Abstract

A porous substrate capable of adsorptive filtration of a fluid having a porous self-supporting substrate and one or more porous, adsorptive coatings comprising from about 1 to about 80% of the void volume of the pores of the substrate. The resultant substrate has good convective and diffusive flow and capacity. The substrate may be crosslinked, have one or more capture chemistries attached to it and is useful as a chromatography media for the selective filtration of desired species including biomolecules such as proteins and DNA fragments.

Claims (22)

1. A porous coated media for adsorption or chromatography based separations comprising a base substrate, the base substrate being a porous non-woven fabrics, self-supporting structure, and one or more porous coatings being formed of agarose and having a thickness of from about 1 to about 20 microns on at least a portion of all surfaces of the substrate, the one or more porous coatings being porous to biomolecules by adsorption in its bulk.

2. The substrate of claim 1 wherein the substrate is in a form selected from the group consisting of a sheet, a fiber and a monolith.

3. The substrate of claim 1 wherein the one or more coatings are formed on substantially all the surfaces of the substrate.

4. The substrate of claim 1 wherein the one or more coatings occupying from about 10 to about 50% of the void volume of the pores of the substrate and the substrate has good convective flow and diffusional flow.

5. The substrate of claim 1 wherein the substrate is a plastic non-woven.

6. The substrate of claim 1 wherein the substrate is a non-woven fabric having a pore size of from about 20 to about 200 microns, and wherein the coating has a thickness of from about 2 to about 20 microns, and an average pore size of from about 1 to about 200 nanometers.

7. The substrate of claim 1 wherein the substrate has a pore size of from about 50 to about 300 microns.

8. The substrate of claim 1 wherein the substrate has a pore size of from about 50 to about 200 microns.

9. The substrate of claim 1 , wherein the coated substrate comprises a fractional porosity of from about 0.35 to about 0.55.

10. The substrate of claim 1 wherein the agarose coating has a thickness of from about 1 to about 20 microns.

11. The substrate of claim 1 wherein the agarose coating has a thickness of from about 5 to about 15 microns.

A porous coated substrate for adsorption based separations comprising a base substrate, the base substrate being a porous, self-supporting structure formed of non-woven fabrics having a pore size of from about 30 to about 200 microns, one or more agarose porous coatings having a thickness of from about 1 to about 20 microns on all surfaces of the substrate and the one or more coatings occupying from about 1 to about 50% of the void volume of the pores of the substrate and the substrate having good convective flow and diffusional flow.

13. The substrate of claim 12 wherein the agarose coating has a thickness of from about 1 to about 20 microns.

14. The substrate of claim 12 wherein the agarose coating has a thickness of from about 5 to about 15 microns.

A porous coated substrate for adsorption based separations comprising a base substrate, the base substrate being a porous, self-supporting structure formed of non-woven fabrics of a material selected from the group consisting of plastic, metals and glasses and one or more porous agarose coatings on all surfaces of the substrate, the one or more agarose coatings having an average pore size from about 1 to about 200 nanometers and a thickness of from about 1 to about 20 microns occupying from about 10 to about 50% of the void volume of the pores of the substrate and the substrate having good convective flow and diffusional flow.

16. The substrate of claim 15 wherein the agarose coating has a thickness of from about 1 to about 20 microns.

A porous coated substrate for adsorption based separations comprising a base substrate, the base substrate being a porous, self-supporting structure formed of plastic non-woven fabrics, and one or more porous coatings on substantially all surfaces of the substrate, the one or more coatings having a thickness of from about 1 to about 20 microns occupying from about 10 to about 50% of the void volume of the pores of the substrate, the one or more coatings being formed of one or more agaroses and the substrate having good convective flow and diffusional flow.

18. The substrate of claim 17 wherein the agarose coating has a thickness of from about 1 to about 20 microns.

A porous coated substrate for adsorption based separations comprising a base substrate, the base substrate being a porous, self-supporting structure formed of a non-woven fabric-having a pore size of from about 30 to about 200 microns and one or more porous agarose coatings being formed of agarose having a thickness of from about 1 to about 20 microns on all surfaces of the substrate and having an average pore size of from about 1 to about 100 nanometers, the one or more coatings occupying from about 1 to about 50% of the void volume of the pores of the substrate and the capacity of the substrate is independent of the convective flow properties of the substrate.

20. The substrate of claim 19 wherein the agarose coating has a thickness of from about 1 to about 15 microns.

A porous coated substrate for adsorption based separations comprising a base substrate, the base substrate being a porous self supporting structure being made from non-woven fabrics having a pore size of from about 30 to about 300 microns, and one or more porous coatings being formed of agarose and having a thickness of from about 2 to about 20 microns on substantially all surfaces of the substrate, wherein the coated substrate comprises a fractional porosity of from about 0.35 to about 0.55.

22. The substrate of claim 21 wherein the agarose coating has a thickness of from about 2 to about 15 microns.
