Abstract

Isolated nucleic acids encoding allergens of the species Dermatophagoides pteronyssinus and Dermatophagoides farinae , Der p VII and Der f VII, respectively, are disclosed. A cDNA encoding a peptide having a Der p VII activity and a predicted molecular weight of about 22, 177 daltons is described. A cDNA encoding a peptide having Der f VII activity is also described. The nucleic acids of the invention can be used as probes to detect the presence of Der p VII or Der f VII nucleic acid in a sample or for the recombinant production of peptides having a Der p VII or Der f VII activity. Peptides having a Der p VII or Der f VII activity can be used in compositions suitable for pharmaceutical administration or methods of diagnosing sensitivity to house dust mite allergens.

Claims (8)

1. An isolated nucleic acid molecule encoding a peptide of a Dermatophagoides farinae VII (Der f VII) protein, wherein the protein comprises the amino acid sequence shown in SEQ ID NO:7, and wherein the peptide comprises a portion of said protein, but not the full-length protein, and wherein the portion is at least 30, 40, 60, 80, 100, 140 or 200 residues in length.

An isolated nucleic acid molecule comprising a nucleotide sequence that is at least 95% homologous to a portion of the nucleotide sequence shown in SEQ ID NO:6, or a complement thereof, wherein the portion encodes a peptide sequence at least 30, 40, 60, 80, 100, 140 or 200 residues in length, of SEQ ID NO:7, and wherein the peptide encoded by the nucleic acid molecule is not the full-length Der f VII protein amino acid sequence shown in SEQ ID NO:7.

An isolated nucleic acid molecule encoding a Der f VII peptide comprising an amino acid sequence that is at least 95% homologous to the amino acid sequence shown in SEQ ID NO:7, wherein the nucleic acid molecule does not encode the full-length Der f VII protein amino acid sequence shown in SEQ ID NO:7.

An isolated nucleic acid molecule comprising a nucleotide sequence that is at least 95% homologous to a portion of the nucleotide sequence shown in SEQ ID NO: 6, or a complement thereof, wherein the portion encodes a peptide sequence at least 30, 40, 60, 80, 100, 140 or 200 residues in length, of SEQ ID NO:7, and wherein the peptide encoded by the nucleic acid molecule is not full-length Der f VII protein allergen.

5. An isolated nucleic acid molecule encoding a Der f VII protein comprising an amino acid sequence that is at least 95% homologous to the amino acid sequence shown in SEQ ID NO:7, wherein the nucleic acid molecule does not encode full-length Der f VII protein allergen.

6. An isolated nucleic acid molecule encoding a Der f VII peptide comprising an amino acid sequence that is a naturally occurring polymorphic variant of the protein of SEQ ID NO:7, wherein the nucleic acid molecule does not encode the full-length Der f VII protein amino acid sequence of SEQ ID NO:7.

7. A vector comprising the nucleic acid molecule of any one of claim 1 , 2 - 3 or 6 .

8. A host cell containing the vector of claim 7 .
