Abstract

The present invention relates to the field of obesity research. Obesity is a public health problem that is serious and widespread. This application pertains to nucleic acids encoding a compound, globular OBG3 polypeptide fragments, vectors and transformed host cells containing these nucleic acids and methods of making the globular OBG3 polypeptide fragments.

Claims (132)

1. An isolated polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids: 87-244 of SEQ ID NO: 6, 88-244 of SEQ ID NO: 6, 89-244 of SEQ ID NO: 6, 90-244 of SEQ ID NO: 6, 91-244 of SEQ ID NO: 6, 92-244 of SEQ ID NO: 6, 93-244 of SEQ ID NO: 6, 94-244 of SEQ ID NO: 6, 95-244 of SEQ ID NO: 6, 96-244 of SEQ ID NO: 6, 97-244 of SEQ ID NO: 6, 98-244 of SEQ ID NO: 6, 99-244 of SEQ ID NO: 6, 100-244 of SEQ ID NO: 6, 111-191 of SEQ ID NO: 6, 144-199 of SEQ ID NO: 6,191-244 of SEQ ID NO: 6, 108 to 244 of SEQ ID NO: 6, amino acids 110 to 244 of SEQ ID NO: 6, amino acids 108 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added, or amino acids 110 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added or a polynucleotide that is complementary to said nucleic acid sequence.

2. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 87-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

3. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 88-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

4. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 89-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

5. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 90-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

6. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 91-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

7. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 92-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

8. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 93-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

9. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 94-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

10. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 95-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

11. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 96-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

12. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 97-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

13. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 98-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

14. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 99-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

15. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 100-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

16. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 111-191 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

17. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 144-199 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

18. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 191-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

19. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 108-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

20. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 108 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added or a polynucleotide complementary thereto.

21. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 110 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added or a polynucleotide complementary thereto.

22. The isolated polynucleotide according to claim 1 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 110 to 244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

A composition comprising a carrier and a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids: 87-244 of SEQ ID NO: 6, 88-244 of SEQ ID NO: 6, 89-244 of SEQ ID NO: 6, 90-244 of SEQ ID NO: 6, 91-244 of SEQ ID NO: 6, 92-244 of SEQ ID NO: 6, 93-244 of SEQ ID NO: 6, 94-244 of SEQ ID NO: 6, 95-244 of SEQ ID NO: 6, 96-244 of SEQ ID NO: 6, 97-244 of SEQ ID NO: 6, 98-244 of SEQ ID NO: 6, 99-244 of SEQ ID NO: 6, 100-244 of SEQ ID NO: 6, 111-191 of SEQ ID NO: 6, 144-199 of SEQ ID NO: 6, 191-244 of SEQ ID NO: 6, 108 to 244 of SEQ ID NO: 6, amino acids 110 to 244 of SEQ ID NO: 6, amino acids 108 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added, or amino acids 110 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added or a polynucleotide that is complementary to said nucleic acid sequence.

24. The composition according to claim 23 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 87-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

25. The composition according to claim 23 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 88-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

26. The composition according to claim 23 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 89-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

27. The composition according to claim 23 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 90-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

28. The composition according to claim 23 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 91-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

29. The composition according to claim 23 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 92-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

42. The composition according to claim 23 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 108 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added or a polynucleotide complementary thereto.

43. The composition according to claim 23 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 110 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added or a polynucleotide complementary thereto.

68. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 87-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

69. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 88-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

70. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 89-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

71. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 90-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

72. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 91-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

73. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 92-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

74. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 93-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

78. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 97-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

79. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 98-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

80. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 99-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

81. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 100-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

82. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 111-191 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

83. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 144-199 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

84. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 191-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

85. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 108-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

86. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 108 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added or a polynucleotide complementary thereto.

87. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 110 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added or a polynucleotide complementary thereto.

88. The composition according to claim 67 , wherein said vector comprises a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids 110 to 244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.

An isolated transformed host cell comprising a polynucleotide consisting of a nucleic acid sequence encoding consecutive amino acids: 87-244 of SEQ ID NO: 6, 88-244 of SEQ ID NO: 6, 89-244 of SEQ ID NO: 6, 90-244 of SEQ ID NO: 6, 91-244 of SEQ ID NO: 6, 92-244 of SEQ ID NO: 6, 93-244 of SEQ ID NO: 6, 94-244 of SEQ ID NO: 6, 95-244 of SEQ ID NO: 6, 96-244 of SEQ ID NO: 6, 97-244 of SEQ ID NO: 6, 98-244 of SEQ ID NO: 6,99-244 of SEQ ID NO: 6, 100-244 of SEQ ID NO: 6, 111-191 of SEQ ID NO: 6, 144-199 of SEQ ID NO: 6, 191-244 of SEQ ID NO: 6, 108 to 244 of SEQ ID NO: 6, amino acids 110 to 244 of SEQ ID NO: 6, amino acids 108 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added, or amino acids 110 to 244 of SEQ ID NO: 6 to which an N-terminal methionine has been added or a polynucleotide that is complimentary to said nucleic acid sequence.

90. The isolated transformed host cell according to claim 89 , wherein said polynucleotide consists of a nucleic acid sequence encoding consecutive amino acids 87-244 of SEQ ID NO: 6 or a polynucleotide complementary thereto.
