Abstract

Compounds, compositions and methods are provided for inhibiting the expression of human STAT3. The compositions comprise antisense oligonucleotides targeted to nucleic acids encoding STAT3. Methods of using these oligonucleotides for inhibition of STAT3 expression and for promotion of apoptosis are provided. Methods for treatment of diseases, particularly inflammatory diseases and cancers, associated with overexpression or constitutive activation of STAT3 or insufficient apoptosis are also provided.

Claims (13)

1. A modified or unmodified antisense compound 20 to 30 nucleobases in length targeted to a nucleic acid molecule encoding human STAT 3, comprising the nucleotide sequence shown in SEQ ID NO: 184, or a pharmaceutically acceptable salt thereof.

2. The antisense compound of claim 1 , which comprises at least one modified intemucleoside linkage.

3. The antisense compound of claim 2 , wherein said modified internucleoside likage linkage is a phosphorothioate linkage.

4. The antisense compound of claim 3 , which comprises at least one modified sugar moiety.

5. The antisense compound of claim 4 , wherein said modified sugar moiety is a 2′-O-(2-methoxyethyl) moiety.

6. The antisense compound of claim 5 , which comprises at least one modified nucleobase.

7. The antisense compound of claim 6 , wherein said modified nucleobase is a 5-methyl cytosine.

8. The antisense compound of claim 1 , wherein all internucleoside linkages are phosphorothioate, nucleotides 1-5 and 16-20 reading from the 5′ end to 3′ end of SEQ ID NO: 184 each comprise a 2′-O-(2-methoxyethyl) sugar, or a pharmaceutically acceptable salt thereof.

9. The antisense compound of claim 8 , wherein all cytidine residues are 5-methylcytidines.

10. The antisense compound of claim 9 , wherein said pharmaceutically acceptable salt is a sodium salt.

11. A pharmaceutical composition, comprising said antisense compound of claim 1 and a pharmaceutically acceptable carrier or diluent.

An antisense oligonucleotide, consisting of the nucleotide sequence shown in SEQ ID NO: 184, wherein every intemucleoside linkage is a phosphorothioate linkage, nucleotides 1-5 and 16-20 reading from the 5′ end to the 3′ end each comprise a 2′-O-(2-methoxyethyl) sugar, nucleotides 6-15 are 2′-deoxynucleotides, every cytidine residue is a 5-methylcytidine, and which is in the form of a sodium salt.

13. A pharmaceutical composition, comprising said antisense oligonucleotide of claim 12 , and a pharmaceutically acceptable carrier or diluent.
