Formation of polyampholytes in the presence of a polyion
View Patent ↗Polyampholyte are able to condense nucleic acid to form small complexes which can be utilized in the delivery of nucleic acid to mammalian cells. The polyampholytes can be formed prior to interaction with nucleic acid or they can be formed in the presence of nucleic acid. Stabilized polycation/nucleic acid complexes can be modified to reduce the positive charge of the polycation and add targeting ligands without destabilizing the complex. The resultant particles retain their small size and are more effective in delivery of nucleic acid to cells in vivo.
1. A process for delivering a nucleic acid to a cell, comprising:
a) condensing the nucleic acid with a polycation to form a complex with a positive surface charge;
b) crosslinking the polycation;
c) modifying the polycation to reduce the surface charge of the complex; and,
d) contacting the cell with the reduced surface charge complex.
2. The process of claim 1 further comprising modifying the polycation to add a targeting ligand.
3. The process of claim 1 wherein modifying the polycation consists of chemically modifying positively charged moieties on the polycation.
4. The process of claim 3 wherein chemically modifying the polycation consists of adding a cation-reactive reagent to the complex.
5. The process of claim 4 wherein the cation-reactive reagent forms a labile bond with the polycation.
6. The process of claim 4 wherein the cation-reactive reagent is charge neutral.
7. The process of claim 4 wherein the cation-reactive reagent is negatively charged.
8. The process of claim 4 wherein the cation-reactive reagent contains a labile bond.
9. The process of claim 4 wherein the cation-reactive reagent contains a polyethylene glycol group.
10. The process of claim 1 wherein crosslinking the polycation comprises adding a multivalent crosslinker to the complex.
11. The process of claim 10 wherein the crosslinker consists of a labile crosslinker.
12. The process of claim 11 wherein the crosslinker consists of a cleavable crosslinker.
13. A complex for delivering a nucleic acid to a cell formed by the process comprising:
a) condensing the nucleic acid with a polycation to form a complex with a positive surface charge;
b) crosslinking the polycation; and,
c) adding a cation-reactive reagent to modify the polycation thereby reducing the surface charge of the complex.
14. The complex of claim 13 wherein a targeting ligand is attached to the polycation.
15. The complex of claim 13 wherein the complex is crosslinked with a labile crosslinker.
16. The complex of claim 13 wherein the cation-reactive reagent forms a labile linkeage to the polycation.
17. The complex of claim 13 wherein the cation-reactive reagent has a net negative charge.
18. The complex of claim 13 wherein the cation-reactive reagent is charge neutral.
19. The complex of claim 13 wherein the cation-reactive reagent attached a polyethyleneglycol group to the complex.
