Abstract

The present invention provides methods for isolating a PHA. The method comprises combining the PHA, a first solvent and a second solvent to form a combination, the first solvent and the second solvent being capable of forming an azeotrope with the second solvent; and heating the combination to substantially remove the first solvent from the combination. Alternatively, the method comprises combining the PHA, a first solvent, a second solvent and a third solvent to form a combination, the first solvent and the second solvent being capable of forming an azeotrope with the third solvent; and heating the combination to substantially remove the first solvent and the second solvent from the combination.

Claims (24)

1. A method of isolating a PHA, the method comprising:

a) extracting a biomass comprising one or more PHAs with an extraction solvent to form a mixture including a PHA phase and a residual biomass phase, wherein the PHA phase comprises the PHA dissolved in the extraction solvent;

b) isolating the PHA phase from the residual biomass phase;

c) combining the PHA phase with a PHA receiving solvent to form a first combination, wherein the receiving solvent forms an azeotrope with the extraction solvent;

d) removing substantially the PHA extraction solvent from the first combination as an azeotropic mixture with a portion of the PHA receiving solvent to form a second combination comprising the PHA and the PHA receiving solvent; and

e) isolating the PHA from the second combination.

2. The method of claim 1 , wherein the PHA extraction solvent is selected from the group consisting of MIBK, butyl acetate, ethyl acetate, cyclopentanone and cyclohexanone.

3. The method of claim 1 , wherein the PHA receiving solvent is water.

4. The method of claim 1 , wherein the mixture in step a) is formed by agitating the biomass with the extraction solvent.

5. The method of claim 1 , wherein the PHA extraction solvent is selected from the group consisting of ketones, esters, alcohols and alkanes.

6. The method of claim 1 , wherein the PHA extraction solvent is a single solvent and forms a binary azeotrope with a single PHA receiving solvent.

7. The method of claim 1 , wherein the PHA extraction solvent is a single solvent and forms a ternary azeotrope with two PHA receiving solvents.

8. The method of claim 7 , wherein the PHA receiving solvents are each selected from water, an alkane, methanol or ethanol.

9. The method of claim 8 , wherein the PHA receiving solvent is a straight-chain alkane selected from heptane, hexane, octane, nonane and dodecane.

10. The method of claim 1 , wherein the PHA is a homopolymer.

11. The method of claim 9 , wherein the homopolyer is selected from the group consisting of poly-4-hydroxybutyrate, poly-4-hydroxyvalerate, poly-3-hydroxypropionate, poly-3-hydroxybutyrate, poly-3-hydroxyhexanoate, poly-3-hydroxyheptanoate, poly-3-hydroxyoctanoate, poly-3-hydroxydecanoate and poly-3-hydroxydodecanoate.

12. The method of claim 11 , wherein the homopolymer is poly-3-hydroxybutyrate.

13. The method of claim 12 , wherein the extraction solvent is cyclohexanone, cyclopentanone or a mixture of MIBK and cyclohexanone.

14. The method of claim 13 , wherein the receiving solvent is water.

15. The method of claim 1 , wherein the PHA is a copolymer.

The method of claim 15 , wherein the poly-3-hydroxybutyrate copolymer is selected from the group consisting of poly-3-hydroxybutyrate-co-3-hydroxypropionate, poly-3-hydroxybutyrate-co-3-hydroxyvalerate, poly-3-hydroxybutyrate-co-3-hydroxyhexanoate, poly-3-hydroxybutyrate-co-4-hydroxybutyrate, poly-3-hydroxybutyrate-co-4-hydroxyvalerate, poly-3-hydroxybutyrate-co-6-hydroxyhexanoate, poly 3-hydroxybutyrate-co-3-hydroxyheptanoate, poly-3-hydroxybutyrate-co-3-hydroxyoctanoate, poly-3-hydroxybutyrate-co-3-hydroxydecanoate, poly-3-hydroxybutyrate-co-3-hydroxydodecanotate, poly-3-hydroxybutyrate-co-3-hydroxyoctanoate-co-3-hydroxydecanoate, poly-3-hydroxydecanoate-co-3-hydroxyoctanoate, and poly-3-hydroxybutyrate-co-3-hydroxyoctadecanoate.

17. The method of claim 16 , wherein the poly-3-hydroxybutyrate copolymer is poly-3-hydroxybutyrate-co-4-hydroxybutyrate.

18. The method of claim 17 , wherein the PHA extraction solvent is MIBK, butyl acetate or cyclohexanone.

19. The method of claim 18 , wherein the PHA receiving solvent is water.
