moving relative positions of the sample and the focal zone;

monitoring the emissions of the acoustic energy from the sample at a plurality of relative positions; and

determining the state of fluidization of the sample based, at least in part, on the emissions of acoustic energy from the sample at the plurality of relative positions.

The method of claim 1 , wherein the sample includes a plurality of samples and the providing step includes providing a plurality of the focused acoustic fields, each to a particular sample, and the method includes concurrently monitoring emissions of acoustic energy from each particular sample, and determining the state of fluidization of each particular sample based, at least in part, on the emissions of acoustic energy from each particular sample.

The method of claim 1 , wherein the sample includes a plurality of samples and the providing step includes providing a plurality of the focused acoustic fields, each to a particular sample, and the method includes concurrently monitoring reflections of acoustic energy from each particular sample, and determining the state of fluidization of each particular sample based, at least in part, on the reflections of acoustic energy from each particular sample.

The method of claim 1 , wherein the providing step includes providing the focused acoustic field to a plurality of samples, and the method includes concurrently monitoring emissions from each of the plurality of samples, and determining the state of fluidization of each of the plurality of samples based, at least in part, on the emissions from each of the plurality of samples.

6. The method of claim 1 , including flowing the sample through the focused acoustic field.

7. The method of claim 1 , including providing the focused acoustic field to a reaction vessel containing the sample.

8. The method of claim 1 , including concurrently monitoring an optical response signal from the sample, and determining the state of fluidization based, at least in part, on the optical response signal.

9. The method of claim 8 , wherein the optical response signal includes at least one of reflective, absorptive and fluorescent spectral content.

10. The method of claim 1 , including concurrently monitoring temperature information from the sample, and determining the state of fluidization based, at least in part, on the temperature information.

11. The method of claim 1 , wherein the sample includes organic material.

12. The method of claim 1 , wherein the sample includes inorganic material.

13. The method of claim 1 , wherein the sample includes a mineral.

14. The method of claim 1 , wherein the sample includes a biological material.

15. The method of claim 1 , including suspending the sample in a fluid.

16. The method of claim 15 , wherein the fluid includes a solvent.

17. A method for achieving a desired state of mixing of a sample with a constituent comprising:

providing a focused acoustic field to a sample, the focused acoustic field having a frequency of between about 100 kilohertz and about 100 megahertz and a focal zone having a diameter of less than about 2 centimeters;

monitoring acoustic energy from the sample;

determining the state of mixing of the sample with the constituent based, at least in part, on the acoustic energy from the sample; and

providing a modified focused acoustic field to the sample, the modified focused acoustic field having acoustic wave variables selected, at least in part, based on the determined state of mixing to achieve the desired state of mixing of the sample, wherein the selected acoustic wave variables include a frequency of between about 100 kilohertz and about 100 megahertz and a focal zone having a diameter of less than about 2 centimeters.

18. The method of claim 17 , wherein the acoustic energy from the sample includes reflected acoustic energy.

19. The method of claim 17 , wherein the acoustic energy from the sample includes emitted acoustic energy.

20. The method of claim 17 , wherein the focused acoustic field has a focal zone smaller than a size of the sample, and the method includes:

moving relative position of the sample and the focal zone;

monitoring the acoustic energy from the sample at a plurality of relative positions; and

determining the state of mixing of the sample with the constituent based, at least in part, on the acoustic energy from the sample at the plurality of relative positions.

21. The method of claim 17 , wherein the providing step includes providing a plurality of the focused acoustic fields, each to a particular sample and associated constituent, and the method includes concurrently monitoring acoustic energy from each particular sample, and determining the state of mixing of each particular sample with its associated constituent based, at least in part, on the acoustic energy from each particular sample.

The method of claim 17 , wherein the providing step includes providing the focused acoustic field to a plurality of samples, and the method includes concurrently monitoring acoustic energy from each of the plurality of samples, and determining the state mixing of each of the plurality of samples with an associated constituent based, at least in part, on the acoustic energy from each of the plurality of samples.

23. The method of claim 17 , including the step of flowing the sample and the constituent through the focused acoustic field.

24. The method of claim 17 , including providing the focused acoustic field to a reaction vessel containing the sample and the constituent.

25. The method of claim 17 , including concurrently monitoring an optical response signal from the sample, and determining the state of mixing of the sample with the constituent based, at least in part, on the optical response signal.

26. The method of claim 25 , wherein the optical response signal includes at least one of reflective, absorptive and fluorescent spectral content.

27. The method of claim 17 , including concurrently monitoring temperature information from at least one of the sample and the constituent, and determining the state of mixing of the sample with the constituent based, at least in part, on the temperature information.

28. The method of claim 17 , wherein the constituent includes a solvent.

29. The method of claim 17 , wherein sample includes a first compound and the constituent includes a second compound, different from the first compound.

30. The method of claim 17 , wherein the sample includes an antibody and the constituent includes a molecule to which the antibody binds.

31. The method of claim 17 , wherein the sample includes a substrate and the constituent includes a ligand.

32. The method of claim 17 , wherein the sample includes at least one of an antibody and a receptor and the constituent include a support surface for immobilizing the at least one of the antibody and the receptor.

33. The method of claim 17 , wherein the sample includes a first nucleic acid and the constituent includes a second nucleic acid, different from the first nucleic acid.

34. The method of claim 33 , wherein the first nucleic acid molecule is a primer and the second nucleic acid is a substrate.
