Abstract

The genome sequences and the nucleotide sequences coding for the PWD circovirus polypeptides, such as the circovirus structural and non-structural polypeptides, vectors including the sequences, and cells and animals transformed by the vectors are provided. Methods for detecting the nucleic acids or polypeptides, and kits for diagnosing infection by a PWD circovirus, also are provided. Method for selecting compounds capable of modulating the viral infection is further provided. Pharmaceutical, including vaccines, compositions for preventing and/or treating viral infections caused by PWD circovirus and the use of vectors for preventing and/or treating diseases also are provided.

Claims (30)

1. A vector capable of expressing at least one polypeptide encoded by a nucleic acid sequence comprising a sequence having at least 90% sequence identity to SEQ. ID. NO: 25.

2. The vector of claim 1 , wherein the vector is a baculovirus vector.

3. A vector capable of expressing at least one polypeptide encoded by a nucleic acid sequence comprising a sequence having at least 95% sequence identity to SEQ. ID. NO: 25.

4. The vector of claim 3 , wherein the vector is a baculovirus vector.

5. A vaccine composition, comprising the vector of claim 1 and a pharmaceutically or veterinarily acceptable carrier, wherein said nucleic acid sequence encodes an immunogenic protein that induces a protective response effective against infection by a piglet weight loss disease virus.

6. A vaccine composition, comprising the vector of claim 2 and a pharmaceutically or veterinarily acceptable carrier, wherein said nucleic acid sequence encodes an immunogenic protein that induces a protective response effective against infection by a piglet weight loss disease virus.

7. A vaccine composition, comprising the vector of claim 3 and a pharmaceutically or veterinarily acceptable carrier, wherein said nucleic acid sequence encodes an immunogenic protein that induces a protective response effective against infection by a piglet weight loss disease virus.

8. A vaccine composition, comprising the vector of claim 4 and a pharmaceutically or veterinarily acceptable carrier, wherein said nucleic acid sequence encodes an immunogenic protein that induces a protective response effective against infection by a piglet weight loss disease virus.

9. An immunogenic composition, comprising an immunizing amount of the vector of claim 1 and a pharmaceutically or veterinarily acceptable carrier.

10. An immunogenic composition, comprising an immunizing amount of the vector of claim 2 and a pharmaceutically or veterinarily acceptable carrier.

11. An immunogenic composition, comprising an immunizing amount of the vector of claim 3 and a pharmaceutically or veterinarily acceptable carrier.

12. An immunogenic composition, comprising an immunizing amount of the vector of claim 4 and a pharmaceutically or veterinarily acceptable carrier.

13. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 5 .

14. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 6 .

15. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 7 .

16. A method for treating or preventing porcine circovirus type B infection in a mammalian subject, comprising administering to said subject a therapeutically effective amount of a vaccine according to claim 8 .

17. A recombinant polynucleotide comprising a sequence having at least 90% sequence identity to SEQ. ID. NO: 25.

18. A recombinant polynucleotide comprising a sequence having at least 95% sequence identity to SEQ. ID. NO: 25.

19. A method for detecting and quantifying at least one polynucleotide of a porcine circovirus type B (PCVB) by a hybridization method, wherein said polynucleotide comprises a sequence with at least 90% identity to SEQ ID NO: 25.

20. A method for detecting and quantifying at least one polynucleotide of a porcine circovirus type B (PCVB) by a hybridization method, wherein said polynucleotide comprises a sequence with at least 95% identity to SEQ ID NO: 25.

21. A method for expressing ORF′2 of a porcine circovirus type B (PCVB) in vitro, comprising transfecting cells in culture with the vector of claim 1 .

22. A method for expressing ORF′2 of a porcine circovirus type B (PCVB) in vitro, comprising transfecting cells in culture with the vector of claim 2 .

23. The method of claim 22 , wherein the cells are SF9 cells.

24. A method for expressing ORF′2 of a porcine circovirus type B (PCVB) in vitro, comprising transfecting cells in culture with the vector of claim 3 .

25. A method for expressing ORF′2 of a porcine circovirus type B (PCVB) in vitro, comprising transfecting cells in culture with the vector of claim 4 .

26. The method of claim 25 , wherein the cells are SF9 cells.

27. A method for expressing ORF′2 of a porcine circovirus type B (PCVB) in vivo, comprising administering the vector of claim 1 to a mammal.

28. The method of claim 27 , wherein the mammal is a pig.

29. A method for expressing ORF′2 of a porcine circovirus type B (PCVB) in vivo, comprising administering the vector of claim 3 to a mammal.

30. The method of claim 29 , wherein the mammal is a pig.
