Abstract

A method of enhancing the permeability of a biological membrane, including the skin or mucosa of an animal or the outer layer of a plant to a permeant is described utilizing microporation of selected depth and optionally one or more of sonic, electromagnetic, mechanical and thermal energy and a chemical enhancer. Microporation is accomplished to form a micropore of selected depth in the biological membrane and the porated site is contacted with the permeant. Additional permeation enhancement measures may be applied to the site to enhance both the flux rate of the permeant into the organism through the micropores as well as into targeted tissues within the organism.

Claims (41)

1. A device for ablating stratum corneum of skin on the body of a subject, comprising:

a plurality of heat probes, which are each formed from a thin layer and adapted to be applied to the skin of the subject at respective points; and

a power source, which is adapted (i) to deliver sufficient thermal energy to at least one of the plurality of the heat probes such that the at least one of the plurality of the heat probes is cycled through a series of one or more modulations of temperature from an initial point of ambient temperature to a peak temperature in excess of 123° C., and back to ambient surface temperature, and (ii) to apply electrical energy to at least one of the plurality of heat probes so as to induce an electrical current to flow through the stratum corneum, causing ablation of narrow channels through the stratum corneum at the respective points.

2. The device according to claim 1 , wherein the power source is adapted to apply the electrical energy such that skin layers beneath the stratum corneum are substantially not ablated.

3. The device according to claim 1 , wherein the power source is adapted to apply the electrical energy such that the ablation of the narrow channels causes a puncturing of the stratum corneum.

4. The device according to claim 1 , wherein the device is adapted to ablate the narrow channels in order to allow a substance to pass therethrough.

5. The device according to claim 4 , wherein the device is adapted to ablate the narrow channels so as to allow the substance to pass through the skin into the body of the subject.

6. The device according to claim 5 , and comprising a housing having a surface that defines a storage region adapted to store the substance in preparation for passage of the substance through the stratum corneum.

7. The device according to claim 4 , wherein the device is adapted to ablate the narrow channels so as to allow the substance to pass through the stratum corneum and into the body of the subject.

8. The device according to claim 4 , wherein the power source is adapted to apply the electrical energy during a first time period, so as to ablate the narrow channels in order to facilitate passage of the substance during a second time period subsequent to the first time period.

9. The device according to claim 1 , wherein the power source is further adapted to

generate an electric field which causes the electrical current to flow through the stratum corneum, and

reduce power dissipated in the stratum corneum responsive to a variation of a characteristic of the current.

10. The device according to claim 9 , wherein the characteristic of the current is a magnitude of the current.

11. The device according to claim 1 , wherein a distance between two of the plurality of heat probes is determined such that contact between the skin is confined to a small area less than 1000 μm.

12. The device according to claim 11 , wherein the distance between the two heat probes is between about 1 to 1000 μm.

13. The device according to claim 1 , wherein the device is configured to monitor dynamic impedance of the heat probes for closed loop control of a temperature of a point of contact between the skin and at least one of the plurality of heat probes.

14. The device according to claim 1 , wherein the power source is adapted to generate alternating current, a frequency thereof being above about 100 Hz.

15. The device according to claim 14 , wherein the power source is adapted to generate the alternating current such that the frequency is between about 1 kHz and about 300 kHz.

16. The device according to claim 1 , wherein the power source is adapted to generate alternating current and to modulate a frequency thereof between a first frequency value and a second frequency value.

17. The device according to claim 1 , wherein the power source is adapted to apply the electrical energy for less than about 1 second.

18. The device according to claim 17 , wherein the power source is adapted to apply the electrical energy for less than about 200 milliseconds.

19. The device according to claim 1 , wherein the heat probes comprise at least one set of two or more heat probes, and wherein the power source is adapted to apply the electrical energy so as to induce the current to flow between the heat probes in the at least one set.

20. The device according to claim 1 , wherein the plurality of heat probes are each formed from a thin layer having a thickness of about 5 to 1000 μm.

21. A device for ablating stratum corneum of skin on the body of a subject, comprising:

a housing having a surface that defines a storage region adapted to store an active substance; a plurality of heat probes, which are each formed from a thin layer and adapted to be applied to the skin of the subject at respective points; and

a power source, which is adapted (i) to deliver sufficient thermal energy to at least one of the plurality of the heat probes such that the at least one of the plurality of the heat probes is cycled through a series of one or more modulations of temperature from an initial point of ambient temperature to a peak temperature in excess of 123° C., and back to ambient surface temperature, and (ii) to apply electrical energy between two or more of the plurality of heat probes, to cause ablation of an area of the stratum corneum so as to facilitate passage of the active substance through the ablated area.

22. The device according to claim 21 , wherein a distance between two of the plurality of heat probes is determined such that contact between the skin is confined to a small area less than 1000 μm.

23. The device according to claim 22 , wherein the distance between the two heat probes is between about 1 μm to 1000 μm.

24. The device according to claim 21 , wherein the device is configured to monitor dynamic impedance of the heat probes for closed loop control of a temperature of a point of contact between the skin and at least one of the plurality of heat probes.

25. The device according to claim 21 , wherein the plurality of electrodes comprise at least one set of two or more heat probes, and wherein the power source is adapted to apply the electrical energy so as to induce the current to flow between the heat probes in the at least one set.

26. The device according to claim 21 , wherein the power source is adapted to generate alternating current, a frequency thereof being above about 100 Hz.

27. The device according to claim 26 , wherein the power source is adapted to generate the alternating current such that the frequency is between about 1 kHz and about 300 kHz.

28. The device according to claim 26 , wherein ablation of an area of the stratum corneum occurs during a first time period, and wherein the first time period is less than about 1 second.

29. The device according to claim 28 , wherein the first time period is less than about 200 milliseconds.

30. The device according to claim 21 , wherein the active substance is a polypeptide.

31. The device according to claim 21 , wherein the plurality of heat probes are each formed from a thin layer having a thickness of about 5 to 1000 μm.

32. A device for ablating stratum corneum of skin of a subject, comprising: a plurality of heat probes, which are each formed from a thin layer and adapted to be applied to the skin of the subject at respective points on the skin confined to a small area less than 1000 μm; a housing having a surface that defines a storage region adapted to store an active substance; and a power source adapted (i) to deliver sufficient thermal energy to at least one of the plurality of the heat probes such that the at least one of the plurality of the heat probes is cycled through a series of one or more modulations of temperature from an initial point of ambient temperature to a peak temperature in excess of 123° C., and back to ambient surface temperature, and (ii) to apply electrical energy between two of the plurality of heat probes applied to the skin to cause ablation of the stratum corneum, so as to facilitate passage of the active substance through the skin.

33. The device according to claim 32 , wherein a distance between the two electrodes is between about 1 to 1000 μm.

34. The device according to claim 32 , wherein the power source is adapted to generate alternating current, a frequency thereof being above about 100 Hz.

35. The device according to claim 34 , wherein the power source is adapted to generate the alternating current such that the frequency is between about 1 kHz and about 300 kHz.
