Abstract

Antigenic isolates and vaccines for Infectious Bursal Disease Virus include variants of the molecular Group 6 family of IBDV isolates, in particular the 28-1 isolate.

Claims (26)

1. A vaccine composition comprising an Infectious Bursal Disease Virus (IBDV) antigen comprising the amino acid sequence of SEQ ID NO: 1 further comprising two or more amino acid substitutions,

wherein a first amino acid substitution of G to D at amino acid position 318 is the only amino acid substitution within the major hydrophilic peak B of IBDV Viral Protein-2 (VP-2); and

wherein a second amino acid substitution is selected from the group consisting of 222-T, 249-K, 254-S, 279-N and 286-I.

2. The vaccine composition of claim 1 , further comprising a pharmacologically acceptable carrier.

3. The vaccine composition of claim 2 , further comprising at least one additional vaccine antigen.

4. The vaccine composition of claim 2 , wherein said carrier is selected from the group consisting of aqueous carriers and emulsions.

5. The vaccine composition of claim 4 , wherein said carrier is a water-in-oil emulsion.

The vaccine composition of claim 3 , wherein said at least one additional vaccine antigen is selected from the group consisting of chicken herpes virus antigens, chicken anemia virus (CAV) antigens, Newcastle Disease virus antigens, Infectious Brochitis (IB) virus antigens, and Infectious Bursal Disease Virus (IBDV) variant antigens.

7. The vaccine composition of claim 1 , wherein said antigen is a live, live-attenuated or killed-inactivated antigen.

8. The vaccine composition of claim 7 , wherein said antigen is live-attenuated and has been serially passaged.

9. The vaccine composition of claim 1 , wherein the antigen comprises the 28-1 antigenic isolate of IBDV, which is deposited with the ATCC under Accession Number PTA-5848.

10. The vaccine composition of claim 6 , wherein said at least one additional vaccine antigen is a reovirus antigen.

11. A vaccine composition comprising an Infectious Bursal Disease Virus (IBDV) antigenic isolate comprising an IBDV antigen comprising the amino acid sequence of SEQ ID NO: 1, wherein said IBDV antigen further comprises two or more amino acid substitutions;

wherein a first amino acid substitution of G to D at amino acid position 318 is the only amino acid substitution within the major hydrophilic peak B of IBDV Viral Protein-2 (VP-2); and

wherein a second amino acid substitution is selected from the group consisting of 222-T, 249-K, 254-S, 279-N, and 286-I.

12. A method for inducing protection against infection from Infectious Bursal Disease Virus, which comprises administering to a poultry animal the vaccine composition according to claim 1 .

13. A method for inducing protection against Infectious Bursal Disease, which comprises administering to a poultry animal the vaccine composition according to claim 11 .

14. The method of claim 12 , wherein said vaccine is administered in ovo or to young post-hatch chicks.

15. The method of claim 12 , wherein said IBDV antigenic isolate is administered with at least one other antigen for poultry.

16. The method of claim 15 , wherein said other antigen is reovirus.

17. A vaccine composition comprising an Infectious Bursal Disease Virus (IBDV) Viral Protein 2 (VP-2) antigen comprising the amino acid sequence of SEQ ID NO:1 and further comprising a 318 G→D amino acid substitution, wherein said substitution is the only amino acid substitution in the hydrophilic peak region B of the VP-2 of Infectious Bursal Disease Virus.

An Infectious Bursal Disease Virus (IBDV) Viral Protein 2 (VP-2) antigen comprising the amino acid sequence of SEQ ID NO:1 and further comprising a 318 G→D amino acid substitution, wherein said substitution is the only amino acid substitution in the hydrophilic peak region B of the VP-2 amino acid sequence.

19. The vaccine composition af claim 11 , wherein said Infectious Bursal Disease Virus antigenic isolate comprises the amino acid sequence set forth in SEQ ID NO: 6.

20. The vaccine composition of claim 6 , wherein said at least one additional vaccine antigen is a Lukert IBDV strain antigen.

21. The vaccine composition of claim 1 , wherein the antigen comprises the amino acid sequence set forth in the SEQ ID NO: 6.

22. The vaccine composition of claim 11 , wherein the Infectious Bursal Disease Virus antigen is isolated from ATCC Accession Number PTA-5848.
