Abstract

The present invention provide purified Flt4 receptor tyrosine kinase polypeptides and fragments thereof, polynucleotides encoding such polypeptides, antibodies that specifically bind such polypeptides, and uses therefor.

Claims (78)

1. A fusion protein comprising:

(a) a polypeptide comprising a portion of a human Vascular Endothelial Growth Factor Receptor-3 (VEGFR-3) extracellular domain (EC),

wherein said portion consists essentially of the first, second, and third immunoglobulin-like domains of the VEGFR-3-EC, and

(b) an immunoglobulin-like domain of human VEGFR-2 or an immunoglobulin Fc peptide,

wherein (a) is fused to (b); and

wherein said fusion protein binds to VEGFR-3 ligands VEGF-C and VEGF-D.

2. A soluble fusion protein according to claim 1 .

3. A fusion protein comprising:

(a) a polypeptide comprising a portion of a human Vascular Endothelial Growth Factor Receptor-3 (VEGFR-3) extracellular domain (EC);

wherein said portion and said polypeptide bind to VEGFR-3 ligands VEGF-C and VEGF-D and comprise at least the first, second, and third immunoglobulin-like domains of the VEGFR-3-EC,

wherein the polypeptide lacks VEGFR-3-EC immunoglobulin-like domains IV-VII; and

(b) an immunoglobulin-like domain of human VEGFR-2 or an immunoglobulin Fc peptide,

wherein (a) is fused to (b), and

wherein the fusion protein binds VEGF-C and VEGF-D, is soluble, and lacks any transmembrane domain.

4. The fusion protein according to claim 3 , wherein the portion consists of the first, second, and third immunoglobulin-like domains of the VEGFR-3-EC.

5. The fusion protein according to claim 1 or 3 , wherein the portion of the VEGFR-3-EC consists of the first through third immunoglobulin-like domains of SEQ ID NO: 2.

6. The fusion protein according to claim 1 or 3 , wherein the polypeptide comprises amino acids 47-314 of SEQ ID NO: 2.

7. The fusion protein according to claim 1 or 3 , wherein the polypeptide is fused to an immunoglobulin-like domain of human VEGFR-2.

8. The fusion protein according to claim 1 or 3 , comprising the polypeptide is fused to an immunoglobulin Fc peptide.

9. A composition comprising the fusion protein according to claim 8 and a pharmaceutically-acceptable carrier.

10. A composition comprising the fusion protein according to claim 1 or 3 and a pharmaceutically-acceptable carrier.

11. A method of treating a neoplastic disorder comprising administering to a mammalian subject having a neoplastic disorder a composition comprising the fusion protein of claim 1 or 3 , in an amount effective to inhibit neoplastic cell growth.

12. The method according to claim 11 , wherein the neoplastic disorder is a tumor characterized by blood vessel or lymphatic vessel neovascularization, and wherein the neovascularization comprises endothelial cells that express VEGFR-3.

13. The method according to claim 12 , wherein the neoplastic disorder is breast cancer.

14. The method according to claim 11 , wherein the composition is administered in an amount effective to inhibit tumor metastasis.

15. The method according to claim 11 , comprising administering to the mammalian subject the fusion protein that comprises the polypeptide fused to the immunoglobulin Fc peptide.

16. The method according to claim 11 , wherein the mammalian subject has been diagnosed with a disease characterized by proliferation of endothelial cells that express VEGFR-3, and wherein said composition inhibits at least one of angiogenesis and lymphangiogenesis in said subject.

17. The method according to claim 16 , wherein the disease is cancer.

18. The method according to claim 17 , further comprising administering a second cancer therapeutic agent to the subject.

19. The method according to claim 18 , wherein said second cancer therapeutic agent is selected from the group consisting of a chemotherapeutic agent, a nucleic acid encoding a cancer therapeutic agent, an anti-lymphangiogenic agent, and an anti-angiogenic agent.

20. The method according to claim 17 , wherein the subject has a cancer of a tissue, organ, or cell selected from the group consisting of brain, lung, liver, spleen, kidney, lymph node, small intestine, blood cells, pancreas, colon, stomach, breast, endometrium, prostate, testicle, ovary, skin, head and neck, esophagus, bone marrow and blood.

21. The method according to claim 11 , wherein the mammalian subject is human.

22. The fusion protein of claim 1 or 3 , wherein the fusion protein is capable of inhibiting VEGF-C-mediated stimulation of endothelial cells that express VEGFR-3.

23. The fusion protein of claim 1 or 3 , wherein the fusion protein is capable of inhibiting VEGF-C-induced phosphorylation of VEGFR-3.

24. The fusion protein of claim 1 or 3 , wherein the fusion protein is capable of inhibiting VEGF-C-induced and VEGF-D-induced signaling of VEGFR-3.

25. The fusion protein of claim 1 or claim 3 , wherein the fusion protein is purified.

26. A polypeptide consisting essentially of:

(i) a fragment of human Vascular Endothelial Growth Factor Receptor-3 (VEGFR-3) consisting essentially of the first through third immunoglobulin-like domains of human VEGFR-3 extracellular domain (EC), and

(ii) an immunoglobulin Fc peptide,

wherein (i) is fused to (ii), and

wherein the polypeptide binds both VEGF-C and VEGF-D.

