Abstract

Reagents, methods and immunodiagnostic test kits for the accurate detection of hepatitis B virus (HBV) infection are disclosed. The methods and kits employ novel rabbit monoclonal antibodies directed against HBV surface antigens (HBsAg) with mutations in the “a” determinant region of HBsAg.

Claims (9)

1. A hybridoma selected from Hybridoma 99S6 (ATCC Accession number PTA-6015) or Hybridoma 99S9 (ATCC Accession number PTA-6014).

2. An antibody produced by the hybridoma of claim 1 .

3. A rabbit monoclonal antibody that recognizes the same epitope as an antibody produced by hybridoma 99S6 (ATTC Accession No. PTA-6015) or 99S9 (ATCC Accession No. PTA-60 14).

4. A solid support comprising at least one rabbit monoclonal antibody according to either of claims 2 or 3 .

5. The solid support of claim 4 , wherein the support further comprises one or more additional antibodies directed against a wild-type HBsAg or an HBsAg mutant with a mutation in the “a” determinant region.

6. The solid support of claim 5 , wherein the one or more additional antibodies comprise an additional monoclonal antibody.

7. The solid support of claim 6 , wherein the one or more additional antibodies comprise a mouse monoclonal antibody.

8. The solid support of claim 4 , further comprising at least two internal controls, wherein one of the controls defines the lower detection limit for a positive result in an immunoassay using the solid support and the other control defines a highly positive result in an immunoassay using the solid support.

9. The solid support of claim 4 , wherein the solid support is a nitrocellulose strip.
