α-1,3-fucosyltransferase
The invention provides a novel species of α-1,3-fucosyltransferase expressed by a gene cloned from animal cells, a cDNA coding for the α-1,3-fucosyltransferase, a method of detecting, or inhibiting the production of, the α-1,3-fucosyltransferase which involves the use of the cDNA, a recombinant vector containing the DNA as an insert, a cell harboring the recombinant vector, and a method of producing same. The α-1,3-fucosyltransferase of the invention is useful in the production of carbohydrate chains having useful physiological activity, for example sialyl Lewis x, and modifications thereof.
1. An isolated DNA which hybridizes with the DNA consisting of the nucleotide sequence defined in SEQ ID NO:1 under stringent conditions which comprises performing hybridization at 65° C. in the presence of 1.0M sodium chloride and then washing at 65° C. in a 0.1-fold concentrated SSC solution, and which encodes a protein having α-1,3-fucosyltransferase activity to synthesize sialyl Lewis x from sialyl-lacto-N-neotetraose.
2. A recombinant vector comprising, as an insert, the DNA of claim 1 .
3. An isolated host cell transformed with the recombinant vector of claim 2 .
4. An Escherichia coil strain transformed with the recombinant vector of claim 2 .
5. A method of producing an α-1,3-fucosyltransferase comprising culturing the cell according to claim 3 under conditions such that said sequence is expressed and said α-1,3-fucosyltransferase is thereby produced.
6. A method of producing an α-1,3-fucosyltransferase comprising culturing the cell according to claim 4 under conditions such that said sequence is expressed and said α-1,3-fucosyltransferase is thereby produced.

A method of introducing the sialyl Lewis x structure onto a carbohydrate chain of a glycoprotein or glycolipid having a NeuAcα2, 3Galβ1, 4GlcNAc structure at a non-reducing end of said carbohydrate chain comprising culturing a cell producing said glycoprotein or glycolipid and transformed with a recombinant vector comprising, as an insert, a DNA of claim 1 , under conditions such that said DNA is expressed and an α-1, 3-fucosyltransferase encoded by said DNA is thereby produced, and such that said α-1, 3-fucosyltransferase effects the introduction of said sialyl Lewis x structure into a carbohydrate chain of the glycoprotein or glycolipid present in said cell.
