Abstract

The invention discloses the nanoparticles composed of chitosan/poly-γ-glutamic acid characterized with a positive surface charge and their enhanced permeability through Caco-2 cells for paracellular drug delivery.

Claims (19)

1. A method of enhancing intestinal paracellular transport of at least one bioactive agent in a patient comprising orally administering nanoparticles composed of said at least one bioactive agent, γ-PGA and low molecular weight chitosan.

2. The method of claim 1 , wherein the low molecular weight chitosan has a molecular weight of about 50 kDa.

3. The method of claim 1 , wherein the low molecular weight chitosan has a molecular weight of less than about 40 kDa.

4. The method of claim 1 , wherein said chitosan dominates on a surface of said nanoparticle.

5. The method of claim 1 , wherein a surface of said nanoparticles is characterized with a positive surface charge.

6. The method of claim 1 , wherein the nanoparticles have a mean particle size between about 50 and 400 nanometers.

7. The method of claim 1 , wherein the nanoparticles have a mean particle size between about 100 and 300 nanometers.

8. The method of claim 1 , wherein the nanoparticles have a mean particle size between about 100 and 200 nanometers.

9. The method of claim 1 , wherein the nanoparticles are formed via a simple and mild ionic-gelation method.

10. The method of claim 1 , wherein the nanoparticles are loaded with a therapeutically effective amount of said at least one bioactive agent.

11. The method of claim 10 , wherein the bioactive agent is a protein.

12. The method of claim 10 , wherein the bioactive agent is calcitonin.

13. The method of claim 10 , wherein the bioactive agent is an Alzheimer antagonist.

14. The method of claim 1 , wherein means for enhancing said paracellular transport is to increase an absorption rate of the at least one bioactive agent.

15. The method of claim 10 , wherein the bioactive agent is a peptide.

16. The method of claim 10 , wherein the bioactive agent is cyclosporin.

17. The method of claim 10 , wherein the bioactive agent is insulin.

18. The method of claim 10 , wherein the bioactive agent is a tumor necrosis factor.

19. The method of claim 10 , wherein the bioactive agent is antibiotic.
