View Patent ↗The invention encompasses ureido substituted benzoic acid compounds, compositions comprising the compounds and methods for treating or preventing diseases associated with nonsense mutations of mRNA by administering these compounds or compositions.
1. A method of modulating premature translation termination and/or nonsense-mediated mRNA decay in a cell in vitro, comprising contacting a cell exhibiting premature translation termination and/or nonsense-mediated mRNA decay with an effective amount of a compound having the structure:
or a pharmaceutically acceptable salt, hydrate, clathrate or stereoisomer thereof, wherein:
X is C(═O), C(═S), S, S(═O) or S(O) 2 ;
Y is substituted or unsubstituted alkyl, substituted or unsubstituted aryl, or substituted or unsubstituted cycloalkyl;
R is hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted arylalkyl, or substituted or unsubstituted cycloalkylalkyl;
n is an integer ranging from 0-4;
R 1 and R 2 together form:
and
R 3 -R 6 are each independently hydrogen, halogen, substituted or unsubstituted alkyl, substituted or unsubstituted alkenyl, substituted or unsubstituted alkynyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted arylalkyl, substituted or unsubstituted cycloalkylalkyl, or alkoxy.
2. A method for treating cystic fibrosis or Duchenne muscular dystrophy, comprising administering to a patient an effective amount of a compound having the structure:
or a pharmaceutically acceptable salt, hydrate, clathrate or stereoisomer thereof, wherein:
X is C(═O), C(═S), S, S(═O) or S(O) 2 ;
Y is substituted or unsubstituted alkyl, substituted or unsubstituted aryl, or substituted or unsubstituted cycloalkyl;
R is hydrogen, substituted or unsubstituted alkyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted arylalkyl, or substituted or unsubstituted cycloalkylalkyl,
n is an integer ranging from 0-4;
R 1 and R 2 together form:
and R 3 -R 6 are each independently hydrogen, halogen, substituted or unsubstituted alkyl, substituted or unsubstituted alkenyl, substituted or unsubstituted alkynyl, substituted or unsubstituted cycloalkyl, substituted or unsubstituted aryl, substituted or unsubstituted arylalkyl, substituted or unsubstituted cycloalkylalkyl, or alkoxy.
