Abstract

The present invention provides isolated polypeptides isolatable from a Fusobacterium spp. Also provided by the present invention are compositions that include one or more of the polypeptides, and methods for making and methods for using the polypeptides.

Claims (20)

1. A composition comprising: at least two isolated polypeptides having molecular weights between 76 kDa and 86 kDa, between 62 kDa and 68 kDa, between 45 kDa and 53 kDa, between 34 kDa and 43 kDa, or between 24 kDa and 35 kDa and are natively expressed at a detectable level by a Fusobacterium spp. when incubated in medium comprising a metal chelator and are not natively expressed by the Fusobacterium spp. at a detectable level when grown in the medium without the metal chelator, wherein molecular weight is determined by electrophoresis on a sodium dodecyl sulfate-polyacrylamide gel and further comprising an adjuvant.

2. The composition of claim 1 wherein the composition further comprises a pharmaceutically acceptable carrier.

3. The composition of claim 1 wherein the composition does not comprise a polypeptide having leukotoxin activity.

4. The composition of claim 1 wherein each of the at least two polypeptides has a molecular weight between 78 kDa and 84 kDa, between 64 kDa and 66 kDa, between 47 kDa and 51 kDa, between 36 kDa and 41 kDa, or between 27 kDa and 32 kDa.

5. The composition of claim 1 wherein each of the at least two polypeptides has a molecular weight of 83 kDa, 79 kDa, 65 kDa, 49 kDa, 39 kDa, 31 kDa, or 28 kDa.

The composition of claim 1 further comprising at least one isolated polypeptide having a molecular weight of between 136 kDa and 144 kDa, between 69 kDa and 77 kDa, between 39 kDa and 47 kDa, between 29 kDa and 37 kDa, and is natively expressed by a Fusobacterium spp. when grown in medium with or without a metal chelator, wherein native expression of the polypeptide is enhanced during growth in the medium comprising a metal chelator.

7. The composition of claim 1 wherein the Fusobacterium spp. is F. necrophorum or F. nucleatum.

8. The composition of claim 1 wherein Fusobacterium spp. is the F. necrophorum strain deposited in the American Type Culture Collection (ATCC) and given the ATCC Accession No. PTA-10318.

9. A method for detecting antibody comprising:

providing a biological sample comprising antibody;

contacting the biological sample with an isolated polypeptide of the composition of claim 1 to form a mixture;

incubating the mixture; and

identifying a polypeptide:antibody complex, wherein the presence of a polypeptide:antibody complex indicates the biological sample comprises antibody that specifically binds to the polypeptide.

10. The method of claim 9 wherein the biological sample is blood, milk, or colostrum.

11. A kit for detecting antibody that specifically binds a polypeptide, comprising in separate containers:

an isolated polypeptide of the composition of claim 1 ; and

a reagent that detects an antibody that specifically binds the polypeptide.

A composition comprising an isolated Fusobacterium spp. polypeptide, wherein the polypeptide has a molecular weight between 76 kDa and 86 kDa, between 62 kDa and 68 kDa, between 45 kDa and 53 kDa, between 34 kDa and 43 kDa, or between 24 kDa and 35 kDa and is natively expressed at a detectable level by a Fusobacterium spp. when incubated in medium comprising a metal chelator and is not natively expressed at a detectable level by the Fusobacterium spp. when grown in the medium without the metal chelator, wherein molecular weight is determined by electrophoresis on a sodium dodecyl sulfate-polyacrylamide gel, a pharmaceutically acceptable carrier, and an adjuvant.

13. The composition of claim 1 wherein the metal chelator is 2,2′-dipyridyl.

14. The composition of claim 12 wherein the metal chelator is 2,2′-dipyridyl.
