Abstract

Methods and compositions for producing flowable compositions, e.g. pastes, that set into calcium phosphate products are provided. In the subject methods, dry reactants that include a calcium source and a phosphate source are combined with a setting fluid to produce the flowable composition. A feature of the subject methods is that the dry reactants include a particulate calcium and/or phosphate reactant having a mean particle size of less than about 8 μm and narrow size distribution. Also provided are the compositions themselves as well as kits for use in practicing the subject methods. The subject methods and compositions produced thereby find use in a variety of applications, including the repair of hard tissue defects, e.g., bone defects.

Claims (46)

1. A method of producing a flowable composition that sets into a calcium phosphate containing product, said method comprising:

combining:

(a) a setting fluid; and

(b) a dry reactant component comprising a first particulate calcium and/or phosphate reactant having a mean particle size of about 8 μm or less and narrow particle size distribution;

in a ratio of (a) to (b) sufficient to produce said flowable material that has a setting value of about 400 Newtons or more in 6 minutes.

2. The method according to claim 1 , wherein said dry reactant component consists of said first particulate calcium and/or phosphate reactant.

3. The method according to claim 1 , wherein said dry reactant component comprises a second calcium and/or phosphate reactant having a mean particle size that is about 10 μm or greater.

4. The method according to claim 3 , wherein said dry reactant component further comprises a source of phosphate ion.

5. The method according to claim 4 , wherein said source of phosphate ion comprises NaH 2 PO 4 .

6. The method according to claim 1 , wherein said dry reactant component further comprises an emulsifier.

7. The method according to claim 6 , wherein said emulsifier is a cellulose ether.

8. The method according to claim 7 , wherein said cellulose ether is carboxymethyl cellulose.

9. The method according to claim 1 , wherein said flowable material has a setting value of about 200 Newtons or more in 3 minutes.

10. The method according to claim 2 , wherein the ratio of said first to second calcium and/or phosphate reactants in said dry reactant component ranges from about 1 to about 10.

11. The method according to claim 1 , wherein said first particulate calcium and/or phosphate reactant is a calcium phosphate compound.

12. The method according to claim 11 , wherein the ratio of calcium to phosphate of said calcium phosphate compound ranges from about 1 to about 2.

13. The method according to claim 12 , wherein said calcium phosphate compound is a tricalcium phosphate.

14. The method according to claim 1 , wherein said first particulate calcium and/or phosphate reactant has a mean particle size of about 4 μm.

15. The method according to claim 1 , wherein said setting fluid is a solution of a soluble silicate.

16. The method according to claim 1 , wherein said ratio of dry reactants to setting fluid ranges from about 0.2:1 to 0.7:1.

17. The method according to claim 1 , wherein said first particulate calcium and/or phosphate reactant is air pulverized.

18. A method of producing a paste that sets into a calcium phosphate containing product, said method comprising:

combining:

(a) a dry reactant component comprising:

(i) a particulate first calcium phosphate reactant having a mean particle size of about 4 μm or less and standard deviation that does not exceed about 4 μm; and

(i) a second particulate calcium and/or phosphate reactant; and

(b) a setting fluid;

in a ratio of (a) to (b) sufficient to produce said paste that has a setting value of about 400 Newtons or more in 6 minutes.

19. The method according to claim 18 , wherein said second calcium and/or phosphate reactant has a mean particle size that is about 9 μm or greater.

20. The method according to claim 18 , wherein the ratio of calcium to phosphate of said first calcium phosphate reactant ranges from about 1 to about 2.

21. The method according to claim 20 , wherein said first calcium phosphate reactant is a tricalcium phosphate.

22. The method according to claim 18 , wherein said first calcium phosphate reactant has a mean particle size of about 3 μm.

23. The method according to claim 18 , wherein said setting fluid is a solution of a soluble silicate.

24. The method according to claim 18 , wherein said first particulate calcium and/or phosphate reactant is air pulverized.

25. The method according to claim 18 , wherein said paste has a setting value of about 200 Newtons or more in 3 minutes.

26. A flowable composition that sets into a calcium phosphate containing product, wherein said composition is produced by the method according to claim 1 .

27. A kit comprising:

a dry reactant component comprising a first particulate calcium and/or phosphate reactant having a mean particle size of about 8 μm or less and narrow particle size distribution; and

a setting fluid;

wherein when said dry reactant and setting fluid are combined according to the method of claim 1 , a flowable material is produced that has a setting value of about 400 Newtons or more in 6 minutes.

28. The kit according to claim 27 , wherein said flowable material has a setting value of about 200 Newtons or more in 3 minutes.

29. A system comprising:

(a) a dry reactant component comprising first particulate calcium and/or phosphate reactant having a mean particle size of about 8 μm or less and a narrow particle size distribution; and

(b) a setting fluid;

wherein when said dry reactant and setting fluid are combined according to the method of claim 1 , a flowable material is produced that has a setting value of about 400 Newtons or more in 6 minutes.

30. The system according to claim 27 , wherein said flowable material has a setting value of about 200 Newtons or more in 3 minutes.
