Abstract

The present invention relates to a canine pre-proGHRH polypeptide, a canine mature GHRH peptide, an isolated polynucleotide which encodes the canine pre-proGHRH or the canine mature GHRH. The invention also encompasses vectors encoding and expressing the canine pre-proGHRH or the canine GHRH which can be used to treat disease and growth hormone deficiencies by gene therapy in vertebrates, in particular in dogs.

Claims (32)

1. An expression vector for expressing a canine pro-GHRH deleted of the propeptide N-terminus comprising a polynucleotide operatively linked to a promoter and optionally to an enhancer, wherein the polynucleotide comprises

(a) a polynucleotide encoding a canine proGHRH N-terminus deleted polypeptide having at least 95% sequence identity to amino acids 31 to 106 of SEQ ID NO: 1; or

(b) a polynucleotide having at least 95% sequence identity to nucleotides 91 to 321 of SEQ ID NO:3 and encoding a canine proGHRH N-terminus deleted polypeptide; and

(c) a polynucleotide encoding a secretory signal peptide sequence fused to the nucleotide base sequence from nucleotide 91 to nucleotide 321 of SEQ ID NO: 3;

wherein the canine pro-GHRH polypeptide deleted of the propeptide N-terminus, when expressed in vivo in a canine is able to be cellularly processed to mature canine GHRH with the ability to stimulate growth hormone secretion.

2. An expression vector for expressing a canine mature GHRH comprising a polynucleotide operatively linked to a promoter and optionally to an enhancer, wherein the polynucleotide comprises

(a) a polynucleotide encoding a canine mature GHRH polypeptide having at least 95% sequence identity to SEQ ID NO: 2; or

(b) a polynucleotide having at least 97% sequence identity to SEQ ID NO:4 and encoding a canine mature GHRH; and

(c) a polynucleotide encoding a secretory signal peptide sequence for the secretion of the canine GHRH of SEQ ID NO: 4, fused to the nucleotide sequence of SEQ ID NO: 4;

wherein the canine mature GHRH polypeptide, when expressed in vivo in a canine is able to be cellularly processed to mature canine GHRH with the ability to stimulate growth hormone secretion.

3. The expression vector of claim 1 , wherein the signal peptide sequence fused to the nucleotide base sequence is an IGF-1 signal peptide.

4. The expression vector of claim 2 , wherein the signal peptide sequence fused to the nucleotide base sequence is an IGF-1 signal peptide.

5. A formulation comprising the vector of claim 1 and a pharmaceutically or veterinarily acceptable carrier, vehicle or excipient.

6. A formulation comprising the vector of claim 2 and a pharmaceutically or veterinarily acceptable carrier, vehicle or excipient.

7. The formulation of claim 5 , wherein the carrier, vehicle or excipient facilitates transfection and/or improves preservation of the vector or protein.

8. The formulation of claim 6 , wherein the carrier, vehicle or excipient facilitates transfection and/or improves preservation of the vector or protein.

9. The vector of claim 1 wherein the polynucleotide has at least 96% identity to SEQ ID NO:3.

10. The vector of claim 1 wherein the polynucleotide has at least 98% identity to SEQ ID NO:3.

11. The vector of claim 1 wherein the polynucleotide has at least 98% identity to SEQ ID NO:3.

12. The vector of claim 1 wherein the polynucleotide has the nucleic acid sequence set forth in SEQ ID NO:3.

13. The vector of claim 1 wherein the canine proGHRH N-terminus deleted polypeptide has at least 98% identity to amino acids 31 to 106 of SEQ ID NO:1.

14. The vector of claim 1 wherein the canine proGHRH N-terminus deleted polypeptide has the sequence set forth in amino acids 31 to 106 of SEQ ID NO:1.

15. The vector of claim 2 wherein the polynucleotide has at least 99% identity to SEQ ID NO:4.

16. The vector of claim 2 wherein the polynucleotide has the nucleic acid sequence set forth in SEQ ID NO:4.

17. The vector of claim 2 wherein the canine mature GHRH polypeptide has at least 97% identity to SEQ ID NO:2.

18. The vector of claim 2 wherein the canine mature GHRH polypeptide has at least 98% identity to SEQ ID NO:2.

19. The vector of claim 2 wherein the canine mature GHRH polypeptide has the sequence as set forth in SEQ ID NO:2.

20. An expression vector for expressing a canine pro-GHRH comprising a polynucleotide operatively linked to a promoter and optionally to an enhancer, wherein the polynucleotide comprises

(a) a polynucleotide encoding a canine proGHRH polypeptide having at least 95% sequence identity to amino acids 21 to 106 of SEQ ID NO: 1; or

(b) a polynucleotide having at least 95% sequence identity to nucleotides 61 to 321 of SEQ ID NO: 3 and encoding a canine proGHRH; and

(c) a polynucleotide encoding a secretory signal peptide sequence fused to the nucleotide base sequence from nucleotide 61 to nucleotide 321 of SEQ ID NO: 3;

wherein the canine pro-GHRH polypeptide, when expressed in vivo in a canine is able to be cellularly processed to mature canine GHRH with the ability to stimulate growth hormone secretion.
