Abstract

The present invention provides assays for the study of the interaction of EPF and its EPF-related peptides with a hDRR receptor. The assays are useful to identify whether a test compound can bind to the hDRR receptor under conditions in which EPF or related peptide can bind to the receptor. The assays are also useful to determine whether the test compound is an agonist or antagonist of hDRRs. The above assays can be performed in a variety of formats including competitive, non-competitive and comparative assays in which the interaction of EPF or EPF-related peptides with hDRRs is assessed as a positive or negative control or compared to the results obtained with the test compound.

Claims (11)

1. An isolated and purified hDRR7 binding fragment of EPF of between 16 and 21 amino acids having at least 70% sequence identity with EPF, said fragment being selected from the group consisting of:

a sequence consisting of amino acid sequence SEQ ID NO:8; or

SEQ ID NO:18, SEQ ID NO:19, SEQ ID NO:20 or SEQ ID NO:21; and

a sequence comprising the amino acid sequence AFRKFLPLFDRVL (SEQ ID NO: 28).

2. An isolated and purified nucleic acid molecule encoding hDRR7 binding fragment of EPF of between 16 and 21 amino acids having at least 70% sequence identity with EPF, said fragment being selected from the group consisting of:

a sequence consisting of amino acid sequence SEQ ID NO:8;

SEQ ID NO:18, SEQ ID NO:19, SEQ ID NO:20 or SEQ ID NO:21; and

a sequence comprising the amino acid sequence AFRKFLPLFDRVL (SEQ) ID NO: 28).

A pharmaceutical composition comprising an hDDR7 binding fragment of EPF of between 16 and 21 amino acids having at least 70% sequence identity with EPF, said fragment being selected from the group consisting of SEQ ID NO: 8, SEQ ID NO:18, SEQ ID NO:19, SEQ ID NO:20 or SEQ ID NO:21 and a sequence comprising the amino acid sequence AFRKFLPLFDRVL (SEQ ID NO:28).

A pharmaceutical composition for the treatment of autoimmune diseases or other conditions where immunosuppressive actions are desired or to prevent transplant rejection, wherein the pharmaceutical composition comprises the hDRR7 binding fragment of EPF of between 16 and 21 amino acids having at least 70% sequence identity with EPF, said fragment being selected from the group consisting of SEQ ID NO:8, SEQ ID NO:18, SEQ ID NO: 19, SEQ ID NO: 20 and SEQ ID NO: 21 or a sequence comprising the amino acid sequence AFRKFLPLFDRVL (SEQ ID NO: 28).

5. The pharmaceutical composition of claim 4 , wherein said autoimmune disease or condition where immunosuppressive action is desired is selected from rheumatoid arthritis, multiple sclerosis and Inflammatory Bowel Disease (IBD).
