Abstract

T-cells are generated with enhanced immunostimulatory capabilities for use in self therapy treatment protocols, by utilizing a biodegradable device with a biodegradable support that has one or more agents that are reactive to T-cell surface moieties. The biodegradable devices are mixed with the T-cells sufficiently so that the one or more agents cross-link with the T-cells' surface moieties and deliver a signal to the T-cells to enhance immunostimulatory capabilities.

Claims (18)

1. A method for generating T-cells with enhanced immunostimulatory capabilities for use in cell therapy treatment protocols, the method comprising:

providing an universal crosslinking agent comprising a device with a support coated with a first material wherein the first material is capable of cross-linking more than one second material, the second materials capable of binding moieties on the surface of T-cells;

mixing the universal crosslinking agent with a first array comprising at least one of the second materials and the T-cells sufficiently so that the first and second material cross-link and the second material binds with the T-cell surface moieties and delivers a signal to the T-cells; and

adding a second array comprising one or more additional second materials to the universal crosslinking agent and the T-cells at a later time in the T-cell response, wherein at least one of the second materials added in the second array is different than at least one of the second materials added in the first array of the mixing step.

2. The method of claim 1 wherein the second material is hound to moieties on the surface of the T-cells prior to mixing with the universal crosslinking agent.

3. The method of claim 1 wherein the second material is an antibody that has specificity to T-cell surface moieties.

4. The method of claim 1 wherein the first material is an antibody.

5. The method of claim 1 wherein the device is a biodegradable microsphere.

6. The method of claim 5 wherein the microsphere degrades into a substance that is nontoxic to humans.

7. The method of claim 1 wherein the universal crosslinking agent is crosslinked with the second material prior to mixing with the T-cells.

8. The method of claim 1 and further including:

centrifuging the universal crosslinking agents with the T-cells to enhance cross-linking.

9. The method of claim 1 and further including:

culturing the universal crosslinking agent with the T-cells to a high cell density.

10. The method of claim 5 wherein the biodegradable micro spheres after being mixed with the T-cells degrade in culture media or physiological fluids in less than 14 days.

11. The method of claim 1 wherein the device is a biodegradable device and the support is a biodegradable support.

12. The method of claim 1 further comprising adding additional universal crosslinking agent in the adding step.

13. The method of claim 1 further comprising adding a third array comprising one or more additional second materials to the universal crosslinking agent and the T-cells at a later time in the T-cell response, wherein at least one of the second materials added in the third array is different than at least one of the second materials added in the second array.
