Abstract

Efficient sequence specific gene silencing is possible through the use of siRNA technology. By selecting particular siRNAs by rationale design, one can maximize the generation of an effective gene silencing reagent, as well as methods for silencing genes.

Claims (34)

1. A pool comprised of a first siRNA and a second siRNA, wherein said first siRNA consists of a first duplex region and either no overhangs or at least one overhang, wherein each overhang has up to six bases, wherein said first duplex region consists of 19 base pairs that form a first sense region and a first antisense region, wherein said first antisense region is complementary to a first region of a target, and said second siRNA consists of a second duplex region and either no overhangs or at least one overhang, wherein each overhang has up to six bases and said second duplex region consists of 19 base pairs that form a second sense region and a second antisense region, wherein said second antisense region is complementary to a second region of said target, and each of said first sense region and said second sense region satisfy the following criteria: an A base at position 19 and a U base at position 10.

2. The pool of claim 1 , wherein said first siRNA has an A base at position 3 of the sense region.

3. The pool of claim 2 , wherein said second siRNA has an A base at position 3 of the sense region.

4. The pool of claim 1 , wherein said first siRNA has an G base at position 1 of the sense region.

5. The pool of claim 4 , wherein said second siRNA has a G base at position 1 of the sense region.

6. The pool of claim 1 , wherein said first siRNA has an A base at position 14 of the sense region.

7. The pool of claim 6 , wherein said second siRNA has an A base at position 14 of the sense region.

8. The pool of claim 2 , wherein said first siRNA has an A base at position 14 of the sense region.

9. The pool of claim 8 , wherein said second siRNA has an A base at position 14 of the sense region.

10. The pool of claim 4 , wherein said first siRNA has an A base at position 14 of the sense region.

11. The pool of claim 10 , wherein said second siRNA has an A base at position 14 of the sense region.

12. The pool of claim 1 , wherein said first siRNA has a GC content of between about 30 and about 52 percent.

13. The pool of claim 12 , wherein said second siRNA has a GC content of between about 30 and about 52 percent.

14. The pool of claim 1 , wherein said first siRNA has a base other than G at position 13 of the first sense region.

15. The pool of claim 14 , wherein said second siRNA has a base other than G at position 13 of the second sense region.

16. The pool of claim 1 , wherein said first siRNA has an A base at position 7 of the sense region.

17. The pool of claim 16 , wherein said second siRNA has an A base at position 7 of the sense region.

18. The pool of claim 1 , wherein said first siRNA has a base other than A at position 11 of the sense region.

19. The pool of claim 18 , wherein said second siRNA has a base other than A at position 11 of the sense region.

20. The pool of claim 15 , wherein the first siRNA has an A base at position 3 of the first sense region.

21. The pool of claim 20 , wherein the second siRNA has an A base at position 3 of the second sense region.

22. The pool of claim 15 , wherein the first siRNA has a G base at position 1 of the first sense region.

23. The pool of claim 22 , wherein the second siRNA has a G base at position 1 of the second sense region.

24. The pool of claim 20 , wherein the first siRNA has a G base at position 1 of the first sense region.

25. The pool of claim 24 , wherein the second siRNA has a G base at position 1 of the second sense region.

26. The pool of claim 14 , wherein the first siRNA has an A base at position 14 of the first sense region.

27. The pool of claim 26 , wherein the second siRNA has an A base at position 14 of the second sense region.

28. The pool of claim 1 , wherein said pool is within a cell.

29. The pool of claim 12 , wherein said pool is within a cell.

30. The pool of claim 14 , wherein said pool is within a cell.

31. The pool of claim 20 , wherein said pool is within a cell.

32. The pool of claim 22 , wherein said pool is within a cell.

33. The pool of claim 24 , wherein said pool is within a cell.

34. The pool of claim 26 , wherein said pool is within a cell.
