View Patent ↗Engineered strains of the oleaginous yeast Yarrowia lipolytica capable of producing greater than 10% arachidonic acid (ARA, an ω-6 polyunsaturated fatty acid) in the total oil fraction are described. These strains comprise various chimeric genes expressing heterologous desaturases, elongases and acyltransferases, and optionally comprise various native desaturase and acyltransferase knockouts to enable synthesis and high accumulation of ARA. Production host cells are claimed, as are methods for producing ARA within said host cells.
1. A recombinant oleaginous Yarrowia yeast cell for the production of arachidonic acid comprising at least one gene encoding a Δ8 desaturase wherein;
a) the at least one gene encoding Δ8 desaturase encodes a polypeptide having at least 95% identity to the amino acid sequence as set forth in SEQ ID NO:49; and
b) the oleaginous Yarrowia yeast cell accumulates in excess of about 25% of its dry cell weight as oil; and
c) the oleaginous Yarrowia yeast cell produces about 14% arachidonic acid of total lipid produced.
2. The recombinant oleaginous Yarrowia yeast cell according to claim 1 further comprising at least one gene encoding Δ12 desaturase having at least 95% identity to the amino acid sequence as set forth in SEQ ID NO:28.
3. The recombinant oleaginous Yarrowia yeast cell according to claim 2 wherein the Yarrowia cell is devoid of any native gene encoding a polypeptide having Δ12 desaturase activity.
4. The recombinant oleaginous Yarrowia yeast cell according to claim 1 , wherein the at least one gene encoding Δ8 desaturase is under the control of a promoter sequence having the nucleic acid sequence selected from the group consisting of SEQ ID NOs:158-168 and 364.
5. The recombinant oleaginous Yarrowia yeast cell according to claim 1 further comprising at least one gene encoding C16/18 elongase having at least 95% identity to the amino acid sequence as set forth in SEQ ID NO:54.
6. The recombinant oleaginous Yarrowia yeast cell according to claim 1 wherein the yeast cell produces a microbial oil comprising arachidonic acid and wherein the microbial oil is devoid of any γ-linoleic acid.
7. A method for the production of a microbial oil comprising arachidonic acid comprising:
a) culturing the host of claim 1 wherein a microbial oil comprising arachidonic acid is produced; and
b) optionally recovering the microbial oil of step (a).
