Abstract

The invention provides isolated polynucleotide molecules, including plasmids; viral vectors; and transfected host cells that comprise a DNA sequence encoding an infectious RNA sequence encoding a North American PRRS virus; and also North American PRRS viruses encoded thereby. The invention further provides isolated infectious RNA molecules encoding a North American PRRS virus. The invention also provides isolated polynucleotide molecules, infectious RNA molecules, viral vectors, and transfected host cells encoding genetically-modified North American PRRS viruses; and genetically-modified North American PRRS viruses encoded thereby. The invention also provides vaccines comprising such plasmids, RNA molecules, viral vectors, and North American PRRS viruses, and methods of using these vaccines in swine and in other animals. Also provided are isolated polynucleotide molecules, viral vectors, and transfected host cells that comprise a nucleotide sequence encoding a peptide of a North American PRRS virus. These viral vectors and transfected host cell lines are useful in providing peptides to compensate for mutated peptide coding sequences of DNA sequences encoding genetically-modified North American PRRS viruses so that functional virions can be generated.

Claims (14)

1. An isolated polynucleotide molecule comprising a DNA sequence encoding an infectious RNA molecule encoding a North American PRRS virus that is genetically modified such that when it infects a porcine animal it is unable to produce PRRS in the animal, wherein said DNA sequence is SEQ ID NO:1, or a sequence homologous thereto that has at least 70% identity to SEQ ID NO: 1, or that hybridizes to the complement of SEQ ID NO:1 under highly stringent conditions which comprise hybridization to filter bound DNA in 0.5M NaHPO 4 , 7% SDS, 1 mM EDTA at 65° C., and washing in 0.1×SSC/0.1% SDS at 68° C.

An isolated polynucleotide molecule of claim 1 , comprising a DNA sequence encoding an infectious RNA molecule encoding a North American PRRS virus that is genetically modified such that it comprises one or more detectable heterologous antigenic epitopes, wherein the DNA sequence encoding the infectious RNA molecule is the same as or homologous to the sequence SEQ ID NO:1 or a sequence homologous thereto except for that it comprises one or more further nucleotide sequences that each encode a detectable heterologous antigenic epitope.

3. The isolated polynucleotide molecule of claim 2 , wherein said nucleotide sequences that each encode a heterologous antigenic epitope are inserted between ORF 1b and ORF 2 within said DNA sequence encoding an infectious RNA molecule encoding a North American PRRS virus.

4. The isolated polynucleotide molecule of claim 2 , wherein said nucleotide sequences that each encode a heterologous antigenic epitope are inserted in ORF 1a or ORF 1b within said DNA sequence encoding an infectious RNA molecule encoding a North American PRRS virus.

The isolated polynucleotide molecule of claim 2 , wherein transcription of said polynucleotide molecule results in formation of a genomic RNA and subsequent formation of subgenomic RNA molecules, and wherein at least one heterologous antigenic epitope is translated directly from the genomic RNA molecule.

The isolated polynucleotide molecule of claim 2 , wherein transcription of said polynucleotide molecule results in formation of a genomic RNA and subsequent formation of subgenomic RNA molecules, and wherein at least one heterologous antigenic epitope is translated from a subgenomic RNA molecule.

The isolated polynucleotide molecule of claim 5 , wherein translation of said genomic or subgenomic RNA molecules results in formation of individual fusion proteins following translation, said proteins consisting essentially of a fusion between a PRRS virus protein and said heterologous antigenic epitope.

8. The isolated polynucleotide molecule of claim 5 , wherein translation of said genomic or subgenomic RNA molecules results in formation of a protein consisting essentially of the heterologous antigenic epitope.

The isolated polynucleotide molecule of claim 6 , wherein translation of said genomic or subgenomic RNA molecules results in formation of individual fusion proteins following translation, said proteins consisting essentially of a fusion between a PRRS virus protein and said heterologous antigenic epitope.

10. The isolated polynucleotide molecule of claim 6 , wherein translation of said genomic or subgenomic RNA molecules results in formation of a protein consisting essentially of the heterologous antigenic epitope.

11. A plasmid that comprises the polynucleotide of claim 1 , and a promoter capable of transcribing said DNA molecule in a suitable host cell.

12. The plasmid of claim 11 , wherein the North American PRRS virus-encoding sequence thereof further encodes one or more detectable heterologous antigenic epitopes.

13. A transfected host cell that comprises the plasmid of claim 11 .

14. A transfected host cell that comprises the plasmid of claim 12 .
