Abstract

The present invention concerns methods and compositions for isolating, enriching, and/or labeling miRNA molecules and for preparing and using arrays or other detection techniques for miRNA analysis. Moreover, the present invention concerns methods and compositions for generating miRNA profiles and employing such profiles for therapeutic, diagnostic, and prognostic applications.

Claims (24)

1. A method for identifying a patient as having a marker for lung cancer comprising

a) analyzing mir-205 expression in a lung tissue sample from a patient suspected of having lung cancer; and

b) identifying the patient as having a marker correlated with lung cancer when an increase in miR-205 of at least 50% in the lung tissue sample compared to a noncancerous lung tissue sample is determined or identifying the patient as not having the marker when the increase in miR-205 in the lung tissue sample fails to be determined.

2. The method of claim 1 , wherein analyzing the sample comprises generating an miRNA profile from the sample, wherein the profile comprises multiple miRNAs.

3. The method of claim 2 , wherein the generating the miRNA profile comprises:

(a) labeling miRNA isolated from the lung tissue sample;

(b) hybridizing the miRNA to an miRNA array; and,

(c) determining miRNA hybridization to the array.

4. The method of claim 1 , further comprising analyzing the lung tissue sample for a decrease in miR-126 expression compared to the noncancerous sample.

5. The method of claim 1 , wherein the patient is determined to have an increase in miR-205 expression.

6. The method of claim 4 , wherein the patient is determined to have a decrease in miR-126 expression.

7. The method of claim 1 , further comprising analyzing the lung tissue sample for a decrease in expression of let-7, miR-26a, miR-30a-AS, miR-95, miR-125a, miR-331, or miR-342, or an increase in expression of miR-200b compared to the noncancerous sample.

8. A method for identifying a patient as having a marker correlated with lung cancer comprising:

a) obtaining from a patient a lung tissue sample, wherein the patient is suspected of having lung cancer;

b) identifying the patient as having a marker correlated with lung cancer if an increase of at least 50% in miR-205 expression in the lung tissue sample compared to a noncancerous lung tissue sample is determined or identifying the patient as not having the marker if the increase is not determined.

9. The method of claim 8 , wherein the increase in the lung tissue sample was determined by generating an miRNA profile from the sample.

10. The method of claim 9 , wherein generating the miRNA profile comprises:

(a) labeling the miRNA isolated from the sample;

(b) hybridizing the miRNA to an miRNA array; and,

(c) determining miRNA hybridization to the array.

11. The method of claim 8 , further comprising identifying the patient as having a marker correlated with lung cancer if a decrease in miR-126 expression compared to the noncancerous sample is determined.

12. The method of claim 8 , wherein the patient is determined to have an increase in miR-205 expression.

13. The method of claim 11 , wherein the patient is determined to have an decrease in miR-126 expression.

14. The method of claim 8 , wherein the lung tissue sample from the patient is analyzed for a decrease in expression of let-7, miR-26a, miR-30a-AS, miR-95, miR-125a, miR-331, or miR-342 or an increase in expression of miR-200b compared to the noncancerous sample.
