Abstract

An auto-injection system provides high throughput screening of fluidic samples. A sample injection valve has a first position which applies a reduced pressure to a sample sipper tube for aspirating a fluidic sample into the sample sipper tube, and a second position which delivers the fluidic sample to a sample supply loop. A column control valve has a first position which delivers the fluidic sample from the sample supply loop to a sample chromatography column, and a second position which reverses direction of fluid flow through the sample chromatography column to deliver the fluidic sample to a sample analyzer. A wash control valve has a first position which supplies a wash buffer solution to the sample chromatography column in a forward fluid flow direction, and a second position which supplies elution solvent to flush the sample supply loop.

Claims (53)

1. An auto-injection system for high throughput screening of fluidic samples, the system comprising:

a sample injection valve having:

i. a first position which applies a reduced pressure to a sample sipper tube for aspirating a fluidic sample into the sample sipper tube, and

ii. a second position which delivers the fluidic sample to a sample supply loop;

a column control valve in fluid communication with a sample analyzer, a source of elution solvent, and a sample chromatography column, and configured to facilitate a continuous flow of an elution solvent to a sample analyzer, the column control valve having:

i a first position which simultaneously delivers the fluidic sample from the sample supply loop to the sample chromatography column in a first direction and delivers an elution solvent to the sample analyzer, and

ii. a second position which flows the elution solvent through the sample chromatography column in a second direction to deliver the fluidic sample and the elution solvent to the a sample analyzer;

a wash control valve having:

i a first position which supplies a wash buffer solution to the sample chromatography column in a forward fluid flow direction, and

ii. a second position which supplies elution solvent to flush the sample supply loop;

means for presenting individual microplate sample wells to the sample sipper tube; and

automated control means for creating a cycle of repeatedly introducing samples and actuating the sample injection valve, column control valve, and wash control valve.

2. An auto-injection system according to claim 1 , wherein the first position of the sample injection valve further delivers wash buffer solution from the wash control valve to the sample chromatography column.

3. An auto-injection system according to claim 1 , wherein the second position of the sample injection valve further delivers elution solvent from the wash control valve to the sample supply loop.

4. An auto-injection system according to claim 1 , wherein the first position of the column control valve further supplies elution solvent to flush the sample analyzer.

5. An auto-injection system according to claim 1 , wherein the second position of the column control valve further allows elution solvent to exit from the sample supply loop.

6. An auto-injection system according to claim 1 , wherein the second position of the wash control valve further delivers wash buffer solution to flush the sample supply loop.

7. An auto-injection system according to claim 1 , wherein the sample analyzer is a mass spectrometer.

8. An auto-injection system according to claim 1 , further comprising: a wash buffer supply pump which provides wash buffer solution to the wash control valve.

9. An auto-injection system according to claim 1 , further comprising:

a first elution solvent pump which provides elution solvent to the column control valve.

10. An auto-injection system according to claim 9 , further comprising:

a second elution solvent pump which provides elution solvent to the wash control valve.

11. An auto-injection system according to claim 1 , wherein samples are processed at a rate of at least twice per minute.

12. An auto-injection system according to claim 1 wherein the sipper tube is flushed from a wash solvent reservoir at a time during the cycle that overlaps selection of the wash control valve to supply elution solvent to the sample supply loop.

13. An auto-injection system for high throughput screening of fluidic samples, the system comprising:

a sample injection valve coupled to a sipper, a vacuum, and a sample loop; a column control valve coupled to the sample injection valve, a first elution solvent pump, a chromatography column, and a sample analyzer;

a wash control valve coupled to the sample injection valve, the column control valve, a wash solvent pump, and a second elution solvent pump;

wherein the sample injection valve is configured for actuation to:

a first position, wherein the sample loop is in communication with the vacuum and the sipper; and

a wash control valve and the column control valve;

wherein the column control valve is configured to facilitate a continuous flow of an elution solvent to the sample analyzer by actuation to:

a first position, wherein the chromatography column is in communication with the sample injection valve to deliver the sample loop's contents to the chromatography column in a first direction and wherein the analyzer is in communication with the first elution solvent pump; and

a second position, wherein the sample analyzer is in communication with the first elution solvent pump via the chromatography column to flow the elution solvent through the chromatography column in a second direction and present the elution solvent and an eluted sample to the sample analyzer; and

wherein the wash control valve is configured for actuation to:

a first position, wherein the wash solvent pump is in communication with the sample injection valve; and

a second position, wherein the second elution pump is in communication with the sample injection valve.

14. A method for high throughput screening of fluidic samples, the method comprising:

providing a sample injection valve having:

i. a first position which applies a reduced pressure to a sample sipper tube for aspirating a fluidic sample into the sample sipper tube, and

ii. a second position which delivers the fluidic sample to a sample supply loop; providing a column control valve in fluid communication with a sample analyzer, a source of elution solvent, and a sample chromatography column, and configured to facilitate a continuous flow of an elution solvent to the sample analyzer, the column control valve having:

i. a first position which simultaneously delivers the fluidic sample from the sample supply loop to the sample chromatography column in a first direction and delivers an elution solvent to the sample analyzer, and

ii. a second position which flows the elution solvent through the sample chromatography column in a second direction to deliver the fluidic sample and the elution solvent to the sample analyzer; providing a wash control valve having:

i. a first position which supplies a wash buffer solution to the sample chromatography column in a forward fluid flow direction, and

ii. a second position which supplies elution solvent to flush the sample supply loop;

providing means for presenting individual microplate sample wells to the sample sipper tube; and

actuating automated control means for creating a cycle of repeatedly introducing samples and actuating the sample injection valve, column control valve, and wash control valve.

15. The method of claim 14 , wherein the cycle of repeatedly introducing samples and actuating the sample injection valve, column control valve, and wash control valve comprises:

actuating the sample injection valve to the first position while actuating the wash control valve to the first position and actuating the column control valve to the first position;

actuating the sample injection valve to the second position while maintaining the wash control valve in the second position and maintaining the column control valve to the second position; and

actuating the wash control valve to a second position and actuating column control valve to a second position, while maintaining the sample injection valve in the second position.

16. A method according to claim 14 , in which the cycle is performed at a speed of greater than two samples per minute.

17. A method according to claim 14 , wherein the sample analyzer is a mass spectrometer.
