Abstract

The present invention relates to substituted triazole compounds and compositions comprising substituted triazole compounds. The invention further relates to methods of inhibiting the activity of Hsp90 in a subject in need thereof and methods for preventing or treating hyperproliferative disorders, such as cancer, in a subject in need thereof comprising administering to the subject a substituted triazole compound of the invention, or a composition comprising such a compound.

Claims (297)

1. A compound represented by the following structural formula

a tautomer, or a pharmaceutically acceptable salt thereof, wherein:

R 2 is a substituted phenyl, wherein the phenyl group is substituted with:

i) one substituent selected from nitro, cyano, a haloalkoxy, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, an optionally substituted heteraralkyl, hydroxylalkyl, alkoxyalkyl, guanadino, —NR 10 R 11 , —O—R 20 , —C(O)R 7 , —C(O)OR 20 , —OC(O)R 7 , —C(O)NR 10 R 11 , —NR 8 C(O)R 7 , —SR 7 , —S(O) p R 7 , —OS(O) p R 7 , —S(O) p OR 7 , —NR 8 S(O) p R 7 , —S(O) p NR 10 R 11 , —S(O) p OR 7 , —OP(O)(OR 7 ) 2 , and —SP(O)(OR 7 ) 2 ; or

ii) two to five substituents selected from the group consisting of an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, an optionally substituted heteraralkyl, hydroxyalkyl, alkoxyalkyl, —F, —Br, —I, cyano, nitro, guanadino, a haloalkyl, a heteroalkyl, —NR 10 R 11 , —OR 7 , —C(O)R 7 , —C(O)OR 20 , —OC(O)R 7 , —C(O)NR 10 R 11 , —NR 8 C(O)R 7 , —SR 7 , —S(O) p R 7 , —OS(O) p R 7 , —S(O) p OR 7 , —NR 8 S(O) p R 7 , —S(O) p NR 10 R 11 , —OP(O)(OR 7 ) 2 , —SP(O)(OR 7 ) 2 , and —S(O) p OR 7 ;

R 7 and R 8 , for each occurrence, are, independently, —H, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralky, or an optionally substituted herteraralkyl;

R 10 and R 11 , for each occurrence, are independently —H, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, or an optionally substituted heteraralkyl; or R 10 and R 11 , taken together with the nitrogen to which they are attached, form an optionally substituted heterocyclyl or an optionally substituted heteroaryl;

R 26 is a C1-C6 alkyl;

p, for each occurrence, is, independently, 0, 1 or 2;

m, for each occurrence, is independently, 1, 2, 3, or 4;

R 20 , for each occurrence, is independently an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, or an optionally substituted heteraralkyl;

provided that R 18 is not cyclohexyl.

3. The compound of claim 2 , wherein R 18 is an optionally substituted cycloalkyl or an optionally substituted cycloalkenyl.

4. The compound of claim 2 , wherein R 18 is a substituted alkyl.

3-(2-Hydroxyphenyl)-4-(naphthalen-1-yl)-5-mercapto-triazole;

30. The compound of claim 6 , wherein said compound is selected from the group consisting of:

3-(2,4-dihydroxy-5-ethyl-phenyl)-4-(1-ethyl-benzimidazol-4-yl)-5-mercapto-[1,2,4]triazole,

3-(2,4-dihydroxy-5-ethyl-phenyl)-4-(1-ethyl-benzimidazol-4-yl)-5-mercapto-[1,2,4]triazole,

3-(2,4-dihydroxy-5-ethyl-phenyl)-4-(2-methyl-3-ethyl-benzimidazol-5-yl)-5-mercapto-[1,2,4]triazole,

3-(2,4-dihydroxy-5-ethyl-phenyl)-4-(1-ethyl-2-methyl-benzimidazol-5-yl)-5-mercapto-[1,2,4]triazole, and

3-(2,4-dihydroxy-5-isopropyl-phenyl)-4-(1-methyl-2-trifluoromethyl-benzimidazol-5-yl)-5-mercapto-[1,2,4]triazole,

a tautomer, or a pharmaceutically acceptable salt therof.

31. The compound of claim 6 , wherein said compound is 3-(2,4-dihydroxy-5-isopropyl-phenyl)-4-(N-methyl-indol-5-yl)-5-mercapto-[1,2,4]triazole or a tautomer, or a pharmaceutically acceptable salt therof.

32. A pharmaceutical composition, comprising a pharmaceutically acceptable carrier and one or more compounds represented by the formula of claim 6 .

