Abstract

Polyampholyte are able to condense nucleic acid to form small complexes which can be utilized in the delivery of nucleic acid to mammalian cells. The polyampholytes can be formed prior to interaction with nucleic acid or they can be formed in the presence of nucleic acid. Stabilized polycation/nucleic acid complexes can be modified to reduce the positive charge of the polycation and add targeting ligands without destabilizing the complex. The resultant particles retain their small size and are more effective in delivery of nucleic acid to cells in vivo.

Claims (21)

1. A process for delivering a nucleic acid to a cell, comprising:

a) condensing the nucleic acid with a polycation to form a complex with a positive surface charge;

b) crosslinking the polycation;

c) reducing the surface charge of the complex by chemically modifying positively charged moieties on the polycation with a maleic anhydride derivative; and,

d) contacting the cell with the reduced surface charge complex.

2. The process of claim 1 further comprising modifying the polycation to add a targeting ligand.

3. The process of claim 2 wherein the targeting ligand consists of galactose.

4. The process of claim 1 wherein the maleic anhydride derivative is charge neutral.

5. The process of claim 1 wherein the maleic anhydride derivative negatively charged.

6. The process of claim 1 wherein the maleic anhydride derivative contains an interaction modifier group.

7. The process of claim 6 wherein the interaction modifier group consists of polyethylene glycol.

8. The process of claim 1 wherein the maleic anhydride derivative contains a targeting group.

9. The process of claim 8 wherein the targeting group contains a galactose molecule.

10. The process of claim 1 wherein the maleic anhydride derivative consists of a dimethylmaleic anhydride derivative.

11. The process of claim 1 wherein the maleic anhydride derivative consists of a 2-propionic-3-methylmaleic anhydride.

12. The process of claim 11 wherein the 2-propionic-3-methylmaleic anhydride consists of a 2-propionic-3-methylmaleic anhydride-polyethylene glycol.

13. The process of claim 1 wherein crosslinking the polycation comprises adding a multivalent crosslinker to the complex.

14. The process of claim 13 wherein the crosslinker consists of a labile crosslinker.

15. The process of claim 14 wherein the crosslinker consists of a cleavable crosslinker.

16. The process of claim 15 wherein the cleavable crosslinker consists of a disulfide bond containing crosslinker.

17. The process of claim 16 wherein the disulfide bond containing crosslinker consists of a disulfide bond dialdehyde.