27. A chimeric polypeptide comprising a fragment of a human Vascular Endothelial Growth Factor Receptor-3 (VEGFR-3) extracellular domain (EC) fused to a heterologous polypeptide;

wherein said fragment binds to VEGF-C and VEGF-D and comprises at least the first through third immunoglobulin-like domains of the VEGFR-3-EC,

wherein the polypeptide lacks VEGFR-3-EC immunoglobulin-like domains IV-VII; and

wherein the polypeptide is a soluble polypeptide lacking any transmembrane domain.

28. The chimeric polypeptide according to claim 27 , wherein the heterologous polypeptide increases serum solubility or serum half-life of the fragment of human VEGFR-3-EC.

29. A composition comprising the polypeptide according to claim 26 or 27 and a pharmaceutically-acceptable carrier.

30. The polypeptide according to claim 26 or 27 , wherein the fragment comprises amino acids 47-314 of SEQ ID NO: 2.

31. The polypeptide of claim 26 or 27 , wherein the polypeptide is capable of inhibiting VEGF-C-induced phosphorylation of VEGFR-3.

32. The polypeptide according to claim 26 or 27 , wherein the polypeptide is capable of inhibiting VEGF-C-induced and VEGF-D-induced signaling of VEGFR-3.

33. The polypeptide according to claim 26 or 27 that is purified.

34. A method of treating a neoplastic disorder comprising administering to a mammalian subject having a neoplastic disorder a composition according to claim 29 , in an amount effective to inhibit neoplastic cell growth.

35. The method according to claim 34 , wherein the neoplastic disorder is a tumor characterized by blood vessel or lymphatic vessel neovascularization, and wherein the neovascularization comprises endothelial cells that express VEGFR-3.

36. The method according to claim 35 , wherein the neoplastic disorder is breast cancer.

37. The method according to claim 34 , wherein the composition is administered in an amount effective to inhibit tumor metastasis.

38. The method according to claim 34 , wherein the mammalian subject has been diagnosed with a disease characterized by proliferation of endothelial cells that express VEGFR-3, and wherein said composition inhibits at least one of angiogenesis and lymphangiogenesis in said subject.

39. The method according to claim 38 , wherein the disease is cancer.

40. The method according to claim 39 , further comprising administering a second cancer therapeutic agent to the subject.

41. The method according to claim 4 , wherein said second cancer therapeutic agent is selected from the group consisting of a chemotherapeutic agent, a nucleic acid encoding a cancer therapeutic agent, an anti-lymphangiogenic agent, and an anti-angiogenic agent.

42. The method according to claim 39 , wherein the subject has a cancer of a tissue, organ, or cell selected from the group consisting of brain, lung, liver, spleen, kidney, lymph node, small intestine, blood cells, pancreas, colon, stomach, breast, endometrium, prostate, testicle, ovary, skin, head and neck, esophagus, bone marrow and blood.

43. The method according to claim 34 , wherein the mammalian subject is human.

44. A method of treating a neoplastic disorder comprising administering to a mammalian subject having a neoplastic disorder a composition comprising a polypeptide, in an amount effective to inhibit neoplastic cell growth,

wherein the polypeptide comprises a portion of a human Vascular Endothelial Growth Factor Receptor-3 (VEGFR-3) extracellular domain (EC);

wherein said portion and said polypeptide bind to VEGFR-3 ligands VEGF-C and VEGF-D and comprise at least the first, second, and third immunoglobulin-like domains of the VEGFR-3-EC,

wherein the polypeptide lacks VEGFR-3-EC immunoglobulin-like domains IV-VII; and

wherein the polypeptide is a soluble polypeptide lacking any transmembrane domain.

45. The method according to claim 44 , wherein the neoplastic disorder is a tumor characterized by blood vessel or lymphatic vessel neovascularization, and wherein the neovascularization comprises endothelial cells that express VEGFR-3.

46. The method according to claim 45 , wherein the neoplastic disorder is breast cancer.

47. The method according to claim 44 , wherein the composition is administered in an amount effective to inhibit tumor metastasis.

48. The method according to claim 44 , wherein the mammalian subject has been diagnosed with a disease characterized by proliferation of endothelial cells that express VEGFR-3, and wherein said composition inhibits at least one of angiogenesis and lymphangiogenesis in said subject.

49. The method according to claim 48 , wherein the disease is cancer.

50. The method according to claim 49 , further comprising administering a second cancer therapeutic agent to the subject.

51. The method according to claim 50 , wherein said second cancer therapeutic agent is selected from the group consisting of a chemotherapeutic agent, a nucleic acid encoding a cancer therapeutic agent, an anti-lymphangiogenic agent, and an anti-angiogenic agent.

52. The method according to claim 49 , wherein the subject has a cancer of a tissue, organ, or cell selected from the group consisting of brain, lung, liver, spleen, kidney, lymph node, small intestine, blood cells, pancreas, colon, stomach, breast, endometrium, prostate, testicle, ovary, skin, head and neck, esophagus, bone marrow and blood.

53. The method according to claim 44 , wherein the mammalian subject is human.

54. The method according to claim 44 , wherein the portion of the VEGFR-3-EC consists of the first through third immunoglobulin-like domains of the VEGFR-3-EC.

55. The method according to claim 44 , wherein the polypeptide comprises amino acids 47-314 of SEQ ID NO: 2.

56. A method of inhibiting VEGF-C or VEGF-D comprising contacting the VEGF-C or VEGF-D with a fusion protein of claim 1 or 3 , or a polypeptide of claim 26 or 27 .
