Abstract

The present invention provides an improved coating for surfaces of medical implants. The coating comprises at least one interfacial biomaterial (IFBM) which is comprised of at least one binding module that binds to the surface of an implant or implant-related material (“implant module”) and at least one binding module that selectively binds to a target analyte or that is designed to have a desired effect (“analyte module”). The modules are connected by a linker. In some embodiments, the IFBM coating acts to promote the recognition and attachment of target analytes to surface of the device. The IFBM coating improves the performance of implanted medical devices, for example, by promoting osteointegration of the implant.

Claims (16)

1. A coating for an implant, the coating comprising:

at least one peptide consisting of 7 to 60 amino acids, which peptide binds specifically to a surface of the implant, wherein the surface is selected from the group consisting of a polymer, a ceramic, a plastic, a metal, a metal oxide, a non-metal oxide, and a combination thereof; and at least one peptide consisting of 7 to 60 amino acids, which peptide binds specifically to a bone morphogenetic protein and has a core binding motif consisting of SEQ ID NO:28 or a conservatively substituted variant consisting of one or more conservative substitutions in the core binding motif other than for Pro and Gly residues in the core binding motif, and wherein the surface- and the bone morphogenetic-binding peptide are linked and can be modified to comprise one or more of a terminal modification and a modification to facilitate the linking between the surface- and the bone morphogenetic-binding peptides which binds specifically to a surface of the implant and the at least one peptide which binds specifically to bone morphogenetic protein.

2. The coating according to claim 1 , wherein the surface is a polymer selected from the group consisting of a mixture of polymers, a natural polymer, a synthetic polymer, and a combination thereof.

3. The coating according to claim 1 , wherein the linker is selected from the group consisting of bonds of the peptides to be linked, an amino acid linker, a polymer linker, and a chemical linker.

4. The coating according to claim 1 , wherein the surface is a plastic selected from the group consisting of polyethylene, polyetheretherketone, and polytetrafluoroethylene.

5. The coating according to claim 1 , wherein the surface is a metal selected from the group consisting of titanium, stainless steel, cobalt chromium alloys, titanium alloy, silicon, and bioactive glass.

6. The coating according to claim 1 , wherein the surface comprises a demineralized bone matrix.

7. The coating according to claim 1 , wherein the at least one peptide, which binds specifically to the bone morphogenetic protein, binds specifically to at least two different bone morphogenetic proteins.

The coating according to claim 1 , wherein the conservatively substituted variant of the core binding motif of SEQ ID NO:28 is selected from the group consisting of SEQ ID NO:25, SEQ ID NO:26, SEQ ID NO:78, SEQ ID NO:79, SEQ ID NO:80, SEQ ID NO:81, SEQ ID NO:83, SEQ ID NO:84, SEQ ID NO:89, SEQ ID NO:91, SEQ ID NO:92, SEQ ID NO:93, and SEQ ID NO:94.

9. A method for coating an implant, the method comprising applying a coating according to claim 1 to a surface of an implant to be coated so that coated is the surface of the implant.

10. The method according to claim 9 , further comprising contacting the coating to bone morphogenetic protein, in binding bone morphogenetic protein to the coated implant.

11. The method according to claim 10 , wherein contacting the coating to bone morphogenetic protein, in binding bone morphogenetic protein to the coated implant, is performed prior to placing the implant into a subject in need of the implant.

12. The method according to claim 10 , wherein contacting the coating to bone morphogenetic protein, in binding bone morphogenetic protein to the coated implant, is performed after placing the implant into a subject in need of the implant.

13. The method according to claim 9 , wherein the surface comprises a demineralized bone matrix.

The method according to claim 9 , wherein the conservatively substituted variant of the core binding motif of SEQ ID NO:28 is selected from the group consisting of SEQ ID NO:25, SEQ ID NO:26, SEQ ID NO:78, SEQ ID NO:79, SEQ ID NO:80, SEQ ID NO:81, SEQ ID NO:83, SEQ ID NO:84, SEQ ID NO;89, SEQ ID NO:91, SEQ ID NO:92, SEQ ID NO:93, and SEQ ID NO:94.

15. The method according to claim 9 , wherein the linker is selected from the group consisting of bonds of the peptides to be linked, an amino acid linker, a polymer linker, and a chemical linker.
