Abstract

Poly(ethylene glycol) carbamate derivatives useful as water-soluble prodrugs are disclosed. These degradable poly(ethylene glycol) carbamate derivatives also have potential applications in controlled hydrolytic degradation of hydrogels. In such degradable hydrogels, drugs may be trapped in the gel and released by diffusion as the gel degrades, or they may be covalently bound through hydrolyzable carbamate linkages. Hydrolysis of these carbamate linkages releases the drug at a controllable rate as the gel degrades.

Claims (41)

1. A method of preparing a prodrug conjugate, said method comprising:

a) providing a polymer having the structure:

where

POLY is a water soluble, non-peptidic polymer;

L is a hydrolytically stable linking group;

Ar is an aromatic group; and

X is an activating group capable of reacting with an amino group of a biologically active agent to form a carbamate linkage, and

b) reacting said polymer with a biologically active agent, H 2 N—Y, wherein H 2 N—Y is a biologically active agent comprising at least one amino group, under conditions effective to displace X from said polymer to thereby form a prodrug having the structure:

2. The method of claim 1 , wherein said POLY is selected from the group consisting of poly(alkylene oxides), poly(oxyethylated polyols), poly(vinyl alcohols), dextran, carbohydrate-based polymers, poly(oxazoline), difunctional poly(acryloylmorpholine), and poly(vinylpyrrolidone).

3. The method of claim 2 , wherein said POLY is selected from the group consisting of homopolymers, random and block copolymers, and random and block terpolymers.

4. The method of claim 2 , wherein said POLY is straight chain or branched.

5. The method of claim 2 , wherein said POLY is selected from the group consisting of poly(ethylene glycol), poly(propylene glycol), copolymers of ethylene glycol and propylene glycol, poly(oxyethylated glycerol), poly(oxyethylated sorbitol, and poly(oxyethylated glucose).

6. The method of claim 5 , wherein said POLY is polyethylene glycol.

7. The method of claim 1 , wherein the molecular weight of POLY ranges from about 100 to about 100,000.

8. The method of claim 7 , wherein the molecular weight of POLY ranges from about 2,000 to about 80,000.

9. The method of claim 1 , wherein POLY further comprises a terminal capping group selected from the group consisting of HO—, alkoxy, and

where

L′ is a hydrolytically stable linking group;

Ar′ is an aromatic group; and

X′ is an activating group capable of reacting with an amino group of a biologically active agent to form a carbamate linkage,

and L′, Ar′, and Y′ may be the same or different from L, Ar, and Y, respectively.

10. The method of claim 9 , wherein said terminal capping group is a C 1-4 alkoxy.

11. The method of claim 10 , wherein said capping group is methoxy.

12. The method of claim 6 , wherein said polyethylene glycol has the formula: —(CH 2 CH 2 O) n —CH 2 CH 2 —, wherein n ranges from about 8 to about 4000.

13. The method of claim 6 , wherein said poly(ethylene glycol) is straight chain or branched.

14. The method of claim 13 , wherein said poly(ethylene glycol) is a branched poly(ethylene glycol) having a central core based upon pentaerythritol or glycerol, and exending therefrom, a number of arms ranging from 3 to 100.

15. The method of claim 13 , wherein said poly(ethylene glycol) is branched and is selected from the group consisting of pendant and forked poly(ethylene glycol).

16. The method of claim 13 , wherein said poly(ethylene glycol) is a branched poly(ethylene glycol) comprising a central core based upon lysine or glycerol, and extending therefrom, 2 or 3 methoxy poly(ethylene glycol) arms.

17. The method of claim 1 , wherein L is selected from the group consisting of ether, amine, imine, imide, amide, carbamide, ester, thioester, carbonate, and urea.

18. The method of claim 1 , wherein L is —O— or —NHCO—.

19. The method of claim 1 , wherein Ar is selected from the group consisting of phenyl, substituted phenyl, biphenyl, substituted biphenyl, polycyclic aryl, substituted polycyclic aryl, heterocyclic aryl, and substituted heterocylic aryl.

20. The method of claim 19 , wherein Ar is phenyl that is ortho-, meta- or para-substituted.

21. The method of claim 20 , wherein Ar is

22. The method of claim 1 , wherein X is selected from the group consisting of halogen, N-succinimidyloxy, 1 -benzotriazolyloxy, and p-nitrophenyloxy.

23. The method of claim 22 , wherein X is chlorine or bromine.

24. The method of claim 1 , wherein H 2 N—Y is an amino-bearing biologically active agent selected from the group consisting of proteins, enzymes, peptides, aminolipids, polysaccharides, amino-oligonucleotides, organic compounds, inorganic compounds.

25. The method of claim 1 , wherein said biologically active agent is water-insoluble or immunogenic.

26. The method of claim 1 , further comprising the step of:

c) administering to a mammal the prodrug formed in step (b).

27. The method of claim 26 , wherein said administering is to the blood circulation.

28. The method of claim 26 , wherein subsequent to said administering, POLY-L-Ar—O—C(O)—NH—Y is hydrolyzed to thereby release the parent biologically active agent, H 2 N—Y.