33. The compound of claim 1 wherein R 1 , R 3 and R 25 are each independently —OH, —SH, —NHR 7 , —OC(O)NR 10 R 11 , —SC(O)NR 10 R 11 , —OC(O)R 7 , —SC(O)R 7 , —OC(O)OR 7 , —SC(O)OR 7 , —OS(O) p R 7 , —S(O) p OR 7 , —SS(O) p R 7 , —OS(O) p OR 7 , —SS(O) p OR 7 , —OC(S)R 7 , —SC(S)R 7 , —OC(S)OR 7 , —SC(S)OR 7 , —OC(S)NR 10 R 11 , —SC(S)NR 10 R 11 , —OC(R 8 )R 7 , —SC(NR 8 )R 7 —OC(NR 8 )OR 7 , —SC(NR 8 )OR 7 , —OP(O)(OR 7 ) 2 or —SP(O)(OR 7 ) 2 .

34. The compound of claim 33 , wherein R 1 and R 3 are each, independently, —OH, —SH, or —NHR 7 .

35. The compound of claim 34 , wherein the compound is represented by the following structural formula:

a tautomer, or a pharmaceutically acceptable salt thereof.

36. The compound of claim 35 , wherein: R 1 is —SH or —OH; R 3 and R 25 are —OH; and R 6 is a lower alkyl, C3-C6 cycloalkyl, lower alkoxy, a lower alkyl sulfanyl, or —NR 10 R 11 .

37. The compound of claim 36 , wherein R 6 is a C1-C6 alkyl, a C1-C6 alkoxy, a C1-C6 alkyl sulfanyl or a C3-C6 cycloalkyl.

38. The compound of claim 2 wherein R 1 , R 3 and R 25 are each independently —OH, —SH, —NHR 7 , —OC(O)NR 10 R 11 , —SC(O)NR 10 R 11 , —OC(O)R 7 , —SC(O)R 7 , —OC(O)OR 7 , —SC(O)OR 7 , —OS(O) p R 7 , —S(O) p OR 7 , —SS(O) p R 7 , —OS(O) p OR 7 ,—SS(O) p OR 7 , —OC(S)OR 7 , —SC(S)OR 7 , —OC(S)OR 7 , —SC(S)OR 7 , —OC(S)(NR 10 R 11 , —SC(S)(NR 10 R 11 , —OC(NR 8 )R 7 , —SC(NR 8 )R 7 , —OC(NR 8 )OR 7 , —SC(NR 8 )OR 7 , —OP(O)(OR 7 ) 2 , or —SP(O)(OR 7 ) 2 .

39. The compound of claim 38 , wherein R 1 and R 3 are each, independently, —OH, —SH, or —NHR 7 .

40. The compound of claim 39 , wherein the compound is represented by the following structural formula:

a tautomer, or a pharmaceutically acceptable salt thereof.

41. The compound of claim 40 , wherein:

R 1 is —SH or —OH;

R 3 and R 25 are —OH; and

R 6 is a lower alkyl, C3-C6 cycloalkyl, lower alkoxy, a lower alkyl sulfanyl, or —NR 10 R 11 .

42. The compound of claim 2 , wherein R 6 is a C1-C6 alkyl, a C1-C6 haloalkyl, a Cl-C6 alkoxy, a C1-C6 haloalkoxy, a C1-C6 alkyl sulfanyl or a C3-C6 cycloalkyl.

43. The compound of claim 1 , selected form the group consisting of

3-(2,4-Dihydroxyphenyl)-4-(2-methyl-4-bromophenyl)-5-mercapto-triazole;

3-(2,4-Dihydroxyphenyl)-4-(2,6-diethylphenyl)-5-mercapto-triazole;

3-(2,4-Dihydroxyphenyl)-4-(2-methy-6-ethylphenyl)-5-mercapto-triazole;

3-(2,4-Dihydroxyphenyl)-4-(2,6-diisopropylphenyl)-5-mercapto-triazole;

47. A pharmaceutical composition, comprising a pharmaceutically acceptable carrier and one or more compounds represented by the formula of claim 2 .

48. 3-(2,4-Dihydroxy-5-ethyl-phenyl)-4-(1,3-dimethyl-indol-5-yl)-5-hydroxy-[1,2,4]triazole, or a tautomer, or a pharmaceutically acceptable salt thereof.

49. 3-(2,4-Dihydroxy-5-isopropyl-phenyl)-4-(1,3-dimethyl-indol-5-yl)-5-hydroxy-[1,2,4]triazole, or a tautomer, or a pharmaceutically acceptable salt thereof.

50. 3-(2,4-Dihydroxy-5-isopropyl-phenyl)-4-(1-methyl-indol-5-yl)-5-hydroxy-[1,2,4]triazole, or a tautomer, or a pharmaceutically acceptable salt thereof.

51. 3-(2,4-Dihydroxy-5-isopropyl-phenyl)-4-(1-isopropyl-indol-4-yl)-5-hydroxy-[1,2,4]triazole, or a tautomer, or a pharmaceutically acceptable salt thereof.
