Abstract

The present invention provides an antibody or peptide which immunologically reacts with human VEGF receptor Flt-1 and cells in which human VEGF receptor Flt-1 is expressed on the cell surface and an antibody or peptide which inhibits binding of human VEGF to human VEGF receptor Flt-1. It also provides a means for the diagnosis or treatment of diseases in which their morbid states progress by abnormal angiogenesis, such as proliferation or metastasis of solid tumors, arthritis in rheumatoid arthritis, diabetic retinopathy, retinopathy of prematurity, psoriasis, and the like.

Claims (14)

1. A method of treating a disease in which the morbid state progresses by abnormal angiogenesis, comprising administering to a human or an animal in need of such treatment an effective amount of a recombinant monoclonal antibody selected from the group consisting of a humanized antibody and an antibody fragment thereof, said recombinant monoclonal antibody specifically binding to the human VEGF receptor Flt-1, and

wherein the recombinant monoclonal antibody binds to an epitope present in a region of the 100 th to 204 th positions from the N-terminal amino acid including a signal sequence of the human VEGF receptor Flt-1, and

said recombinant monoclonal antibody comprises an antibody heavy chain (H chain) variable region (V region), and an antibody light chain (L chain) variable region (V region), and

wherein said H chain V region comprises the amino acid sequence of SEQ ID NO:91.

A method of treating a disease in which the morbid state progresses by abnormal angiogenesis, comprising administering to a human or an animal in need of such treatment an effective amount of a recombinant monoclonal antibody selected from the group consisting of a humanized antibody and an antibody fragment thereof, said recombinant monoclonal antibody specifically binding to the human VEGF receptor Flt-1, and

wherein the recombinant monoclonal antibody binds to an epitope present in a region of the 100 th to 204 th positions from the N-terminal amino acid including a signal sequence of the human VEGF receptor Flt-1, and

said humanized antibody is selected from the group consisting of a human chimeric antibody and a human complementarity-determining region (CDR)-grafted antibody, and

wherein the CDR-grafted antibody comprises:

CDR in a heavy chain (H chain) variable region (V region) comprising the amino acid sequences of SEQ ID NOs:15, 16, and 17, and

CDR in a light chain (L chain) variable region (V region) comprising the amino acid sequences of SEQ ID NOs:18, 19 and 20.

3. The method according to claim 2 , wherein the human CDR-grafted antibody comprises:

the H chain V region comprising the amino acid sequence of SEQ ID NO:91, and

the L chain V region comprising the amino acid sequence of SEQ ID NO:93, 94 or 96.

4. The method according to either claim 1 or 2 , wherein the antibody fragment is selected from the group consisting of Fab, Fab′, F(ab′) 2 , a single chain antibody, and a disulfide stabilized antibody.
