Abstract

Gene expression profiling in multiple myeloma patients identifies genes that distinguish between patients with subsequent early death or long survival after treatment. Poor survival is linked to over-expression of genes such as ASPM, OPN3 and CKS1B which are located in chromosome 1q. Given the frequent amplification of 1q in many cancers, it is possible that these genes can be used as powerful prognostic markers and therapeutic targets for multiple myeloma and other cancer.

Claims (11)

1. A method of determining the prognosis of a human multiple myeloma patient, comprising the steps of:

obtaining an isolated nucleic acid sample from plasma cells isolated from the patient;

measuring gene expression levels, by DNA microarray or RT-PCR, using probes specific for GNG10, PNPLA4, KIAA1754, AHCYL1, MCLC, EVI5, AD-020, PARG1, CTBS, FUCA1, RFP2, FLJ20489, LTBP1, TRIP13, AIM2, SELI, SLC19A1, LARS2, OPN3, ASPM, CCT2, UBE21, STK6, FLJ13052, FLJ12525, BIRC5, CKS1B, CKAP1, MGC57827, DKFZp7790175, PFN1, ILF3, IFI16, TBRG4, PAPD1, EIF2C2, MGC4308, ENO1, DSG2, EXOSC4, TAGLN2, RUVBL1, ALDOA, CPSF3, MGC15606, LGALS1, RAD18, SNX5, PSMD4, RAN, KIF14, CBX3, TMPO, DKFZP586L0724, WEE1, ROBO1, TCOF1, YWHAZ, and MPHOSPH1 using the isolated nucleic acid sample, thereby obtaining a gene expression profile; and

comparing the gene expression profile to one or more suitable controls and assigning the patient a prognosis on the basis of the comparison, wherein:

reduced expression of GNG10, PNPLA4, KIAA1754, AHCYL1, MCLC, EVI5, AD-020, PARG1, CTBS, FUCA1, RFP2, FLJ20489, or LTBP1, relative to a control with a favorable prognosis, is associated with a poor prognosis; and

overexpression of TRIP13, AIM2, SELI, SLC19A1, LARS2, OPN3, ASPM, CCT2, UBE21, STK6, FLJ13052, FLJ12525, BIRC5, CKS1B, CKAP1, MGC57827, DKFZp7790175, PFN1, ILF3, IFI16, TBRG4, PAPD1, EIF2C2, MGC4308, EN1L, DSG2, EXOSC4, TAGLN2, RUVBL1, ALDOA, CPSF3, MGC15606, LGALS1, RAD18, SNX5, PSMD4, RAN, KIF14, CBX3, TMPO, DKFZP586L0724, WEE1, ROBO1, TCOF1, YWHAZ, or MPHOSPH1, relative to a control with a favorable prognosis, is associated with a poor prognosis.

2. The method of claim 1 , wherein the one or more suitable controls comprises a normal healthy individual or an individual with a favorable prognosis based on a gene expression profile for genes comprising GNG10, PNPLA4, KIAA1754, AHCYL1, MCLC, EVI5, AD-020, PARG1, CTBS, FUCA1, RFP2, FLJ20489, LTBP1, TRIP13, AIM2, SELI, SLC19A1, LARS2, OPN3, ASPM, CCT2, UBE21, STK6, FLJ13052, FLJ12525, BIRC5, CKS1B, CKAP1, MGC57827, DKFZp7790175, PFN1, ILF3, IF116, TBRG4, PAPD1, EIF2C2, MGC4308, ENO1, DSG2, EXOSC4, TAGLN2, RUVBL1, ALDOA, CPSF3, MGC15606, LGALS1, RAD18, SNX5, PSMD4, RAN, KIF14, CBX3, TMPO, DKFZP586L0724, WEE1, ROBO1, TCOF1, YWHAZ, and MPHOSPH.

3. The method of claim 1 , wherein measuring the gene expression levels comprises generating cDNA from the isolated nucleic acid sample isolated from the plasma cells.

4. The method of claim 1 , wherein the patient is undergoing, or previously underwent, treatment comprising high-dose chemotherapy and/or autologous peripheral blood stem cell transplantation.

5. The method of claim 1 , wherein the nucleic acid sample is artificially and detectably labeled.

6. The method of claim 5 , wherein the expression profile is measured by detecting a complex between the artificially and detectably labeled nucleic acid sample and a synthetic microarray.
